The effectiveness of polycyclic aromatic hydrocarbons (PAHs) for selective solubilisation of single walled carbon nanotubes (SWCNTs) has been studied by Raman spectroscopy. Polyphenyl and polyacene PAHs of different lengths are used. Selective interaction between the PAHs and SWCNT is investigated by analyzing the Raman radial breathing modes the frequency positioning of which yields information concerning the diameter distribution of the SWCNT sample. Samples were dispersed at concentrations below the debundling limit and deposited on quartz substrates. This thesis contains a systematic study of the dispersion of pristine HiPco Single Walled Carbon Nanotubes (SWNTs) in a series of organic solvents. A double beamed UV-Vis-NIR absorption spectrometer coupled with an integrating sphere was employed to demonstrate the dispersibility of SWNTs in different solvents. Raman Spectroscopy and Atomic Force Microscopy (AFM) were used to confirm the debundling and exfoliation of SWNTs aggregates. An investigation of the solubility of SWNTs in four chlorinated aromatic solvents demonstrated that the similarity in structure between solvent molecules and nanotube sidewall is not a dominant factor to obtain stable SWNT solutions. An important area of application of holographic optical elements (HOEs) is in optical and electronic speckle pattern interferometry. The design, fabrication and characterization of holographic optical elements (HOEs) for electronic speckle pattern interferometry are presented. Reflection HOEs (RHOEs) were fabricated for use in electronic speckle pattern interferometers (ESPI) and laser Doppler vibrometers (LDV). The HOE-based interferometer is sensitive to out-of-plane displacements only. The results obtained are promising for future applications of the system for modal analysis.

Novel nanocomposites consisting of a water soluble acrylamide–based photopolymer and colloidal zeolite nanoparticles of zeolite Beta and zeolite A were prepared. The interactions between the photopolymer components and zeolite nanoparticles in the photopolymerizable nanocomposites were characterized for the first time by 13C NMR and Visible spectroscopy. It was found that the zeolite Beta nanoparticles (up to 5% wt.) behave as a non-inert additive, resulting in an effective increase in layer thickness that causes doubling of the diffraction efficiency of the nanocomposite in comparison to that of the undoped photopolymer. The study of the interaction of anticancer drugs with mammalian cells in vitro is important to elucidate the mechanisms of action of the drug on its biological targets. In this context, Raman spectroscopy is a potential candidate for high throughput, noninvasive analysis. To explore this potential, the interaction of cis-Diamminedichloroplatinum (II) (Cisplatin) with a human lung adenocarcinoma cell line (A549) was investigated using Raman microspectroscopy. The results were correlated with parallel measurements from the MTT cytotoxicity assay, which yielded an IC50 value of 1.2±0.2 μM. The accuracy of a transient elastography liver-scanning ultrasound system was assessed using a novel application of PVA-cryogel as a tissue-mimicking material with acoustic and shear elasticity properties optimized to best represent those of liver tissue. Although the liver scanning system has been shown to offer a safer alternative for diagnosing liver cirrhosis through stiffness measurement, as compared to the liver needle biopsy exam, the scanner’s accuracy has not been fully established. The development of a panchromatic acrylamide based photopolymer (ABP) for holographic recording application is presented. The scattering of the recording medium was characterised by measuring the Bidirectional Scattering Distribution Function (BSDF). The dynamic range in reflection mode of recording was evaluated by measuring the diffraction efficiencies of the holographic gratings recorded individually at 633nm, 532nm, and 473nm wavelengths at spatial frequencies of 4200 l/mm, 5000 l/mm and 5700 l/mm respectively. Mild centrifugation (945 g) was carried out to remove large bundles. Atomic force microscopy (AFM) studies revealed that the bundle size decreased as the dispersion was diluted. By measuring the UV−vis−NIR absorption before and after centrifugation as a function of the concentration, the dispersion limit of SWNTs in each solvent can be determined. The formation of surface relief profile in photopolymerisable systems when illuminated with a focused beam of light is simulated numerically using a two-way diffusion model that takes account both for monomer and short polymer chains diffusion. The concentration and spatial distribution dynamics of monomer and short and long polymer chains are calculated. The surface profile is obtained from calculated components concentrations considering different densities of monomer and polymer. The influence of the illumination time, intensity