We examined the physiological responses of steady-state iron (Fe)-replete and Fe-limited cultures of the biogeochemically critical marine unicellular diazotrophic cyanobacterium Crocosphaera at glacial (19 Pa; 190 ppm), current (39 Pa; 380 ppm), and projected year 2100 (76 Pa; 750 ppm) CO2 levels. Rates of N2 and CO2 fixation and growth increased in step with increasing partial pressure of CO2 (pCO2), but only under Fe- replete conditions. N2) and carbon fixation rates at 75 Pa CO2 were 1.4-1.8-fold and 1.2-2.0-fold higher, respectively, relative to those at present day and glacial pCO2 …

Panulirus argus Virus 1 (PaV1) is an emerging disease in Caribbean spiny lobster Panulirus argus. It is considered a threat to the lobster industry in the Florida Keys. In order to understand the infection dynamics of the PaV1 virus in the lobster host, a sensitive and specific fluorescence in situ hybridization (FISH) assay was developed for diagnosis of PaV1 in lobsters. The lower limit of detection using the 110-bp DNA probe in a dot-blot hybridization for PaV1 was 10 pg of cloned DNA template and 10 ng of genomic DNA extracted from hemolymph of diseased spiny lobster. Striped bass (Morone saxatilis) in Chesapeake Bay, USA, are experiencing an epizootic of mycobacteriosis. This disease, caused by bacteria in the genus Mycobacterium, causes granulomatous lesions of the skin and viscera. Diseased fish are often emaciated, and fish with skin lesions may be significantly disfigured. The overall goal of this work was to examine aspects of the pathobiology of mycobacteria in striped bass via laboratory exposure studies and cellular assays. Two methods used to measure dinitrogen (N2) fixation (acetylene reduction and 15N2 uptake) often result in different N2 fixation rates. Part of the discrepancy may arise from the observation that Trichodesmium can release a fraction of their recently fixed N2 as dissolved organic nitrogen (DON) and/or ammonium (NH4 +). To resolve outstanding issues regarding N2 fixation and the production of dissolved combined nitrogen (N) by Trichodesmium, we conducted a comprehensive analysis of N2 fixation and the production of DON and NH4+ in cultures of Trichodesmium IMS101. An Investigation Of Dissolved Organic Matter In A Shallow Coastal Bay Subject To Aureococcus Anophagefferens Blooms, Jean-Paul Simjouw

Aureococcus anophagefferens, the pelagophyte responsible for brown tide blooms, was identified in Chincoteague Bay in 1997 and has “bloomed” there since at least 1998. Aureococcus anophagefferens is capable of using dissolved organic nitrogen (DON) and dissolved organic carbon (DOC) substrates to support growth, and this utilization is hypothesized to give the organism a competitive advantage relative to other phytoplankton when inorganic nutrient concentrations are low or depleted. Because previous studies suggest dissolved organic matter (DOM) is important in initiating and sustaining brown tide blooms, a field study of the variations in DOC concentration and DOM composition was performed at …

Perkinsus marinus is the causative agent of the oyster disease Dermo in the eastern oyster, Crassostrea virginica. In vitro propagation of the parasite has led to the establishment of multiple isolates by several investigators. Little work, however, has been done to characterize different isolates. In this study multiple isolates were examined for genetic and biochemical diversity. at two loci, the ITS region and the ATAN region, there was as much intra-isolate genetic variation among DNA sequences of some isolates as there was inter-isolate variation.
