Browntail Moth (BTM; Euproctis chrysorrhoea) is a non-native species currently outbreaking in Maine. BTM are polyphagous folivores and feed on a variety of woody plant species, including many economically and ecologically important ornamental and forest trees such as oaks and apples. Human health is also a concern as BTM larvae produce urticating hairs that can cause severe dermatitis in people. New monitoring and management programs are urgently needed, with the current population densities reaching a 100-year high.

The Maine Forest Service monitors BTM populations by visually assessing winter nest densities and defoliation patterns, both time and labor intensive. This …

BACKGROUND: Biopesticides and transgenic crops based on Bacillus thuringiensis (Bt) toxins are extensively used to control insect pests, but the rapid evolution of insect resistance seriously threatens their effectiveness. Bt resistance is often polygenic and complex. Mutations that confer resistance occur in midgut proteins that act as cell surface receptors for the toxin, and it is thought they facilitate its assembly as a membrane-damaging pore. However, the mechanistic details of the action of Bt toxins remain controversial.

BACKGROUND: Western corn rootworm (WCR; Diabrotica virgifera virgifera) field-evolved resistance to transgenic maize expressing the Cry3Bb1 protein derived from Bacillus thuringiensis (Bt) has been confirmed across the United States Corn Belt. Although use of pyramided hybrids expressing Cry3Bb1 + Cry34/35Ab1 has increased in recent years to mitigate existing WCR Bt resistance, susceptibility of Nebraska WCR populations to this rootworm–Bt pyramid has not been assessed. Plant-based bioassays were used to characterize the susceptibility of WCR populations to Cry3Bb1 and Cry3Bb1 + Cry34/35Ab1 maize. Host-pathogen interactions are central components of ecological networks where the MAPK signaling pathways act as central hubs of these complex interactions. We have previously shown that an insect hormone modulated MAPK signaling cascade participates as a general switch to trans-regulate differential expression of diverse midgut genes in the diamondback moth, Plutella xylostella (L.) to cope with the insecticidal action of Cry1Ac toxin, produced by the entomopathogenic bacterium Bacillus thuringiensis (Bt). The molecular mechanisms of insect resistance to Cry toxins generated from the bacterium Bacillus thuringiensis (Bt) urgently need to be elucidated to enable the improvement and sustainability of Bt-based products. Although downregulation of the expression of midgut receptor genes is a pivotal mechanism of insect resistance to Bt Cry toxins, the underlying transcriptional regulation of these genes remains elusive. The western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte, is an economically important pest species of field corn (Zea mays L.) in the U.S. Corn Belt. Yield losses and control costs exceed greater than $1 billion annually. Over the past 10+ years, growers have adopted transgenic corn hybrids expressing rootworm-active Bacillus thuringiensis (Bt) proteins as a primary WCR management tactic. Field-evolved resistance to Cry3Bb1 and mCry3A proteins expressed in single-trait Bt hybrids has been confirmed in some areas of Nebraska. Greater than expected injury by western corn rootworm (WCR) (Diabrotica virgifera virgifera LeConte) to Cry3Bb1 expressing maize hybrids (Zea mays L.) has been reported in southwestern Nebraska. Affected areas of some fields are often associated with high pH calcareous soils where maize growth is poor and iron chlorosis is common. Understanding the behavior of pests targeted with Bacillus thuringiensis Berliner (Bt) crops is important to define resistance management strategies. Particularly the study of larval movement between plants is important to determine the feasibility of refuge configurations. Exposure to Bt maize, Zea mays L. (Poaceae), has been suggested to increase larval movement in lepidopteran species but few studies have examined the potential for resistance to interact with behavioral responses to Bt toxins. Transgenic plants have been widely adopted by growers to manage the western corn rootworm, Diabrotica virgifera virgifera LeConte, in field corn. Because of reduced efficacy in some Nebraska fields after repeated use of Cry3Bb1-expressing hybrids, single plant bioassays were conducted in 2012 and 2013 to characterize the susceptibility of western corn rootworm populations to the rootwormactive proteins Cry3Bb1, mCry3A, and Cry34/35Ab1. Results demonstrate that there are heritable differences in susceptibility of Nebraska western corn rootworm populations to rootworm-active Bt traits. Crops producing insecticidal toxins from the bacterium Bacillus thuringiensis (Bt) are widely planted and enable management of key insect pests while reducing the use of conventional insecticides. However, the evolution of Bt resistance could diminish these benefits. Fitness costs of Bt resistance occur in the absence of Bt toxin when individuals with resistance alleles show a reduction in fitness relative to susceptible individuals, and they can delay the evolution of resistance. Ecological factors including host-plant variety can affect the magnitude of fitness costs, and consequently, the degree to which fitness costs delay resistance. Four sunflower accessions were compared with a susceptible check, hybrid ‘894’, in the greenhouse to determine their resistance to the banded sunflower moth, Cochylis hospes Walsingham, and their interaction with Bacillus thuringiensis Berliner variety kurstaki. Antibiosis, expressed as lower larval weights, was detected in all of the accessions. In addition to being antibiotic, sunflower accession Ames 3291 was antixenotic to banded sunflower moth oviposition and exhibited an additional impact on larval weight when B. thuringiensis was applied. A study was conducted in 1996 and 1997 to determine if honey bees, Apis mellifera L., could vector Bacillus thuringiensis Berliner variety kurstaki from hives equipped with a pathogen applicator to sunflower capitula and if the amount of B. thuringiensis deposited on the capitula would be sufficient to control the banded sunflower moth, Cochylis hospes Walsingham. The study demonstrated that honey bees became contaminated with B. thuringiensis as they exited hives equipped with filled pathogen applicators and deposited enough B. thuringiensis on the capitula to cause banded sunflower moth larval mortality. This study was conducted to determine dose-mortality response of the banded sunflower moth, Cochylis hospes Walsingham, to Bacillus thuringiensis Berliner and its effect on sunflower seed damage and yield. Of B. thuringiensis products tested, Cutlass AF and Javelin WG had the lowest LC50 values and highest slopes and relative potencies for C. hospes. Javelin WG was superior to the other B. thuringiensis materials tested and to Asana XL in preventing seed damage. Sunflower heads sprayed with Javelin WG had higher yields than sunflower heads sprayed with Asana XL or the other B. thuringiensis products tested. Asana XL is a standard …

