Early left ventricular (LV) remodeling following pediatric cardiac transplantation has not been described. To identify patterns and determinants of change in left ventricular mass and volume posttransplant, we studied 125 consecutive children who underwent cardiac transplantation between January 1, 1989 and July 31, 1993. Two-dimensional imaging-directed M-mode echocardiograms were studied weekly until 26 weeks post-transplant. LV mass and volume (indexed to BSA1.5) were measured. LV mass index increased until 3 weeks post-transplant, and then decreased. The mean decrement in LV mass index after 8 weeks post-transplant (relative to baseline) was significantly larger in patients with donor-recipient weight ratio > 1.5 compared …

Little is known of the effects of low concentrations of inhaled nitric oxide (NO) on pulmonary mechanics and lung volumes in premature newborn infants with lung disease. Cardiopulmonary function tests including blood gas analyses were obtained from eleven premature lamb models with respiratory distress syndrome to determine if compliance, airways resistance, tidal volume, functional residual capacity, and gas exchange were affected. The effect of inhaled NO on the direction of shunting through the ductus arteriosus was determined using echocardiography.

Retinoic acid (RA) is an important regulator of growth and differentiation in many cell types, including bone. However, its effects on human osteoblast cell growth and differentiation have not been well studied. Therefore, I investigated the effect of RA on proliferation and differentiation of normal human bone cells (HBCs) and human osteosarcoma, SaOS-2 cells. RA decreased baseline as well as serum-stimulated proliferation in normal HBCs. To determine the effect of RA on differentiation, expression of several osteoblastic differentiation markers were studied. Muscle contraction is thought to be accomplished by sliding of myosin filaments along actin filaments. Although actomyosin naturally uses ATP as an energy source, it can also use other nucleoside triphosphates (NTP) as substrates. In this work, the dependence of rate and equilibrium constants of the various steps in the muscle contraction mechanism upon nucleotide structure was investigated to unravel the dependence of the mechanical properties of the muscle upon changes in different biochemical steps of the mechanism.

The tau mutation affects the circadian system of the golden hamster primarily by decreasing the period of the activity cycle from 24 to 20 hours. To study the effect of this mutation on reproductive function in the golden hamster, 3 experiments have been designed. In the first experiment, wild-type and tau female hamsters will be maintained in conditions of 14:10 LD and 11.7:8.3 LD, respectively. Blood samples will be taken and analyzed by radioimmunoassay (RIA) to determine the timing of the proestrus LH surge. Precise connections in many mammalian nervous systems require a great deal of remodeling during development. In the visual system, many excess synapses are originally formed in the lateral geniculate nucleus and striate cortex. Only the correct set of axon terminals are retained during normal development, while imprecise ones withdraw. The mechanism by which only correct axons are retained requires neural activity, and may be regulated by specific receptors at synapses.

The transmission of neural signals at these synapses is carried out in part by the glutamate-activated NMDA receptor. It is hypothesized that NMDA receptor activation plays a crucial role in …

The myogenic reactivity provides one of the principal mechanisms for blood flow autoregulation. The aims of the performed studies described in this dissertation were to test the role of [Ca2+]i and MLC phosphorylation in arteriolar myogenic reactivity and further examine the source(s) of activator Ca2+ required to initiate and maintain myogenic vasoconstriction. In addition, the possible underlying mechanism of contractile protein expression was also addressed.

We investigated the developmental expression and cellular resolution of connexin32 and 43 mRNA in the rat brain using in situ hybridization. Utilizing 35S-labelled probes, in situ hybridization was performed on sections of embryonic day 20 and postnatal days 3, 10, 15, 30 and adult brain. Connexin32 mRNA was first detected in brainstem nuclei at postnatal day 3 and in the midbrain at postnatal day 15. The level of this message continued to increase to postnatal day 30 where the level of message reached a plateau or slightly decreased by adulthood.
