Periodontal ligament cell viability at the time of tooth replantation is of utmost importance for proper repair of the damaged tissues (1). Multiple different options are currently available for storage of an avulsed tooth. The goal of this study was to investigate new storage media and evaluate their ability to maintain cell viability over time. Materials and Methods: Cultured human periodontal ligament fibroblasts (hPDLF) were seeded on 96-well plates. Introduction: One important step of root canal therapy is the process of cleaning and shaping each canal. This process involves using endodontic rotary files combined with chemical irrigants to remove pulpal remnants and infected dentin from the canal while eliminating pathogenic bacteria. It is essential to maintain proper canal anatomy while cleaning and shaping. The challenge for the practitioner is to select a rotary file system that will be flexible enough to maintain canal anatomy but strong enough to prevent breakage under normal use. Introduction: Viability of periodontal ligament (PDL) cells is a key prognostic factor fora replanted tooth following avulsion. Dehydration results death of PDL cells, which in turn causes ankylosis upon replantation, followed by replacement resorption. Hank’s Balanced Salt Solution (HBSS) has been recommended as the standard storage medium for an avulsed tooth. This study evaluated the effect of residual gutta percha on the retention strength of prefabricated post cemented with four different cements. Forty intact mandibular single canal canines or premolars were selected for standardized size and quality, endodontically treated, and decoronated to the cementoenamel junction. A post space was prepared to accommodate Para post XP size 4 (Coltene, Whaledent Inc., Cuy Falls, Ohio) to a depth of 8 mm. Bacterial lipopolysaccharide (LPS) has been implicated in the pathogenesis of pulpal and periapical diseases. Several studies have evaluated the ability of various medicaments and irrigants used during root canal therapy to chemically detoxify LPS. The purpose of this study was to evaluate the ability of some endodontic sealers to detoxify LPS. This investigation compared the quantities of TNF-α released by human monocytes exposed to LPS or LPS pre-treated with sealers. Periradicular surgery is a viable option to save natural teeth when non-surgical treatment fails or when endodontic retreatment is not feasible or contraindicated. Laboratory and animal studies have demonstrated that MTA is biocompatible, provides an excellent seal against penetrating bacteria, and promotes hard tissue healing. Two variations of MTA (Gray and White) have been introduced to the dental market. The purpose of this study was to histologically compare the in vivo response to immediate furcation perforation repair using gray or white MTA in beagle dogs.

Mineral Trioxide Aggregates (MTA) was recently developed at Loma Linda University. It has been shown to possess good sealing ability when tested with dye, bacteria, and a fluid filtration technique.

Endotoxin is a component of the cell wall of Gram negative bacteria. It has been proposed that it may play a role in the pathogenesis of periradicular lesions. Previous studies have demonstrated that MTA can prevent the leakage of bacteria. However, the bacterial products were not tested during these studies.

Sealing of the root canal system is usually accomplished by the conservative endodontic approach. Cases where it is impossible to treat nonsurgically are usually treated surgically by an apicoectomy and retrofill. A study was undertaken to determine the concentration at which a sealer reaches a point of nontoxicity. An in vitro model utilizing HeLa cells and human fibroblasts was used to observe the effect of various concentrations of eight freshly mixed and eight set root canal sealers. Evaluation of toxicity was determined by cell survival which was measured by incorporation of 3H-thymidine into the nucleic acid components of each cell type used. Determination Of Lead Content In Apical Tissue Following Root Canal Obturation With Sargenti's Formula, G. Merle Hickok

Four Stump-tail monkeys were used to help determine if the lead in Sargenti's formula diffuses out into the apical tissue while confined entirely within the root canal. In each animal two teeth were filled with Sargenti's formula 1-2 millimeters short of the radiographic apex, and two teeth were filled with gutta percha and Proco-Sol sealer for controls, making a total sample size of sixteen. The time ranged from 48 hours to 120 days. Lead analysis using the spectrophotometric method showed a higher lead content in the teeth filled with Sargenti's formula than with the gutta percha controls.

An in vitro study using radioactively-labeled tissue culture cells was conducted over a seven-month period on Grossman's sealer, Rickert's sealer, N2 Permanent, and Cavit to determine the lasting and relative tissue toxicity of these materials.

From this study it was observed that all materials possess lasting tissue toxicity. When relative toxicity is considered, Grossman's is the most toxic, followed by N2, Rickert's and Cavit in descending order of toxicity. Both Grossman's and N2 would be considered highly toxic, with Rickert's displaying moderate toxicity. Cavit would be considered mild to moderate in toxicity.

A histological comparison of forty-four human teeth in which vital partial pulpectomies were performed, and the pulp stumps covered with dycal, N-2 and ZOE was carried out. The time interval at which the teeth were extracted for study was ten, twelve and fourteen weeks. These teeth were on patients twelve through seventeen years of age and were to he extracted for orthodontic purposes. The teeth were caries free, vital and asymptomatic. An in vitro study using polyethylene tubes filled with Grossman's sealer, Rickert's sealer, and N2 permanent was conducted over a six month period in an effort to determine their lasting antimicrobial properties against eight different microorganisms.

From the study it was observed that N2 permanent possesses a long-lasting antimicrobial effect, but this effect appears to diminish greatly with time. Further investigation is needed to determine whether or not the effect is actually permanent in nature. This study compared endodontic overfills of N2 and gutta percha-Grossman's Sealer from a histopathological basis. The study employed four male rhesus monkeys and a sample of 31 teeth.

The most obvious difference between the samples filled with N2 and the gutta percha-Grossman's Sealer samples was the recurring presence of ankylosis in the N2-treated teeth. Results also included bone necrosis, inflammation and root and bone resorption.

The N2 samples were considered to have evidenced greater violence of reaction apically.

The purpose of this investigation was to test the effect of different types of condensation, irrigants, filling materials and techniques, and variations in root canal preparation, on the success of sealing lateral and accessory canals, whose presence was determined before hand, and to relate certain inherent anatomic and morphologic characteristics of these canals to the amount of sealing obtained. This paper was undertaken to study the use of Cavit an[sic] an endodontic filling material.

Part I examines the physical and biological properties of Cavit. This material has a 14% linear setting expansion, which results in a very tight interphase seal. It is theorized that this type of seal in connection with a three dimensional obturation is necessary to prevent the flow of irritant from the oral environment to the periapical tissue.

The biological properties are tentatively shown to be similar to ZOE.

The main purpose of this investigation was to test the endodontic sealing abilities of silver cones, N2, warm gutta percha, chemically softened gutta percha, laterally condensed gutta percha and Cavit under carefully controlled conditions. Evaluations were done by radiographic, macroscopic, microscopic, and autoradiographic examinations.

The four points which must be considered when evaluating the efficacy of sealing root canal systems, in vivo, were presented and discussed.

Cavit as a total endodontic filling material was presented, investigated and discussed.