Transgenic Bt crops expressing Cry and Vip toxins from Bacillus thuringiensis (Bt) have been increasingly planted to manage insect pest damage on agricultural crops. The high adoption of Bt-based insecticidal technologies suggests an increase selection pressure for the evolution of resistance in insect populations. Transgenic corn expressing Cry1F protein from Bacillus thuringiensis Berliner has been registered for Spodoptera frugiperda control since 2003. Unexpected damage to Cry1F corn was reported in 2006 in Puerto Rico and Cry1F resistance in S. frugiperda was documented. The inheritance of Cry1F resistance was characterized in a S. frugiperda resistant strain from Puerto Rico which displayed >387-fold resistance to Cry1F. Concentration-response bioassays of reciprocal crosses of resistant and susceptible parental populations indicated that resistance is recessive and autosomal. Bioassays of the backcross of the F1 generation crossed with the resistant parental strain suggest that a single locus is responsible …

The European corn borer, Ostrinia nubilalis Hübner (Lepidoptera: Crambidae) has been a major pest of corn and other crops in North America since its accidental introduction nearly a hundred years ago. Wide adoption of transgenic corn hybrids that express toxins from Bacillus thuringiensis, referred to as Bt corn, has suppressed corn borer populations and reduced the pest status of this insect in parts of the Corn Belt. Mortality of western corn rootworm (Diabrotica virgifera virgifera LeConte) due to feeding on MON863 transgenic maize (Zea mays L.) expressing the Cry3Bb1 protein was evaluated at three Missouri sites in both 2003 and 2004 and at one site each in South Dakota, Nebraska, and Iowa in 2004. To do this, survivorship relative to survivorship on isoline maize (i.e., the same genetic background, but without Cry3Bb1) was evaluated. Transgenic corn (Zea mays L.) products expressing multiple Bt proteins targeting a same group of insect pests have become commercially available in the U.S. The first commercialized pyramided Bt corn technologies for managing lepidopteran pests include Genuity® VT Triple Pro™ and Genuity® SmartStax™. Sugarcane borer, Diatraea saccharalis (F.), is a major target of Bt maize in South America and many areas of the US mid-south region. Six laboratory strains of D. saccharalis were established from six single-pair F2 families possessing major resistance alleles to Cry1Ab maize hybrids. Susceptibility of the six strains was evaluated on diet treated with each of four purified trypsin-activated Bt proteins, Cry1Ab, Cry1Aa, Cry1Ac, and Cry1F. Bt susceptibility of the six strains was compared with that of known Cry1Ab-susceptible and -resistant strains of D. saccharalis. Transgenic corn hybrids that express toxins from Bacillus thuringiensis (Bt) are highly effective against the European corn borer, Ostrinia nubilalis (Hübner), and the closely related Asian corn borer, Ostrinia furnacalis (Guenée). Since the registration of Bt corn hybrids in the U.S. in 1996, there has been a great deal of information generated on O. nubilalis. However, relatively little information exists for O. furnacalis. The sugarcane borer, Diatraea saccharalis, is a major target pest of transgenic corn expressing Bacillus thuringiensis (Bt) proteins in many areas of the U.S. mid-southern region. A Cry1Ab-resistant (Cry1Ab-RR) strain of D. saccharalis has been developed from a single two-parent family-line. To examine the molecular mechanisms of the Cry1Ab resistance in this insect strain, cDNAs of five types of potential candidate genes related to Bt resistance were sequenced using reverse transcriptase polymerase chain reaction (RT-PCR) and 5’ rapid amplification of cDNA end (5’ RACE). Dietary diversification, including consumption of plant tissues such as pollen, can enhance the fecundity of generalist predators, resulting in improved control of pest prey. Supplemental pollen feeding has been observed in many natural enemies, including sheet-web spiders (Araneae: Linyphiidae), which represent a major component of food webs in agroecosystems. Their horizontal, ground-based webs have the potential to intercept pollen grains during anthesis of crop plants, providing the opportunity for consumption of pollen to occur. In laboratory feeding trials, Frontinella communis and Tennesseellum formicum (Araneae: Linyphiidae) readily fed on pollen grains dusted on their webs, with 82 and 92% of spiders …

