Evaluation Of Different Scaffolds For Bone Regeneration In Rat Calvarial Bone Defects, Jeerawit Sirakittiworapong

Chulalongkorn University Theses and Dissertations (Chula ETD)

Scaffold plays a key role in the context of tissue engineering. It not only provides a structural support for cells, but also creates an appropriate milieu for recruited cells, enhancing the therapeutic effects of cell-based treatments, and enabling the controlled release of biological cues such as growth factors. Collagen is widely used scaffold material in tissue engineering, particularly in periodontal and bone regeneration. Other natural polymers, however, have been developed and can be useful. Therefore, in this study, we aim to investigate the ability of four different natural polymers including collagen, chitosan, silk fibroin, and silk fibroin/gelatin hydrogel in promoting …

Expression Of Vascular Endothelial Growth Factor Protein In Mrna-Transfected Human Periodontal Ligament Cells, Mana Naratippakorn

Chulalongkorn University Theses and Dissertations (Chula ETD)

The complete regeneration of periodontal tissues following current periodontal therapy remain challenging and unpredictable. Nucleoside-modified messenger RNA (mRNA) technology can be a promising novel platform in regenerative medicine. The aims of this study were to evaluate whether pseudouridine modified mRNA encoding vascular endothelial growth factor (VEGF) could induce VEGF production in human periodontal ligament cells (PDLCs) and this translated protein function by promoting in vivo blood vessel formation using chorioallantoic membrane (CAM) assay. Isolated PDLCs from healthy periodontal tissue were transfected with modified mRNA encoding VEGF (VEGF mRNA) complexed with a transfecting agent, Lipofectamine 2000 (L2000) and L2000 alone (control). …

The Immunomodulatory Roles Of Supracrestal Gingival Connective Tissue-Derived Human Mesenchymal Stromal Cells In The Polarization Of Macrophages, Jirawit Inthayat Jan 2020

The Immunomodulatory Roles Of Supracrestal Gingival Connective Tissue-Derived Human Mesenchymal Stromal Cells In The Polarization Of Macrophages, Jirawit Inthayat

Chulalongkorn University Theses and Dissertations (Chula ETD)

MSCs exert their immunomodulatory effects on various immune cells by cell-cell contact and cytokines secretion. Our previous study has demonstrated that supracrestal gingival connective tissue-derived mesenchymal stem cells (SG-MSCs) were recognized to be a good candidate for periodontal regeneration. SG-MSCs showed the similar potential to PDL-MSCs and held significant advantage over PDL-MSCs by which a tooth extraction is not required. In terms of immunomodulatory properties, the effect of SG-MSCs on macrophage has never been explored. This study was aimed to investigate the effects of SG-MSCs on macrophages by cocultured SG-MSCs and THP-1-derived macrophages (THP-1-MPs) in direct cell-cell contact condition. Briefly, …

Protein Expression And Inflammatory Response After Delivery Of Mrna Encoding Platelet-Derived Growth Factor-Bb In Sucrose Citrate Buffer Into Rat Gingiva, Phan Bhongsatiern

Chulalongkorn University Theses and Dissertations (Chula ETD)

Messenger RNA (mRNA) has emerged as a novel therapeutic modality in medical fields, including regenerative medicine. The concept of mRNA-based therapy is to use synthesized mRNA encoding therapeutic protein delivered to target tissue. Thus, mRNA encoding growth factor could be a promising alternative to recombinant protein. Platelet-derived growth factor (PDGF) is one of most extensively studied growth factors for periodontal regeneration. To date, mRNA therapy has never been explored in the field of periodontal regeneration. The aim of the study is to examine the effect of nucleoside-modified mRNA encoding PDGF-BB on the level of PDGF-BB protein and inflammatory response at …

Utilization Of Modified Mrna Encoding Bone Morphogenetic Protein-2 For Periodontal Regeneration: An In Vitro Study, Promphakkon Kulthanaamondhita

Chulalongkorn University Theses and Dissertations (Chula ETD)

Current modalities for periodontal regeneration provide modest success, however complete periodontal regeneration is still not achievable. Recently in vitro synthesized nucleoside-modified messenger RNA (mRNA) has emerged as a novel platform in regenerative medicine. The aims of this study are to investigate the ability of human periodontal ligament cells (PDLCs), clinically relevant target cells, to produce bone morphogenetic protein-2 (BMP-2), a significant protein for bone formation after transfected with modified mRNA that encode this protein. We investigated the ability of translated protein for enhancing PDLC proliferation and promotes endothelial cell tube formation, marker of angiogenesis. Isolated PDLCs from healthy periodontal tissue …
