The Stringent Response In Pseudomonas Aeruginosa Influences The Phenotypes Controlled By The Gac/Rsm System, Michael Shawn Hooker

Pseudomonas aeruginosa is a ubiquitous, opportunistic pathogen that causes acute and chronic infections. Infection is typically initiated via motile and virulent strains. After exposure to stressors, acute infections make both genotypic and phenotypic switches to a chronic, sessile strain. This is due to intricate regulatory networks directing gene expression in response to stressors. One network, GacA/GacS, has been established to control virulence factors. The stringent response of bacteria is mediated by alarmones produced primarily by RelA which responds to starvation.

Microbial communities contain densely packed cells where competition for space and resources are fierce. These communities are generally referred to as biofilms and provide advantages to individual cells against immunological and antimicrobial intervention, dehydration, and predation. High intracellular pools of cyclic diguanylate monophosphate (c-di-GMP) cause cells to aggregate during biofilm formation through the production of diverse extracellular polymers. Genes that encode c-di-GMP catalytic enzymes are commonly mutated during chronic infections where opportunists display enhanced resistance to phagocytosis and antibiotics. The marine cyanobacteria Prochlorococcus and Synechococcus are the most abundant phototrophs in the oceans. They cohabit the oligotrophic ocean and thus have coevolved together, yet they have distinctly different methods for harvesting light. Synechococcus, like other cyanobacteria, possess phycobilisomes with various combinations of phycobiliproteins to capture wavelengths of light not otherwise available to chlorophyll. Prochlorococcus lack phycobilisomes and use divinyl chlorophyll b (Chl b2) as their primary accessory pigment to divinyl chlorophyll a (Chl a2) to capture light energy. A molecular padlock assay was developed and assessed for detection of Listeria monocytogenes that operated in crude plant extracts. The molecular padlock assay was developed by Liu et al. (1996) and modified by Lizardi et al. (1998). We further modified and described a padlock probe that detected L. monocytogenes oligonucleotide, cDNA and genomic DNA containing a 16s rRNA sequence (GenBank Acc. No. X56 153). This technique was effective in the presence of crude potato leaf extracts in contrast to PCR, which failed to detect the presence of L. monocytogenes targets in crude leaf extracts. Infectious salmon anemia virus (ISAV) is an emerging pathogen of farmed Atlantic salmon (Salmo salar). The development of an effective ISA virus vaccine is a high priority for salmon producers in the U.S. and elsewhere. The process of developing a recombinant vaccine requires complete genetic characterization of the virus. Toward this end we have cloned, sequenced and determined the organization of the eight segments of single-stranded RNA from ISA virus isolate CCBB. The virus was grown in cell culture and purified by density gradient ultracentrifugation. Viral RNA was isolated from purified ISAV and used in the construction of two different …
