A murine model was utilized to test the influence of heavy metal protoporphyrins on burn conversion, heme oxygenase – 1 (HO-1) expression, and inflammation. Heavy metal protoporphyrins, such as cobalt protoporphyrin (Co PP) and tin protoporphyrin (Sn PP), were used to influence the heme oxygenase activity. The effects of these heavy metal protoporphyrins on burn wound conversion were examined using a burn comb model in rats. In addition to assessing the extent of conversion, HO-1 expression and parameters of inflammation were also examined in the area of injury (interspace region) subject to conversion. The main function of the cardiovascular system is to deliver a sufficient quantity of oxygenated blood to the tissues, cells, and organs of the body in order to provide the cells with essential nutrients for metabolism and for the removal of waste products. All cells require and utilize oxygen. Oxygen is transported to various cells and tissues via red blood cells flowing through the microcirculation of an organism. Our recent studies have shown that that erectile dysfunction (ED) drugs including Sildenafil (Viagra), Vardenafil (Levitra) and Tadalafil (Cialis) enhance killing of several types of cancer cells by anticancer drug, Doxorubicin (DOX). We observed increased cell death by apoptosis in response to the combined treatment with ED drugs and DOX. However, the mechanism of such enhancement of cell death by combined treatment of ED drugs and DOX is not fully understood. Cushing's syndrome and glucocorticoid therapy lead to central obesity, insulin resistance, and symptoms of altered energy regulation similar to those observed in the metabolic syndrome. We hypothesized that excess glucocorticoids alter energy sensing/signaling in skeletal muscle through mediation of the LKB1/AMPK signaling pathway. To test this hypothesis, three 100 mg pellets of corticosterone were implanted subcutaneously in each of nine rats for two weeks. Responses were compared with sham operated controls fed ad libitum or food restricted to produce the body weights similar to the treatment group rats. Background and Purpose: Whole-body vibration (WBV) is a relatively new form of exercise training that may influence muscle performance. This study investigated the acute effects of high (26 Hz) and low (2 Hz) frequency WBV on isometric muscle torque of the quadriceps and hamstrings in persons with multiple sclerosis (MS).

The purpose of this study was to use Cone Beam Computed Tomography (CBCT) images to quantitatively evaluate buccal bone changes of maxillary first premolar (P1), second premolar (P2) and first molar (M1) shortly after Rapid Maxillary Expansion (RME) and at the end of orthodontic treatment, as well as to evaluate the variables that may be associated with the short-term and long-term changes. High dietary intake of selenium or isoflavones reduces risk factors for prostate cancer. We tested whether combined supplementation of these two dietary components would reduce prostate cancer risk factors in rats more than supplementation of each component individually. Methods Male Noble rat pups were exposed from conception to diets containing an adequate (0.33-0.45 mg/kg diet) or high (3.33-3.45 mg/kg) concentration of selenium as Se-methylselenocysteine and a low (10 mg/kg) or high (600 mg/kg) level of isoflavones in a 2 x 2 factorial design. Kisspeptin, a neuroendocrine regulator of gonadotropin releasing hormone, is hypothesized to integrate nutrition and hormones critical to metabolism and the regulation of reproduction. As aquatic adults, eastern red-spotted newts (Notophthalmus v. viridescens) are generally green with two rows of dorsal spots, which vary in number between individuals and range in colour from orange to red. The function of these spots is unknown, but it is possible that they serve as sexual ornamentations and we examined this hypothesis by testing for sexual dimorphism in spot characteristics. This study examined the effects of cycling posture on subsequent running performance similar to the transition phase of a triathlon. Experienced, non-elite triathletes completed two trials of a cycle-run transition. During the last three minutes of a 30 minute cycling bout, at power output equal to lactate threshold, subjects either remained seated (SEAT), or alternated seated and standing cycling (30 s at a time; ALT). The consumption of a high-fat meal induces transient vascular dysfunction. Aerobic exercise enhances vascular function in healthy individuals. Our purpose was to determine if different levels of aerobic capacity impact vascular function, as measured by flow mediated dilation, following a high-fat meal. Flow mediated dilation of the brachial artery was determined before, two- and four-hours postprandial a high-fat meal in young males classified as highly trained (n = 10; VO2max = 74.6 ± 5.2 ml·kg·min-1) or moderately active (n = 10; VO2max = 47.3 ± 7.1 ml·kg·min-1). Flow mediated dilation was reduced at two- (p < 0.001) and four-hours (p < 0.001) compared to baseline for both groups but was not different between groups at any time point (p = 0.108). Triglycerides and insulin increased at two- (p < 0.001) and four-hours (p < 0.05) in both groups. LDL-C was reduced at four-hours (p = 0.05) in highly trained subjects, and two- and four-hours (p ≤ 0.01) in moderately active subjects. HDL-C decreased at two- (p = 0.024) and four-hours (p = 0.014) in both groups. Glucose increased at two-hours postprandial for both groups (p = 0.003). Our results indicate that a high-fat meal results in reduced endothelium-dependent vasodilation in highly trained and moderately active individuals with no difference between groups. Thus, high aerobic capacity does not protect against transient reductions in vascular function after the ingestion of a single high-fat meal compared to individuals who are moderately active.

