The Evolution Of Spinosad Resistance In Colorado Potato Beetles (Leptinotarsa Decemlineata), Coby Michael Klein

Colorado potato beetles (Leptinotarsa decemlineata) are a major pest of cultivated potato plants worldwide. They are well-known for their ability to rapidly evolve resistance to all major classes of pesticides. Defoliation of potato plants by L. decemlineata can reduce potato yields by a considerable margin. The damage done by resistant beetles is steep and much research is focused on developing new chemical controls, especially those derived from naturally occurring compounds. Egg, larval, and adult life stages of Colorado potato beetle, Leptinotarsa decemlineata (Say), were observed feeding on or attached to a previously undocumented host plant belonging to the genus Chamaesaracha in eastern Colorado on July 2017. At one site, L. decemlineata were more abundant on Chamaesaracha sp. than the accepted ancestral host plant, Solanum rostratum (Dunal). While future studies should confirm the ancestral status of the observed L. decemlineata and suitability of Chamaesaracha sp. for completion of development, our observations suggest a need for further characterization of the ancestral host range of L. decemlineata.

The Colorado potato beetle is one of the most challenging agricultural pests to manage. It has shown a spectacular ability to adapt to a variety of solanaceaeous plants and variable climates during its global invasion, and, notably, to rapidly evolve insecticide resistance. To examine evidence of rapid evolutionary change, and to understand the genetic basis of herbivory and insecticide resistance, we tested for structural and functional genomic changes relative to other arthropod species using genome sequencing, transcriptomics, and community annotation. The Colorado potato beetle, Leptinotarsa decemlineata, is a significant pest of potato, and its impact on agriculture is measured on a global scale. The beetle is mainly controlled by neonicotinoid insecticides, however, resistance development is a growing concern. Resistance to neonicotinoids is thought to involve elevated activity of detoxifying enzymes and xenobiotic transporters that break-down and excrete insecticide molecules. Here, using mRNA sequencing, I identified multiple detoxifying enzyme and xenobiotic transporter genes transcriptionally up-regulated in a neonicotinoid resistant strain of beetles. The adaptation of herbivorous insects to their host plants is hypothesized to be intimately associated with their ubiquitous development of resistance to synthetic pesticides. However, not much is known about the mechanisms underlying the relationship between detoxification of plant toxins and synthetic pesticides. To address this knowledge gap, we used specialist pest Colorado potato beetle (CPB) and its host plant, potato, as a model system. Next-generation sequencing (454 pyrosequencing) was performed to reveal the CPB transcriptome. Factors influencing the susceptibility of adult Colorado potato beetle (CPB), Leptinotarsa decemlineata (Say), to the fungal entomopathogen, Beauveria bassiana (Bals.), were studied. In an assay comparing trials between a): laboratory reared, non-diapausing beetles and b): field collected, post-diapausing beetles, survival of B. bassiana treated beetles was higher for non-diapausing adults, however, control mortality was higher for post-diapausing adults. In a similar assay with field collected pre- and post-diapausing beetles, survival of B. bassiana treated individuals was higher for post-diapause adults and control mortality was higher for post-diapausing adults than pre-diapausing beetles. This work is an investigation of:( 1) mortality of the Colorado potato beetle (CPB), Leptinotarsa decemlineata (Say), under different modes of exposure to the pathogen Beauveria bassiana (Bals.) Vuill., (2) host and environmental factors affecting sporulation, production, and viability of B. bassiana conidia from infected cadavers of CPB larvae, and (3) transmission of the disease in CPB populations. The research leads to an improved understanding of the process of transmission of B. bassiana in CPB populations. This may enhance its use as a biological control agent for this pest insect. Given the ongoing improvement of computer technology and validation of past efforts, scientists can now combine several agricultural submodels into one holistic and user-friendly computer application. Beauveria bassiana (Balsamo) Vuill. (Bb), a fungal pathogen of the Colorado potato beetle has been used to control CPB in Europe. Preliminary studies in the United States gave promising results for CPB control by using the fungus. Based on these studies, a three-year pilot program was initiated to evaluate the potential for using Bb o control the Colorado potato beetle in the U.S. We report herein on the results obtained in southern Maine from using this mycoinsecticide to control the beetle.

It is probable that more than one insect species are capable of spreading some of the potato virus diseases. This conclusion was also indicated by repeated observations in the dry-land section of western Nebraska. While aphids are abundant and are probably the chief vectors of potato virus diseases in the more humid sections of the country, they are usually scarce in the western upland plains area and are not present in large enough numbers to account for the rapid spread of spindle tuber and unmottled curly dwarf which sometimes occurs.
