Purpose/Objective(s): Stereotactic body radiotherapy (SBRT) is considered as the treatment of choice for stage I non-small cell lung cancer (NSCLC) in medically inoperable patients. Purpose/Objective(s): Radiation therapy in lung cancer relies on CT and functional imaging (FDG-PET) to delineate tumor volumes. Semi-automatic contouring tools have been developed for PET to improve on the inter-observer bias of manual contouring and intrinsic differences in imaging equipment. A common method involves using a threshold at a given percentage of the max activity, which may be less accurate with smaller tumors and tumors with low source to background ratio. Purpose/Objective(s): Image guidance (IG) may permit higher radiotherapy (RT) doses (>65 Gy) after radical prostatectomy (RP) without increased toxicity, with improved accuracy and smaller margins. Purpose/Objective(s): It is becoming increasingly evident that the behavior of cancers from different subsites of the head and neck varies greatly despite similar histologic appearance. As current head and neck trials are focusing more on a risk-based approach, it is unclear into which trials patients with hypopharyngeal primaries (HP) should be included. Purpose: While several studies analyze the incidence of brain metastases in patients with non-small cell lung cancer (NSCLC) treated with prophylactic cranial irradiation (PCI), there is little data available on how this treatment can affect medical cost and morbidity. Our goal was to analyze those issues patients encounter secondary to brain metastases, often in the final months of their lives, and to support the hypothesis of economic benefit and cost effectiveness resulting from the use of PCI in this population.

Introduction: Glioblastomas (GBM) frequently overexpress the epidermal growth factor receptor (wtEGFR) or its mutant, EGFRvIII contributing to radioresistance. New treatment strategies for GBM include blockade of EGFR signaling and angiogenesis. Cediranib (CD) is a highly potent VEGFR-2 RTKI that inhibits all three VEGF receptors. This study investigated the radiosensitizing potential of CD in combination with temozolamide (TMZ) in U87 GBM xenografts expressing wtEGFR or EGFRvIII.

Introduction: Tumor cells grow in nutrient and oxygen deprived microenvironments and adapt to the suboptimal growth conditions by altering metabolic pathways. This adaptation process characteristically results in a tumor phenotype that displays upregulated Hif-1α anaerobic glycolysis, chronic acidification, reduced rate of overall protein synthesis, lower rate of cell proliferation and aggressive invasive characteristics. Most transplantable tumors exhibit a pHe of 6.7- 7.0; the DB-1 melanoma xenografts used here have a pHe=6.7. Understanding tumor cell reaction to the microenvironment is a critical factor in predicting the tumor response to radiotherapy. Purpose: The number of patients presenting with pacemakers in our clinic has increased recently. Following AAPM recommendations, a treatment plan is developed that minimizes the dose to the pacemaker. The most efficient way to measure in vivo dose during treatment is to use MOSFETs or diodes which offer a simple but inaccurate (up to 25% error) method of recording. In this abstract, we analyze the dose measured by these different devices in an attempt to assess pacemaker dose.

Purpose: This work tests the possibility of using IMRT to promote organ sparing for the treatment of CSI. The traditional CSI technique uses opposed lateral fields to treat the brain and posterior fields to treat the spine. Published manuscripts focusing on the possibility of IMRT for CSI have only looked at pediatric cases. Here, we will plan IMRT for CSI on adult patients to quantify the dosimetric gains when compared to traditional techniques.

Background: A third of patients with solid malignancies develop brain metastases. Expected overall survival is 4-7 months depending on age, performance status, and extracranial disease. Standard treatment is controversial; however, the majority of patients receive wholebrain radiation therapy at some point. Vorinostat (suberoylanilide hydroxamic acid, SAHA), an FDA-approved HDAC inhibitor, has been demonstrated to radiosensitize tumor cells in vitro, as assessed by both radiation-induced DNA damage and clonogenic cell survival (Munshi et al. Background: A Wiki is a website which can be directly, openly edited; Wikipedia, a collaborative encyclopedia, is a well-known example. Due to Wiki’s lack of editorial control and formal structure, we hypothesized that the content would be less complete and less accurate than that presented on a formal, peer-reviewed web site. Our goal was to compare the coverage, accuracy, and readability of cancer information from a Wiki (Wikipedia) with a peer-reviewed web site, the patientoriented National Cancer Institute’s Physician Data Query (PDQ) comprehensive cancer database.

Background: Treatment of high-grade glioma consists of fractionated radiation therapy (RT) ± chemotherapy. The incidence/significance of neurological toxicity (NT) in this disease is not known. We evaluated the relationship between acute and chronic NT, and ultimate outcome, as well as risk factors for NT from the RTOG database.

Introduction: Recently Temozolomide (TMZ) has become the more commonly used analog of DTIC-related oral agents. Although the response rates achieved by TMZ alone are less than satisfactory, there is great interest in identifying compounds that could be used in combination therapy. We have previously demonstrated that the bioflavonoid quercetin (Qct) promotes a p53-mediated response in melanoma and sensitizes melanoma to DTIC. Here we demonstrate that Qct also sensitizes cells to TMZ by a mechanism that involves the modulation of a truncated p53 family member, ΔNp73.

Introduction: Tumor cells grow in nutrient and oxygen deprived microenvironments and adapt to the suboptimal growth conditions by altering metabolic pathways. This adaptation process characteristically results in a tumor phenotype that displays anaerobic glycolysis, chronic acidification and aggressive tumor characteristics. Understanding the tumor cell reaction to the microenvironment is a critical factor in predicting the tumor response to hyperthermia. The glucose regulatory molecule, 6-Phosphofructo-2-Kinase/Fructose-2,6-Biphosphatase Isoform-3 (PFKFB3), is a bifunctional enzyme central to glycolytic flux and downstream of the metabolic stress sensor AMP-activated protein kinase (AMPK), which has been shown to activate an isoform of Phosphofructokinase (PFK-2).
