Fatty Acid Desaturase 7 (FAD7) is a chloroplast-localized enzyme that alters the fatty acid content of photosynthetic membranes, and that negatively regulates plant defenses against aphids. Previous studies in the model organism Arabidopsis (Arabidopsis thaliana) have shown that loss-of-function mutations in FAD7 decrease population growth of the green peach aphid (GPA; Myzus persicae Sulzer). This study further characterized the effects of a fad7 null mutant on aphids, and investigated the role of reactive oxygen species (ROS), including singlet oxygen (1O2), in plant responses to aphid resistance in fad7 as well as in wild type plants and a mutant with heightened …

Tomato (Solanum lycopersicum) is one of the most economically important vegetable crops grown around globe but is a host for numerous pests and pathogens. In the future, tomato breeders will have to focus on increasing fruit quantity and on enhancing pest resistance. Many accessions of the wild relative of tomato, S. habrochaites display high levels of resistance towards arthropods such as spider mites. The presence of the sesquiterpene hydrocarbon, 7-epi-zingiberene, found in S. habrochaites type IV trichomes has been associated with arthropod resistance. The sugarcane aphid (Melanaphis sacchari) has emerged as a significant pest for sorghum. The use of sugarcane aphid-resistant sorghum germplasm with integrated pest management strategies appears to be an excellent solution to this problem. In this study, a resistant line (RTx2783) and a susceptible line (A/BCK60) were used to characterize the differences in plant responses to the sugarcane aphid through a series of experiments, which examined global sorghum gene expression, aphid feeding behavior and inheritance of aphid resistance. Fatty Acid Desaturase7 (FAD7) is a chloroplast-localized enzyme that converts 16 and 18 carbon dienoic fatty acids to trienoic fatty acids. The suppressor of prosystemin-mediated response2 (spr2) mutant in tomato (Solanum lycopersicum) and the fad7-1 mutant in Arabidopsis (Arabidopsis thaliana) result in the loss of function of FAD7, which alter the fatty acid profiles of chloroplast membranes and enhance resistance against aphids. This research contributes toward the long-term goal of identifying factors that determine aphid resistance in FAD7 mutants. Rice (Oryza sativa L.) is considered one of the most important crops in the world, and the sugarcane borer, Diatraea saccharalis Fabricius (Lepidoptera: Crambidae), is among the key pests damaging the crop in the Americas. The objective of this work was to identify rice genotypes as a source of resistance to D. saccharalis. Rice plants were infested in the greenhouse and subsequently evaluated for damage, larval weight and survival, and stem size. Maize was domesticated from teosinte in Mexico some 7,000 to 10,000 years ago and quickly spread through the Americas. It has become one of the most important crops at a local and global level. Two types, Northern Flint corn and Southern Dent corns provided the basis of the genetic background of modern maize hybrids. The development of hybrids, first double-cross and later single-cross hybrids, along with a transition to high input farming provided huge yield increases, which have continued to improve with improving technology.

Fatty acid desaturases (FADs) are enzymes that act in the chloroplast or the endoplasmic reticulum (ER) to incorporate double bonds into the acyl chains of fatty acids, and recent evidence indicates that at least one of these enzymes, FAD7, also influences plant resistance to aphids. FAD7 is an enzyme in the chloroplast that is found throughout the plant kingdom and that desaturates 16- and 18-carbon fatty acids (FAs) with two double bonds (dienoic acids) to generate FAs with three double bonds (trienoic acids). Understanding the behavior of pests targeted with Bacillus thuringiensis Berliner (Bt) crops is important to define resistance management strategies. Particularly the study of larval movement between plants is important to determine the feasibility of refuge configurations. Exposure to Bt maize, Zea mays L. (Poaceae), has been suggested to increase larval movement in lepidopteran species but few studies have examined the potential for resistance to interact with behavioral responses to Bt toxins. The American chestnut, Castanea dentata, was once one of the most useful and abundant canopy trees in eastern North American forests. Over the last 200 years, the species has been decimated by two exotic pathogens, Phytophthora cinnamomi and Cryphonectria parasitica, killing millions of trees and reducing surviving Castanea dentata to short-lived sprouts. Cryphonectria parasitica-resistance breeding programs are currently producing advanced backcross generations, which are being compared with pure American chestnut in field tests of growth performance and Cryphonectria parasitica resistance. Fourteen U.S plant introduction (PI) accessions of C. lanatus var. lanatus (4), C. lanatus var. citroides (5) C. colocynthis (5) and a known susceptible commercial cultivar ‘Sugar Baby’ were evaluated for resistance to the twospotted spider mite, Tetranychus urticae Koch, in a limited free-choice and free-choice bioassay under laboratory conditions. The limited free choice bioassay, involved nine Petri dish cages that held five randomly assigned leaves individually inoculated with two adult females and one adult male. A recent study in Solanum bulbocastanum (a wild relative of the cultivated potato) aiming to identify potential genes involved in aphid and pathogen resistance mechanisms found that a homolog of the Arabidopsis thaliana cysteine protease gene RD19a is upregulated during aphid infestation. RD19a is upregulated in response to abiotic stresses such as drought and high salinity, and rd19a mutants show increased susceptibility to bacterial infection. Transgenic corn (Zea mays L.) products expressing multiple Bt proteins targeting a same group of insect pests have become commercially available in the U.S. The first commercialized pyramided Bt corn technologies for managing lepidopteran pests include Genuity® VT Triple Pro™ and Genuity® SmartStax™. Part 1: Visitation of potential pollinators to Cornus florida and C. kousa flowers was assessed in East Tennessee in 2008 and 2009. Data regarding insect visitation rates to multiple trees per location were gathered throughout the flowering period. Diurnal and seasonal variations in visitation were recorded. Pollen coverage was assessed on portions of captured insect exoskeletons that were most likely to contact the stigma. Eleven families in four insect orders were collected from C. florida and 26 families in five orders from C. kousa. The most important pollinators in eastern Tennessee were bees in the Andrenidae and Halictidae. The most …