Despite the reported specificity of Bacillus thuringiensis proteins against target pests, a number of studies have indicated that the uptake of Bt-endotoxins from bioengineered crops could have negative effects on natural enemies. It is therefore essential to quantify exposure pathways in non-target arthropod food webs across multiple transgenic events. Adult ground beetles (Coleoptera: Carabidae) were collected from transgenic corn fields expressing lepidopteran-specific Cry1Ab, coleopteran-specific Cry3Bb1, and both Cry1Ab and Cry3Bb1 (stacked event), as well as a non-transgenic isoline. To examine how resistance to Bacillus thuringiensis (Bt) toxins influences movement and survival of European corn borer (Ostrinia nubilalis [Hübner]) neonates, the responses of Cry1Ab-resistant , -susceptible, and hybrid (F1) larvae were examined using two different techniques. First, using an automated videotracking system, aspects of O. nubilalis movement were quantified in the presence of artificial diet incorporating 50% non-Bt or insect-resistant Cry1Ab maize tissue. Second, O. nubilalis dispersal and survival were measured 48–72 h after hatching on a Cry1Ab maize plant surrounded by two non-Bt maize plants. The sugarcane borer, Diatraea saccharalis (F.), is a dominant corn stack boring pest and a major target of Bacillus thuringiensis (Bt)-corn in many areas of the mid-southern region of the United States. A Cry1Ab-resistant strain of D. saccharalis, capable of survival on commercial Bt corn plants, was established using an F2 screening procedure. Larval survival of Cry1Ab-resistant, -susceptible, and -heterozygous genotypes was evaluated on five non-Bt and seven Bt field corn hybrids at two plant stages. During the vegetative stages, all seven Bt corn hybrids were highly efficacious against the three genotypes, while 8-18% of the heterozygous genotype survived on …

Resistance to transgenic cotton, Gossypium hirsutum L., producing Bacillus thuringiensis (Bt) toxin Cry1Ac is linked with three recessive alleles of a cadherin gene in laboratory-selected strains of pink bollworm, Pectinophora gossypiella (Saunders), a major cotton pest. Here, we analyzed a strain (MOV97-R) with a high frequency of cadherin resistance alleles, a high frequency of resistance to 10 μg of Cry1Ac per milliliter of diet, and an intermediate frequency of resistance to 1,000 μg of Cry1Ac per ml of diet. Transgenic corn expressing the Cry1Ab toxin from Bacillus thuringiensis is highly toxic to European corn borer, Ostrinia nubilalis, larvae. A putative Cry1Ab receptor (OnBt-R1) molecule was cloned and sequenced from a cDNA library prepared from midgut tissue of O. nubilalis larvae. The 5.6 Kb gene is homologous with a number of cadherin genes identified as Cry1 binding proteins in other lepidopterans. Brush border membrane vesicles were prepared using dissected midguts from late instars. Susceptibility to the Cry1Ab protoxin and toxin from Bacillus thuringiensis (Berliner) and activity of gut proteinases were assessed in both susceptible and Cry1Ab-selected colonies of European corn borer, Ostrinia nubilalis (Hübner). Resistance in two different selected colonies was at least 6- and 15-fold for the Cry1Ab protoxin and 108- and 484-fold for the Cry1Ab toxin. Activities of trypsin-like, chymotrypsin-like and elastase-like proteinases were variable among the colonies tested and not indicative of a major contribution to Cry1Ab resistance. Activation of the 130-kDa Cry1Ab protoxin occurred rapidly in all colonies, with no apparent differences among colonies. In addition, there were no …
