The mammalian neocortex develops from a thin layer of neuroepithelial cells into a robust mosaic of differentiated neurons organized into distinct layers and functional areas. This process requires a complex choreography of transcription factors and epigenetic regulators to be spatially and temporally activated in a precise manner. Disruptions in any aspect of this delicate process may lead to neurodevelopmental disorders like autism spectrum disorder, schizophrenia, or intellectual disability. One such transcriptional coregulator that is a crucial regulator of neocortical development is Cited2. The majority of neurons within the spinal cord are interneurons. Interneurons make important connections with other cells that allow signals to be relayed between the brain and the rest of the body, and aid in functions such as locomotion. One key functional characteristic of interneurons is the neurotransmitter that they use to communicate with other cells. Interneurons that use GABA or glycine neurotransmitters to communicate have an inhibitory phenotype, while interneurons that use glutamate neurotransmitters have an excitatory neurotransmitter phenotype. Astrocytes play a vital role in orchestrating the precise brain wiring that occurs during development and are essential for maintaining homeostasis into adulthood. The cystine/glutamate antiporter, system xc-, in the central nervous system is especially abundant in astrocytes and itself is known to contribute importantly to the basal extracellular glutamate concentration as well as the intracellular and extracellular glutathione levels, either of which, if perturbed, could alter brain development and/or contribute to degeneration. Bacterial cells often form sessile biofilms that are up to 1,000 times more resistant to antimicrobial agents than free-living cells. Meanwhile, bacteria produce a small subpopulation of slow-growing or non-growing persister cells that exhibit high tolerance to antibiotics. Both biofilms and persister cells play important roles in the recalcitrance of chronic infections. Recent studies have shown that bacterial cell-to-cell communication, named quorum sensing (QS), is involved in the biofilm and persister formation.

In this study, we investigated the effects of quorum sensing signals N-(3-oxododecanoyl)-homoserine lactone (3-oxo-C12-HSL) and N-butyryl homoserine lactone (C4-HSL) on isolated Pseudomonas aeruginosa PAO1 and PDO300 persister cells. …

Aberrant hyperactivation of the phosphatidylinositol 3-kinase (PI3K) cell-signaling pathway, one of the most prominent cell growth, proliferation and survival pathways in the human body, has been observed in many forms of cancer. Upon deregulation, this pathway can facilitate the evasion of programmed cell death, the stimulation of autonomous growth, and the evasion of regular growth-inhibitory signals. My project focuses on the V2 cells of the zebrafish spinal cord. The V2 cells are an unusual class of spinal cells because they all originate from molecularly indistinguishable p2 progenitor cells in the spinal cord. However, as these cells become post-mitotic and differentiate, they start to express different transcription factor genes that allow them to initially develop into two sets of molecularly distinct cells. As differentiation continues, at least one more class of molecularly distinct cells develops. Current treatment for cancer employs a combination of modalities including surgery, radiation, chemotherapeutic drugs, and immunotherapy. In female mammals, proper oocyte development is a vital prerequisite for future gamete viability and fertility. This development of oocytes, known as oogenesis, begins with the migration of primordial germ cells to the genital ridge of the early embryo, where multiple rounds of mitotic division occur without complete cytokinesis. The result is temporary cyst morphology. Cyst breakdown is a crucial process in the next developmental stage, resulting in formation of the single oocytes which will grow in follicles surrounded by granulosa cells and eventually develop into eggs. In the United States alone, more than one million people are blind. One of the leading causes of blindness is the retinal disease, Retinitis Pigmentosa (RP). RP is a heterogeneous group of inherited disorders characterized by the initial loss of rod photoreceptors. Approximately 100,000 people in the United States and 1.5 million people worldwide are affected by RP. Unfortunately, there is no cure. In humans with RP, it is known that the loss of rod photoreceptors leads to degeneration of cones.
