A Gene Expression Profile Associated With Successful Cns Axon Regeneration In Xenopus Laevis, Jamie L. Belrose

Background: The South African claw-toed frog, Xenopus laevis, is uniquely suited for studying differences between regenerative and non-regenerative responses to CNS injury within the same organism, because some CNS neurons (e.g., retinal ganglion cells after optic nerve crush (ONC)) regenerate axons throughout life, whereas others (e.g., hindbrain neurons after spinal cord injury (SCI)) lose this capacity as tadpoles metamorphose into frogs. Tissues from these CNS regions (frog ONC eye, tadpole SCI hindbrain, frog SCI hindbrain) were used in a three-way RNA-seq study of axotomized CNS axons to identify potential core gene expression programs for successful CNS axon regeneration.

Relative to most primate species, Japanese macaques (Macaca fuscata) experience extreme seasonal variation in thermal and ecological stressors that can impact energetic demands. To cope with these environmental changes, levels of metabolic hormones, such as cortisol and triiodothyronine, fluctuate to facilitate energetic adjustments. While previous research in primates has investigated thermal and ecological stressors individually, a combined assessment of these stressors alongside hormone levels can provide a more holistic understanding of the relationship between a primate’s thermoregulation, energetic balance, and stress. Dryland ecosystems need effective restoration strategies to address severe degradation. It is often assumed that voluntary forb weeds either compete with or have no effect on native seedlings. In contrast, theory and empirical work suggest a potential facilitative role for such forb weeds. We conducted a manipulative weed exclusion experiment at a semi-arid site in South Texas targeted for large scale forest restoration and subsequently dominated by early successional forb weeds to discern the net effect of these weeds on the growth and survival of target thornscrub tree and shrub seedlings. The Asian citrus psyllid (ACP), Diaphorina citri Kuwayama (Hemiptera: Liviidae), vectors “Candidatus Liberibacter spp.”, the causative agent of Citrus Greening Disease. Managing ACP is imperative given the continuous increase in HLB positive trees. An aspect of integrated pest management (IPM) is the use of entomopathogenic fungi for the biological control of D. citri.

In order to identify endemic entomopathogenic fungi actively infecting D. citri, ACP were collected from local residential areas, surface sterilized, and plated on a semi-selective agar medium. The role of human disturbance in accelerating weed growth is well understood. While most of these studies have focused on soil mediated disturbance, mowing is also a management practice that could impact weed traits. Using silver leaf nightshade (Solanum elaeagnifolium), a noxious and invasive weed, we asked whether continuous mowing affects growth and plant defense traits. We found that mowed plants produced significantly less fruits, had lower total seed fitness, but had higher seed mass, and germinated significantly faster. Emerging diseases such as Snake Fungal Disease (SFD) caused by the fungus Ophidiomyces ophiodiicola (Oo) have caused population declines in various snake species in the United States which play a crucial role in the ecosystem as a natural pest control. Fish and macroinvertebrate response to restoration and conservation efforts varies in regards to the size and structure of the system (e.g. headwater streams in WV versus large rivers such as the Mississippi River). This project reviews fish and macroinvertebrate rebound in treated acid mine drainage (AMD) streams in WV as well as macroinvertebrate drift patterns in the Mississippi and Missouri Rivers. White-nose syndrome (WNS) is currently the leading cause of death to North American bats and was first documented in the United States in 2006 on the East Coast. Infection with the fungus Pseudogymnoascus destructans (Pd) causes bats to arise from hibernation with unnecessary frequency, leading to depletion of fat reserves that are imperative for extended periods of torpor. During metamorphosis, the steroid hormone 20-hydroxecydysone (20E) triggers a plethora of tissue-specific responses in the common fruit fly, Drosophila melanogaster. The salivary glands are exocrine organs that respond to a large pulse of 20E to secrete a massive cargo of stored glycoproteins called glue. Because the glands are amenable to ex vivo-organ culture and powerful genetic manipulations, they are an excellent model for studying the conserved developmental and physiological signaling pathways that regulate the general function of these specialized secretory tissues. A mammalian species inventory with comparisons between sampled sites was conducted via multiple methodologies to document presence of mammals at two Missouri state parks within the Ozark National Scenic Riverways. Camera traps, small mammal traps, acoustic detectors, and mist nets were used to detect species at the parks, and species similarity indices and occupancy analyses were used to discern use of space. A mammalian inventory was compiled for each area of inquiry. Greater diversity was found at the park with more variable habitat types. Symbiotic relationships are extremely common in virtually all species. These relationships between organisms, particularly mutualistic symbioses, can have an incredibly large impact and can greatly affect the evolution of the species involved. Of particular interest is the suspected mutualistic relationship between green algae and fungi that may hold key evidence of how terrestrial plants evolved. Neurofibromatosis Type 2 (NF2) is a rare genetic disorder, classified as a neurocutaneous syndrome inherited through an autosomal dominant genetic mutation or deletion of the NF2 gene in humans. The tumor suppressor gene NF2 encodes the cytoskeletal protein MERLIN to promote apoptosis and inhibit abnormal cellular proliferation. Mutations in NF2 lead to a deficiency of functional MERLIN in humans and can result in hearing loss, disability, and premature death. This dissertation explores how students reason about genetic engineering socioscientific issues (SSIs) related to a recently developed, powerful genome editing technology called CRISPR/Cas9. It is divided into three chapters. Chapter One describes an exploratory study that characterized students’ moral reasoning using a sociocultural theoretical framework. I used content analysis and logistic regression to investigate how academic and social factors influenced moral reasoning. Students generally opposed the use of CRISPR/Cas9 technology for non-medical enhancements, and the moral considerations used were influenced by genetics knowledge level and demographic variables such as gender and socio-economic status. The Critically Endangered Largetooth Sawfish, Pristis pristis, experienced global declines in range and abundance over the past century and Australia is now their last stronghold. This research aimed to understand whether these declines have been accompanied by a reduction in levels of genetic diversity. Using P. pristis tissue samples sourced from natural history specimens, three fragments (616-bp, 386-bp, 141-bp) in the mitochondrial DNA (mtDNA) control region were PCR-amplified. At each fragment, significant genetic structure was found in P. pristis overall (ΦST=0.946, N=9; 0.813, N=54; 0.771, N=99). The Ediacaran to Cambrian Deep Spring Formation consists of mixed carbonate-siliciclastic strata which contain an increasingly complex and biogeographically important biota. Past investigations of the Deep Spring Formation at Mt. Dunfee, Nevada, explored the highly diverse microbialite reefs consisting of a wide range of stromatolite morphologies which exerted significant control on local sedimentation and topography. Early investigations also documented the biomineralizing metazoan Cloudina (an Ediacaran index fossil). However, recent exploration of the area has resulted in the discovery of several new metazoan fossil communities consisting of a diverse assemblage of Ediacaran soft-tissue tubicolous vermiforms (tube fossils) similar to Cloudina. Aquatic ecosystems are inundated by environmental pesticides. This study, observed the dose-dependent effects of pesticides cocktail (metalachlor, linuron, isoproturon, tebucanazole, aclonifen, atrazine, pendimethalin, and azinphos-methyl) and elevated temperature (32 °C for 4-week exposure) on morphology of gills and kidneys, and expression of nitrotyrosine protein (NTP), dinitrophenyl protein (DNP), catalase (CAT), superoxidase dismutase (SOD), Na+/K+-ATPase, renin, and apoptosis in tissues of goldfish. Additionally, the effects on the free-swimming behavior were also observed. Histological analysis showed wide-spread damage in tissues at higher temperature and pesticides co-exposure. PI3K/AKT/mTOR signaling pathway is activated in 90% of all Glioblastoma Multiforme (GBM) tumors. We used a dual targeting approach to interfere with the phosphoinositide 3-kinase- mammalian target of rapamycin (PI3K-mTOR) pathway by treating U87MG GBM cells with NVP-BEZ235 (PI3K and mTOR a dual inhibitor) and, their downstream effects on molecular regulation on glycolytic genes were investigated using RNA-sequencing and gene set enrichment analysis (GSEA). RNA-seq identified 7803 differentially regulated genes in response to NVP-BEZ235. GSEA identified two glycolysis-related gene sets had significant differences (P < 0.05) in gene expression between control and NVP-BEZ235 treated samples. The GSEA core enrichment reported 17 and 35 genes were significantly downregulated (P adj < 0.05) in response to NVP-BEZ235 treatment in two different glycolysis related gene sets. We validated the expression of genes using qRT-PCR in U87MG cells as well as other GBM cell lines. Our results suggest novel insights on hallmark metabolic reprogramming associated with the PI3K-mTOR inhibition and provides advances in developing molecular targeted therapies.

