A classifier tool was created to identify major serogroups of Moraxella bovis based on spectral profiles collected from bacterial cells through the use of Fourier transformed infrared spectroscopy. M. bovis strains were selected from a group of previously sequenced and genotyped isolates, which provided the pilin sequence type for each isolate. The obtained spectroscopic profiles were analyzed through artificial neural networks to create a classifier tool that could identify the PilA type for unknown strains. Control of Bovine Viral Diarrhea Virus (BVDV) relies on resource-intensive individual animal sampling to detect and remove persistently infected (PI) cattle. Herd-level surveillance tools would be useful for herds with unknown BVDV status and for monitoring herds with BVDV-free status. The overall objective of this thesis is to explore the viability of BVDV surveillance at a herd-level using samples collected without handling individual animals. The first objective was to determine the feasibility of using stable flies as a sampling tool to detect BVDV. As feral swine continue to expand their geographical range and distribution across the United States, their involvement in crop damage, livestock predation, and pathogen transmission is likely to increase. Despite the relatively recent discovery of feral swine involvement in the aetiology of a variety of pathogens, their propensity to transmit and carry a wide variety of pathogens is disconcerting. We examined sera from 2055 feral swine for antibody presence to six serovars of Leptospira that can also infect humans, livestock or domestic animals. About 13% of all samples tested positive for at least one serovar, suggesting that Leptospira infection is …

Caprine Arthritis-Encephalitis Virus (CAEV) is an incurable disease of goats that has both social and economic impacts. Clinical disease in goats includes encephalitis in kids; chronic arthritis, inflammatory mastitis and progressive respiratory disease in adults. In the last 25 years there have been significant changes in the US goat industry with rapid growth in the meat and dairy industries. Recent prevalence studies are lacking and historic studies may not reflect changes in the industry. Bovine herpes virus 1 (BHV-1) is a significant viral pathogen in cattle that induces a myriad of clinical symptoms. These symptoms include: conjunctivitis, upper respiratory tract infections, genital disorders, and abortions. BHV-1 infection can also lead to transient immune-suppression, which predisposes cattle to secondary bacterial infection leading to life-threatening pneumonia referred to as bovine respiratory disease (BRD). Following acute infection, BHV-1 establishes latency in sensory neurons within trigeminal ganglia. Reactivation of the virus can occur periodically, resulting in virus transmission. The latency-related (LR) RNA is the only abundantly expressed transcript in latently infected sensory neurons and it encodes several proteins, …

Sera samples were collected from 21 free-ranging, captured female elk (Cervus elaphus nelsoni) in 1995- 96, and tissue and sera samples were collected from 415 hunter-harvested elk from 1995 to 2006 and tested for selected diseases. Titers for Anaplasma marginale were detected in 81 of 436 (19%) elk. Occurrence of antibodies to anaplasmosis increased from 4 to 40 elk from 2002 to 2006. Titers for bovine viral diarrhea virus (BVDV) were detected in 18 of 346 (5%) samples. Titers for Leptospira interrogans serovars were detected in 21 of 289 (7%) of samples from 1995 to 2004. The protozoan, Tritrichomonas foetus (TF), has been recognized as a cause of bovine infertility for more than 100 years (Skirrow and BonDurant, 1988). As an obligate parasite of the bovine reproductive tract its control and eradication seems achievable (Harding, 1950). However, this disease continues to trouble US cattle producers and a recent epidemic in the Western US has lead to increased interest in research and regulatory efforts (Cima, 2009).

Within the last decade a new etiologic agent has been identified as a possible cause of diarrhea in humans and domestic animals—that being the protozoan parasite Cryptosporidium. The purpose of this study was to determine the incidence and distribution of Cryptosporidium in the state of Nebraska, to determine if Cryptosporidium occurs more frequently in association with other enteropathogens and to determine about the relation of scouring in the calves to infection with Cryptosporidium. Neonatal calf diarrhea typical of that seen in field cases was produced in colostrum-deprived calves by inoculating either feces or bacteria-free filtrates via the duodenum, orally, or by a spray (aerosol). Three calves, kept free of E. coli, developed severe diarrhea following inoculation with a bacteria-free filtrate. Intense fluorescence in the epithelium of sections of small intestine from two experimental calves was observed following staining with ftuorescein-labeled gamma globulin produced with viral antigen prepared from the feces of two experimental diarrhetic E. coli-free calves. Swine brucellosis is a bacterial disease that affects swine, other domestic animals, and humans. Severe exposure causes the disease in cattle, dogs, cats, and poultry. The disease can spread from one species of animal to another, and from animals to humans. However, it does not normally spread from one human to another, or from humans to animals. The disease costs the swine industry about $10 million a year. Losses due to human infection cannot be estimated. The great increase in the number of swine found to be tuberculous presented a problem, the solution of which appeared to be desirable before adequate control measures could be devised or recommended. In the hope that a solution of the problem may be found this station undertook an investigation and attempted by experimental inquiry to determine the infection type of the tuberculosis associated with the animals which constituted the bulk of the retentions.