The aim of the present study was to evaluate the post-exercise blood pressure (BP) responses to two different methods of resistance exercise (RE, traditional multiple set and tri-set). The present study investigated the rate of systolic blood pressure recovery (SBPR) amongst three male age groups. Ninety-six apparently healthy, non-athletic adult males (48 young [23.91±4.58 years], 32 middle-aged [41.78±5.98 years] and 16 elderly [62.87±2.57 years]) participated in the study. Subjects performed a maximal-effort ergometer exercise test. Maximal oxygen uptake (VO2max) was measured during the exercise protocol; heart rate (HR) and SBP were measured before exercise (after 10 and 15 minutes of rest), during exercise (at 2-minute intervals), and at the first minute of post-exercise recovery and subsequently at 2-minute intervals until the recovery of SBP. The authors lead a study abroad trip to Beijing, China during the 2008 Summer Olympic Games. Included are reflections from the students who participated. Throughout the visit, students had the opportunity to complete learning outside of the traditional classroom setting. Exposure to stress can lead to either increased stress vulnerability or enhanced resiliency. Laboratory rats are a key tool in the exploration of basic biobehavioral processes underlying individual differences in the effect of stress on subsequent stressors' impact. The Occidental low (LoS) and high (HiS) saccharin-consuming rats, which differ in emotional reactivity, are useful in this effort. In the present study, footshock affected acoustic startle amplitude 4 h later among LoS but not HiS rats. Firefighters and police have different physical job requirements and selection processes. Firefighters have on-duty exercise time and police do not. This could affect body composition and strength. The purpose of this investigation was to compare body composition and bench press strength between firefighters and police officers in the Charlotte-Mecklenburg departments. Current 2006 fitness records were analyzed to compare firefighters and police (N = 2330). The 5K handicap (5KH), designed to eliminate the body weight (BW) and age biases inherent in the 5K run time (RT), yields an adjusted RT (RTadj) that can be compared between runners of different BW and age. Vascular reactivity changes dramatically during postnatal maturation due in large part to developmental changes in myofilament calcium sensitivity. Recent findings suggest that reactivity of the thick filament component of calcium sensitivity is upregulated in fetal compared to adult arteries. In light of these findings, the present study tests the hypothesis that upregulation of fetal thick filament reactivity is due to upregulation of myosin light chain kinase (MLCK) activity. To test this hypothesis, MLCK abundance and its activity is measured in intact arteries. The results indicate that MLCK abundance is 6.03 ± 0.96 fold greater in adult than in fetal arteries. …

The contention that quantitative profiles of biomolecules contain information about the physiological state of the organism has motivated a variety of high-throughput molecular profiling experiments. However, unbiased discovery and validation of biomolecular signatures from these experiments remains a challenge. Here we show that the Arabidopsis thaliana (Arabidopsis) leaf ionome, or elemental composition, contains such signatures, and we establish statistical models that connect these multivariable signatures to defined physiological responses, such as iron (Fe) and phosphorus (P) homeostasis. Vertebrate development is regulated by cellular communication by mechanisms of cell fate and cell behavior. These crucial mechanisms are regulated by cellular signaling and in the case of cell fate, cellular signaling results in transcription of developmentally important genes. Communication between cells can also result in regulation of cell behavior by acting on cytoskeletal elements rather than nuclear factors. One of the cellular signals that regulate both cell fate and cell behavior is the family of Wnt signaling molecules. The exact mechanisms that regulate limb outgrowth the mouse embryo are unknown. Although there are several models, we favor a hypothesis where cells become polarized by signals secreted from the AER which orient their cell migration and/or divisions causing limb outgrowth. Clonal analysis has provided a mechanism to study cell behavior. We have generated a targeting construct containing the Fgf8 inhibitor, Sprouty2, in order to generate mutant clones for behavioral analyses in the limb. Osteoarthritis (OA) is a degenerative joint disease that affects more than 60% of Americans 65 and older. Because human subjects and samples are not readily available for research, animal models are an invaluable resource for the study of OA. Disproportionate micromelia (Dmm) is one such model that develops OA early in life due to a deletion in the c-propeptide of the Col2a1 gene. Light microscope analysis of the articular cartilage in Dmm has been completed, but is insufficient to show the cellular effects of the deletion mutation in Dmm in adequate detail. Recent evidence makes a compelling case that U.S. Army, Navy, and Air Force health-related physical fitness tests penalize larger, not just fatter, service members. As a result, they tend to receive lower scores than their lighter counterparts, the magnitude of which can be explained by biological scaling laws. Larger personnel, on the other hand, tend to be better performers of work-related fitness tasks such as load carriage, heavy lifting and materiel handling.
