The potato aphid, Macrosiphum euphorbiae, is an important agricultural pest that causes economic losses to potato and tomato production. To establish the transcriptome for this aphid, RNA-Seq libraries constructed from aphids maintained on tomato plants were used in Illumina sequencing generating 52.6 million 75±105 bp paired-end reads. The reads were assembled using Velvet/Oases software with SEED preprocessing resulting in 22,137 contigs with an N50 value of 2,003bp. After removal of contigs from tomato host origin, 20,254 contigs were annotated using BLASTx searches against the non-redundant protein database from the National Center for Biotechnology Information (NCBI) as well as IntereProScan. …

Although regarded as a low-risk commodity, wheat flour-based mixes have been implicated in several food safety incidents. The present thesis is a compilation of five scientific manuscripts on the effect of weather variation, milling steps and implementation of pre-milling interventions on the microbiological quality and safety of wheat and milled products. The first manuscript is a review of the microbiological quality and safety of wheat-based products. Despite the low water activity, wheat flour may harbor dormant but viable microorganisms, which could lead to safety concerns when flour is used to produce refrigerated dough products. Fungal staining (black point) of wheat can reduce the quality of grain. The most serious problem is a discolouration of products. Discolouration is a consequence of infection by microorganisms. The most likely cause in Western Australia is a species of the common fungus Alternaria. Infection and discolouration occur between flowering and grain maturity, and the optimum environmental conditions are probably consecutive days of high relative humidity together with warm temperatures.

Sterols from mixed stages of the com root lesion nematode, Pratylenchus agilis, and uninfected corn root cultures were analyzed by gas chromatography-mass spectrometry. Twenty-eight sterols were identified in P. agilis, including nine not previously detected in nematodes. The major sterols were 24-ethylcholest-22-enol, 24-ethylcholesta-5,22-dienol, 24-methy1cholestanol, 24-ethylcholestanol, isofucostanol, and 24-ethylcholesterol. The principal corn root sterols were 24-ethylcholesta-5,22-dienol, 24-methylcholesterol, 24-ethylcholesterol, isofucosterol, and cycloartenol. Therefore, the major metabolic transformation of sterols by P. agilis was saturation of the sterol nucleus. In addition, very small amounts of 4α-methylsterols were biosynthesized by P. agilis.