Thermal-stress events associated with climate change cause coral bleaching and mortality that threatens coral reefs globally. Yet coral bleaching and coral cover patterns vary spatially and temporally, and few studies have identified bright spots where corals have the potential to survive through climate change. Artemisia californica, Mimulus aurantiacus, Equisetum telmateia, Equisetum hyemale, and Marah fabacea were selected from a list of plants reported as having been used for ailments related to infections by tribes located in California. Freshwater systems experience multi-faceted degradation from a variety of ecological and environmental stressors. Three common stressors in these systems, drought, nutrient pollution, and invasive species, have wide-ranging effects on stream population- community- and ecosystem dynamics. Biogeography of the Himalayan region [to include the Qinghai-Tibetan Plateau (QTP)] evolved over a ~30M year span, catalyzed by the collision of the Indian and Eurasian plates. The resulting uplift produced major ecological and climatic effects, that in turn drove the diversification of biodiversity. As a result, the QTP is designated as a global biodiversity hotspot particularly vulnerable to cumulative climatic effects, including shrinking distributions, declining numbers, and local extinctions. Understanding how the biodiversity within the Himalaya/ QTP was established and maintained is a necessary first step in prioritizing conservation efforts.

This study investigates the relationship between multiple temperature variables, to include annual and pre-lay date temperatures with first-egg and mean first-egg lay dates of the eastern bluebird at the Warner Parks in Nashville, Tennessee, USA. Data is collected by citizen scientists for the Eastern Bluebird Nesting Box Project while visiting artificial nest boxes throughout the park and recording observations made during the breeding season. Temperature data is retrieved from the Northwest Alliance for Computational Science and Engineering’s Parameter-elevation Regressions on Independent Slopes Model (PRISM) Climate Group, based at Oregon State University. Understanding threats and drivers of extinction risk is critical to conserving marine biodiversity. Although marine bony fishes are some of the most diverse and abundant vertebrates in the world, overexploitation is the major threat to these taxa. Species in the family Sciaenidae are important marine resources in many parts of the world, supporting small- and large-scale fisheries for consumption and the international maw (dried swimbladder) trade. Sciaenids exhibit a diverse array of life history characteristics, such as high fecundity, large body size, formation of spawning aggregations, and longevity. Autumn migration is a time when billions of birds move from breeding grounds in North America to wintering grounds in Central and South America, with many individuals relying on stopover habitats en route for resting and refueling purposes.
