Could a combat veteran's horrific experiences in early-2000s Afghanistan have a direct, biological impact on his or her now-adult daughter's risk of a heart attack later in her life? This concept would have been unapologetically mocked a mere twenty years ago, and it has only been in the past decade that the new field of epigenetics has revealed a distinct possibility for this event to actually take place—for parents' experiences to profoundly influence the biology of their children. Posttraumatic stress disorder (PTSD) is a devastating psychiatric disorder characterized by distinctive symptom clusters, including intrusive memories (i.e., flashbacks), avoidance of related stimuli, affective changes, and hyperarousal. Strikingly, patients with PTSD face a significantly increased risk for a number of inflammation-driven pathologies, ranging from cardiovascular to autoimmune disease. Yet, the exact etiology of this increased risk remains unknown. The immune system is known to be strongly influenced by the sympathetic nervous system, and sympathetic overactivity is a hallmark of PTSD. AbstractFear is an evolutionarily conserved mechanism that allows an organism to discern potential harm and act accordingly by engaging in defensive behaviors. While fear is an adaptive response, dysregulation of fear by means of a traumatic event can lead to psychiatric conditions such as post-traumatic stress disorder (PTSD). Animal research using rodents in Pavlovian context fear conditioning experiments allows for the ability to study the underlying neural circuitry of threat-appropriate and aberrant fear learning and memory that may contribute to PTSD. Though there has been substantial progress towards understanding brain-behavior relationships and characterizing the neurobiology of psychiatric disorders, research has not translated as expected into novel prevention and treatment of mental health conditions. One limitation may be the emphasis on individual-level variables (e.g., income) and omission of relevant area-level factors (e.g., neighborhood socioeconomic disadvantage). Recently, attention has been directed towards identifying the biological mechanisms by which neighborhoods impact mental health. The chronic stress associated with living in a disadvantaged neighborhood promotes a cascade of maladaptive events, which in turn impact brain structure and functioning. The ability to effectively withhold an inappropriate response is a critical feature of cognitive control. Prior research indicates alterations in neural processes required for motor response inhibition in anxious individuals, including those with posttraumatic stress disorder (PTSD), and those who engage in regular cannabis use. However, thus far most research has examined how anxiety-related symptoms and cannabis use influence response inhibition in isolation of one another. Posttraumatic stress disorder (PTSD) is a heterogenous psychological disorder that may result from exposure to a traumatic event. Using functional magnetic resonance imaging (fMRI), symptoms of PTSD have been associated with aberrations in brain networks that emerge in the absence of a given cognitive demand or task, called resting state networks. Most previous research in resting state networks and PTSD has focused on aberrations in the static functional connectivity among specific regions of interest (ROI) in the brain and within canonical networks constrained by a priori hypotheses. On January 30th 2020, the World Health Organization (WHO) declared the COVID-19 pandemic a Public Health Emergency of International Concern (PHEIC). Italy has been one of the most affected countries in the world. Research suggests that many veterans with post-traumatic stress disorder (PTSD) struggle with reintegration (Sayer et al., 2010), but are unlikely to seek help or complete treatment (Schottenbauer et al., 2008). To make matters worse, available treatment options are often time consuming, challenging, and/or associated with negative side effects (Carafano & Hutchinson, 2017). The study of how the brain acquires fearful memories has attracted considerable experimental attention, due in part to the promise of discovering novel therapeutic approaches for psychiatric disorders that are characterized by unusually strong and persistent traumatic memories. In recent years, extensive research has focused on studying the neural and molecular mechanisms by which fear memories are acquired, stored, and retrieved in the brain. According to Lynne Weilart (2013), in her article on the reasons why people seek out therapy, trauma is the number one reason people attend counseling. Many different trauma-informed approaches are designed specifically to address the consequences of trauma and to facilitate healing. Some of these approaches are as follows: Cognitive Behavioral Therapy (CBT); Dialectical Behavioral Therapy (DBT);Mentalization Based Therapy (MBT); Trauma Systems Therapy (TST); Trauma Assessment Pathway (TAP); and Attachment, Self-Regulation, and Competency (ARC) (de Arellano, Danielson, Ko, & Sprauge, 2008). This study tested the role of affect lability and disinhibition in mediating associations between PTSD symptoms and two forms of alcohol-related problems, dependence syndrome symptoms (e.g., impaired control over consumption) and conduct problems (e.g., assault, risk behaviors). Genotype at the serotonin transporter linked polymorphic region (5-HTTLPR) was hypothesized to moderate associations between traumatic stress and PTSD symptoms. In addition, the study tested whether childhood traumatic stress moderated associations between combat trauma and PTSD symptoms. Participants were 270 OIF/OEF/OND veterans. Post traumatic stress disorder (PTSD) patients frequently show impairment in safety learning (Jovanovic, Kazama, Bachevalier, & Davis, 2012). Since the amygdala is known to be critical for emotional processing(Wassum & Izquierdo, 2015) and dopamine signaling in the amygdala is important for mediating both fear and reward learning, current experiments examined the role of dopamine signaling in the BLA in mediating both safety learning and reward seeking. Symptom severity among veteran survivors with partial or full posttraumatic stress disorder (PTSD) continues to increase, with approximately 40% of U.S. veterans reporting significant symptomology 10 years after initial onset of the condition. Veteran survivors often struggle to find therapeutic interventions that meet their specific needs and have a difficult time maintaining a therapy that is both equitable and evidenced based. Individuals respond differently to traumatic stress. Social status, which plays a key role in how animals experience and interact with their social environment, may influence how individuals respond to stressors. In this study, we used a conditioned defeat model to investigate whether social status alters susceptibility to the behavioral and neural consequences of traumatic stress. Conditioned defeat is a model in Syrian hamsters in which an acute social defeat encounter results in a long term increase in submissive behavior and a loss of normal territorial aggression.
