Within the last decade, the number of diseases associated with dysregulation of the neural circuits responsible for energy balance, glucosensing, and feeding control has increased. One brain region containing these circuits is the lateral hypothalamic area (LHA), which plays an important role in the sleep/wake cycle, energy balance, and motivated behavior. Although the LHA has been termed a â??feeding centerâ??, other regions within the hindbrain are known for the regulation of similar functions including energy balance, glucosensing, and feeding control. Breast cancer is the second most diagnosed cancer among women and is about 30% of all new cases of female cancers each year. It is projected that 1 in 8 every U.S. woman (about 13%) develop invasive breast cancer over the course of her lifetime. Anticancer drug discovery is a time and resource-consuming process for which exceedingly reliable and efficient modern approaches are needed. Phenotypic drug screenings can generate highly potent and innovative drug candidates; however, deconvolution of the drugâ??s target often presents significant barriers to drug development. To overcome this hurdle, we have originally combined in vitro and in silico analyses to uncover the molecular mechanism(s) driving the anticancer activity of the uniquely structured small molecule drug candidate, Tpz-1. Neurodegeneration, a progressive loss of nerve cells, occurs in many neurological disorders, including Alzheimer's disease (AD) and Parkinson's diseases (PD), as well as in dementia. 4-nonylphenol (4-NP), an endocrine-disrupting compound (EDC), has been shown to affect brain development and may cause neurodegeneration. In the environment, 4-NP arises as a degradation product of alkylphenol polyethoxylates, compounds widely used as nonionic surfactants in commercial production, as well as in herbicides, pesticides, polystyrene plastics, and paints and has been shown to undergo a high level of accumulation in biological tissues. The 26S proteasome has been successfully targeted to treat multiple myeloma. However, the effect can sometimes be reversed as cells acquire resistance to chemotherapy. Recently, combining proteasome inhibitors with indirect proteasome inhibitors has been proposed in order to delay and even reverse resistance. Small molecules derived from 4-piperidinone that maintain an unsaturated β-carbonyl group are thought to interact with the thiol group of catalytically active cysteine residues in deubiquitinases that associate with the 26S proteasome, effectively inhibiting the proteasome.

Tyrosine kinases are an essential component of cell signal transduction pathways, many of which promote cellular proliferation. However, when a tyrosine kinase is aberrantly activated or its negative regulation is lost, the result can be malignancy. In humans, 90 tyrosine kinases are present and of these, 51 have been linked to a malignancy through mutation or overexpression. Janus kinase 3 (JAK3) is one such kinase that upon hyperactivation, due to a somatic mutation, has been linked to cancer including its substrate, signal transducer and activator of transcription (STAT5). The Effect Of Exogenous Leptin On Murine Dendritic Cell Morphology And Function, Christine Delgado

Leptin is a pleiotropic hormone primarily secreted by white adipose tissue. Although originally found to regulate food intake and energy expenditure, it is now known to also significantly affect the immune system. In this study we assessed the effect of leptin on bone marrow derived dendritic cell (BM-DC) cytoskeletal structure and functionality. Our preliminary data demonstrates that the addition of physiological concentrations of exogenous leptin to BM-DC causes cytoskeletal rearrangement specifically that of actin as evidenced by increased number of llamelopodia/dendrites and increased staining for actin within the llamelopodia/dendrite region.