and spot diameter on the surface profiles dynamics is discussed. The in vitro study of cellular species using Raman spectroscopy has proven a powerful non-invasive modality for the analysis of cell constituents and processes. This work uses micro-Raman spectroscopy to study the chemical fixation mechanism in three human cell lines (normal skin, normal bronchial epithelium, and lung adenocarcinoma) employing fixatives that preferentially preserve proteins (formalin), and nucleic acids (Carnoy’s fixative and methanol–acetic acid). Vibrational spectroscopy (Raman and FTIR microspectroscopy) is an attractive modality for the analysis of biological samples since it provides a complete non-invasive acquisition of the biochemical fingerprint of the sample. A theoretical model for formation of a short exposure holographic grating is presented. The model accounts for both monomer and polymer diffusion and distinguishes between short polymer chains capable of diffusing and long polymer chains that are immobile. It is shown that the experimentally observed decrease of diffraction efficiency at higher spatial frequency can be predicted by assuming diffusion of short-chain polymers away from the bright fringes. The time evolution of the refractive index modulation after short exposure is calculated and compared with experimental results. The optical properties of photopolymer layers consisting of an acrylamide–based matrix and microporous aluminophosphate nanocrystals of AEI- type are investigated. The compatibility of the photopolymer doped with the nanoparticles is studied. The surface and volume properties of the layers with different levels of doping with microporous nanocrystals are characterized. The effective refractive indices and absorption coefficients of the doped photopolymer layers are determined and used to calculate the refractive index and porosity of pure AEI nanoparticles used as dopants. We describe the optimisation of the holographic patterning of sub-micrometer surface relief structures in an acrylamide-based photopolymer. A substantial improvement in the photoinduced surface relief resolution was achieved by altering the photopolymer chemical composition and by introducing a single step post recording thermal treatment of the layers. It was observed that, by optimisation of the chemical composition of the photopolymer layers, the maximum achievable spatial frequency increases from 200 l/mm to 550 l/mm. A photopolymerizable material for use in the visible normally consists of a dye sensitizer, a free radical generator, monomer/monomers and, optionally, a binder, and is prepared usually as a dry or liquid film. Upon illumination with light of appropriate wavelength, a photopolymerisation reaction is triggered, leading to a change in the film’s refractive index. Because the presence of the dye is essential, the film is usually made sensitive to light during its preparation. Here we separate the film preparation and the sensitisation processes. The radiation-induced bystander effect is a phenomenon known to occur post irradiation, characterised by the induction of biological effects in unirradiated cells as a result of receiving signals from irradiated cells or their culture medium. Chemicalinduced bystander effects are poorly characterised and there are no reports of a bystander effect induced by metals. Heavy metals and in particular chromium (VI) are known to cause persistent genomic instability. The primary aim of this project was to develop novel anatomically realistic renal phantoms for the evaluation of current and emerging ultrasound techniques capable of diagnosing all grades of renal artery stenosis. Renal artery stenosis is considered the most common cause of potentially curable secondary hypertension which if left untreated can lead to renal failure. The humidity and temperature responses of a reflection hologram recorded in a self-processing photopolymer layer have been characterised by measurement of the position of the maximum intensity in the spectral response of the hologram. It has been demonstrated that such a hologram can be used for indication of the relative humidity (RH) in a specific temperature range. The operational range and reversibility of the holograms have been characterised in a controlled humidity and temperature environment. The humidity studies were carried out in the range from 10 to 80% RH and the temperature range under study was 15 – 50 ºC.

Single walled carbon nanotubes have gained enormous popularity due to a variety of potential applications which will ultimately lead to increased human and environmental exposure to these nanoparticles. This study was carried out in order to evaluate the inflammatory response of immortalised and primary human lung epithelial cells (A549 and NHBE) to single walled carbon nanotube samples (SWCNT). Special focus was placed on the mediating role of lung surfactant on particle toxicity.
