Climatic warming is altering species distributions and ecosystem functions across the globe. Wood is an important carbon pool and the fungal communities in wood are relatively simple compared to those in soil. These factors make decomposing wood an ideal system for exploring the influence of decomposer community on the response of decomposition to warming. My research has focused on the effects of warming wood decomposition rates and wood decomposing communities. Of these viruses, cyanophages are considered especially important because they infect and lyse cyanobacteria, which are some of the main primary producers in marine environments. Bacteria have been widely reported to use quorum sensing (QS) systems, which employ small diffusible metabolites to coordinate gene expression in a population density dependent manner. In Proteobacteria, the most commonly described QS signaling molecules are N-acyl-homoserine lactones (AHLs). Recent studies suggest that members of the abundant marine Roseobacter lineage possess AHL-based QS systems and are environmentally relevant models for relating QS to ecological success. As reviewed here, these studies suggest that the roles of QS in roseobacters are varied and complex. Background

The distribution of genetic diversity of Toxoplasma gondii in wildlife is of interest to understand the transmission of this parasite in the environment. Limited information on T. gondii genotypes has been reported in wildlife in China. The objective of this study was to carry out the genetic characterization of T. gondii isolates from wild animals on the Qinghai-Tibet Plateau.

Methods

Using PCR and multilocous polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technology, we detected genetic diversity of T. gondii isolates from Qinghai vole, Plateau pika and Tibetan ground-tit in these regions.

Background

Toxoplasma gondii is a widely prevalent protozoan parasite that causes serious toxoplasmosis in humans and animals. The present study aimed to determine the genetic diversity of T. gondii isolates from pigs in Jiangxi, Sichuan, Guangdong Provinces and Chongqing Municipality in China using multilocous polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technology.

Methods

The Roseobacter clade is a widely distributed, abundant, and biogeochemically active lineage of marine alpha-proteobacteria. Members of the Roseobacter lineage are prolific surface colonizers in marine coastal environments, and antimicrobial secondary metabolite production has been hypothesized to provide a competitive advantage in colonization. In this work, Phaeobacter sp. strain Y4I was found to produce the water soluble, blue pigment indigoidine via a nonribosomal peptide synthase-based biosynthetic pathway encoded by a novel series of genetically linked genes, termed igiBCDFE. Roseobacters are an abundant and trophically versatile lineage of marine bacteria that are especially dominant in coastal salt marshes. Central to understanding of how members of the Roseobacter clade contribute to biogeochemical cycling in the world’s oceans is how these bacteria physiologically respond to mixtures of usable growth substrates present in their environmental niches. A prior study provided evidence that bacterioplankton transcripts most closely related to the Roseobacter Sagitulla stellata E-37 are among the most abundant in coastal systems for biogeochemically significant processes of N, P, and S cycling. Culture-independent methods have revolutionized environmental microbiology and geomicrobiology studies and next-generation sequencing and metagenomics techniques continue to reveal the vast genetic diversity of microorganisms. But, these approaches provide comparatively little quantitative information about the roles that naturally occurring microbial gene variations play in local biogeochemical cycling. We present the genome of a cyanosiphovirus (KBS2A) that infects a marine Synechococcus sp. (strain WH7803). Diarrheal disease pathogens remain a major concern in developing countries as rotavirus is the leading cause of hospitalization of young children worldwide. A recent study has shown shallow groundwater in rural Bangladesh to be contaminated with bacterial and viral pathogens, but found no correlation between rotavirus and any fecal indicator or environmental parameter during the monsoon season of July, 2009. Background

Cansiliella servadeii (Coleoptera) is an endemic troglobite living in deep carbonate caves in North-Eastern Italy. The beetle constantly moves and browses in its preferred habitat (consisting in flowing water and moonmilk, a soft speleothem colonized by microorganisms) self-preens to convey material from elytra, legs, and antennae towards the mouth. We investigated its inner and outer microbiota using microscopy and DNA-based approaches.

Results

The Odontogenic Ameloblast-associated Protein (ODAM) is expressed in a wide range of normal epithelial, and neoplastic tissues, and we have posited that ODAM serves as a novel prognostic biomarker for breast cancer and melanoma. Transfection of ODAM into breast cancer cells yields suppression of cellular growth, motility, and in vivo tumorigenicity. Herein we have extended these studies to the effects of ODAM on cultured melanoma cell lines.

Microbial communities associated with the human oral cavity are complex, and many oral microbes have yet to be cultured. These uncultured community members are of interest ecologically and phylogenetically, and a number of uncultured species have been positively correlated with oral diseases such as periodontitis. Thus, an approach was adapted to selectively separate single cells from mixed populations of oral bacteria and obtain genomic information for uncultured community members. A combination of fluorescent labeling, cell sorting with flow cytometry and multiple displacement amplification was used to obtain sufficient genomic material for whole-genome pyrosequencing.

Quorum sensing (QS) is a phenomenon that allows bacteria to communicate with each other. Small molecules known as autoinducers are synthesized and released by bacteria, and once enough members of the community are around to ensure survival, i.e. quorum, a phenotype, e.g. bioluminescence, is expressed. There are two types of QS molecules, intra- and inter-species.

Microbes capable of polyaromatic hydrocarbon (PAH) biodegradation can be used to remediate soils contaminated with these persistent pollutants. To monitor in situ PAH-biodegradation, the bioluminescent bio-reporter Pseudomonas fluorescens HK44, containing a lux luminescent gene cassette inserted into its naphthalene degradation operon, was released into PAH-contaminated soil in lysimeters in 1996. Three treatments were imposed: strain HK44 mixed with PAH-contaminated soil (PAH+, HK44+; n=3); strain HK44 mixed with uncontaminated soil (PAH–, HK44+; n=2) and PAH-contaminated soil alone (PAH+, HK44–; n=1). A single cultured marine organism, Nanoarchaeum equitans, represents the Nanoarchaeota branch of symbiotic Archaea, with a highly reduced genome and unusual features such as multiple split genes.

Results

The first terrestrial hyperthermophilic member of the Nanoarchaeota was collected from Obsidian Pool, a thermal feature in Yellowstone National Park, separated by single cell isolation, and sequenced together with its putative host, a Sulfolobales archaeon. Both the new Nanoarchaeota (Nst1) and N. equitans lack most biosynthetic capabilities, and phylogenetic analysis of ribosomal RNA and protein sequences indicates that the two form a deep-branching archaeal lineage. Toxoplasma gondii is an opportunistic pathogenic protozoan parasite, which infects approximately one third of the human population worldwide, causing opportunistic zoonotic toxoplasmosis. G protein-coupled receptors (GPCRs) are the target of 30-50% of all prescribed drugs for human medicine and are therefore the subject of intense study by the scientific community. It has been recognized recently that compounds called allosteric modulators can regulate GPCR activity by binding a GPCR at sites not occupied by the normal receptor-activating molecule. Such allosteric compounds are desirable drug candidates as they may produce fewer toxic side-effects than standard drugs that target GPCRs.
