The transcriptional enhancers of the Moloney and Friend murine leukemia viruses (MLV) are important determinants of viral pathogenicity. We used electrophoretic mobility shift and methylation interference assays to study nuclear factors which bind to a region of these enhancers whose sequence is identical between Moloney and Friend viruses and particularly highly conserved among 35 mammalian type C retroviruses whose enhancer sequences have been aligned (E. Mammalian type C retrovirus enhancer factor 1 (MCREF-1) is a nuclear protein that binds several directly repeated sequences (CNGGN6CNGG) in the Moloney and Friend murine leukemia virus (MLV) enhancers (N. R. Manley, M. O'Connell, W. Sun, N. A. Speck, and N. Hopkins, J. Virol. 67:1967-1975, 1993). In this paper, we describe the partial purification of MCREF-1 from calf thymus nuclei and further characterize the binding properties of MCREF-1. MCREF-1 binds four sites in the Moloney MLV enhancer and three sites in the Friend MLV enhancer. Most studies on the biological activity of selenium have focused on its role at the active site in the ubiquitous enzyme, glutathione peroxidase (GSH-Px) which catalyzes the removal of hydrogen peroxide and organic hydroperoxides (1). A connection between selenium deficiency and a wide range of diseases in animals and humans has been reviewed (2). Development of various forms of human cancer (3), failure of the immune system (4), as well as some cardiovascular diseases (5) have been reported or suspected to correlate with selenium deficiency. Parasite-specific antibody responses were detected using an indirect fluorescent antibody (IFA) test in cattle that were naturally or experimentally infected with Neospora parasites. The test was developed using Neospora tachyzoites isolated from an aborted bovine fetus and grown in bovine cell cultures (isolate BPA1). In all cases, infections were confirmed by the identification of Neospora tachyzoites and/or bradyzoite cysts in fetal or calf tissues using an immunoperoxidase test procedure. Fifty-five naturally infected cows that aborted Neospora-infected fetuses had titers of 320-5,120 at the time of abortion. The performance of all shower dip chemicals for sheep lice can vary depending on the active chemical and the conditions under which they are used.

Department of Agriculture trials have indicated that the wettable powders coumaphos and magnesium fluorosilicate were less effective at eradicating sheep lice than were synthetic pyrethroid and organophosphate dipping chemicals.

However, failure to eradicate sheep lice may be the result of several factors, many of them related to management.

With the termination of Western Australia's Sheep Lice Eradication Campaign in 1993 farmers' views about the eradication of sheep lice are of greater relevance now than in the past. Lice control now rests with individual farmers and continuing local lice cell groups .

A recent survey of producers in the Moora Shire provides some key insights into farmers' thoughts about sheep lice and their eradication.

Heterophil chemotaxis using heterophils isolated from the peripheral blood of five commercial broiler chickens naturally infected with staphylococcal bacteria was compared by the modified Boyden-chamber technique with chemotaxis of heterophils from two chickens from the same flock not infected with Staphylococcus (field controls) and from four healthy laboratory control broiler chickens. The infected chickens had gross and histologic lesions of staphylococcal tenosynovitis and osteomyelitis. Staphylococci were isolated from the lesions. Hematologic parameters and histologic lesions of infected chickens also were examined. Compared with field and laboratory controls, Staphylococcus-infected chickens had heterophilic leukocytosis. Heterophil chemotaxis, in response to chemotactic factors generated by three different strains of staphylococcal bacteria, was measured using the modified Boyden-chamber technique. Heterophils were obtained from healthy 6-to-8-week-old broiler chickens. Each bacterial strain generated factors that were chemotactic for chicken heterophils. Factors generated by two pathogenic isolates of Staphylococcus aureus, however, induced significantly greater chemotaxis in chicken heterophils than those generated by a nonpathogenic Staphylococcus isolate.

To test the hypothesis that epidermal rather than dermal mosaicism determines Blaschko's lines in hypomelanosis of Ito (HI), we studied the distribution of chromosomal mosaicism in four patients. In two, mosaicism had not been detected in lymphocytes or dermal fibroblasts, but was clearly shown in epidermal keratinocytes; furthermore, the abnormal cell line was confirmed to the hypopigmented epidermis and the normal epidermis contained only normal cells. Negative findings in the other two patients might be because of mosaicism which was undetected either because it was submicroscopic or because it was present in melanocytes, which have not yet been studied. The power of molecular biology is unleashed with the ability to clone and sequence genes, and then express these genes in heterologous systems. This sets the stage for the full analysis of proteins that are otherwise difficult to isolate and/or purify, especially when present at very low copy number per cell or when isolated from relatively precious materials. The dystrophin gene has been localised to Xp 21.1. Mutations of this gene can lead to the clinical manifestations of Duchenne and Becker muscular dystrophies (DMD/BMD). In the majority of DMD and BMD patients the disease-causing mutation is a deletion detectable by southern analysis or multiplex PCR, however in 30% of patients no deletion is observed using these conventional tests. Using PCR amplification of cDNA it was possible to detect a deletion in the product of the dystrophin gene of one such individual affected with BMD. The disease caused by the protozoan Perkinsus marinus has been a major source of mortality in the eastern oyster, Crassostrea virginica. Variations in susceptibility to P. marinus infection among eastern oysters collected from the Chesapeake Bay and Gulf of Mexico, as well as between eastern and Pacific (Crassostrea gigas) oysters were determined. Since oyster host defense may play a role in determining susceptibility to pathogen infection, cellular and humoral defense activities of the oyster and their interactions with P. marinus were investigated. Tall fescue (Festuca arundinacea Schreb.) is the most commonly grown cool season grass used for pastures in Arkansas. Most tall fescue contains a fungal endophyte (Acremonium coenophialum Morgan-Jones & Gams), which causes fescue toxicosis in livestock and costs cattle producers millions of dollars annually in lost production. Endophyte presence is known to reduce wild mammal populations in areas where tall fescue is prevalent. The endophyte spends its entire life cycle within the plant and is transmitted through the seed. From an activist's perspective, the 80's was very encouraging,-- alternatives gained acceptance, legitimacy and credibility within the toxicology, corporate and regulatory communities. And The Center for Alternatives to Animal Testing (CAAT) at Johns Hopkins University played a significant role in making this acceptance possible.

Western Australia is well placed to eradicate virulent footrot from the State's sheep and goat populations.

Between 1950 and 1993 the proportion of sheep flocks with footrot fell from about 15 per cent to less than 2 percent, and the 1990-92 outbreak has been turned around. One hundred and eighty properties (]. 7 per cent) of flocks, mostly in the high rainfall South-West, are in quarantine for footrot today.

This high level of footrot control is the result of strong support from the sheep industry and years of good cooperation between farmers and the Department of Agriculture.

The Distribution And Occurrence Of Aerobic Heterotrophic Antimicrobic-Producing Bacteria Isolated From The Lower Chesapeake Bay, Linda Lea Gilmer

Three different estuarine microenvironments in the lower Chesapeake Bay were investigated for the occurrence and geographical distribution of antimicrobic-producing bacteria. Samples were taken from the water column, fish skin, and algal surfaces. A total of 1472 bacterial isolates were assayed for antimicrobial activity against fungal, Gram-negative and Gram-positive bacterial test organisms. Of the total bacterial isolates assayed, 5. 2% were active antimicrobic producers. Algal surf aces were shown to have a higher incidence of antimicrobic-producing bacteria than the water column or fish skin. No antimicrobic activity was demonstrated by bacteria isolated from the flounder fish skin. The majority of the …
