In the course of experiments designed to identify and characterize structural proteins of the nuclear matrix, one antibody was generated which recognized an extraction-resistant cytoplasmic protein. This antibody was used as the starting point in the cloning and molecular characterization of a novel protein of the inter-membrane space of the mitochondrion which has been named mitofilin. Mitofilin is expressed in all human cell types, and murine homologues also exist. Mitofilin associates only with mitochondria and not with other membrane-bounded organelles such as Golgi or endoplasmic reticulum. Human zona pellucida protein 3 (hZP3) is the putative receptor on the zona pellucida of the mature oocyte that recognizes and binds sperm, and therefore plays a critical role in fertilization.

A full length cDNA of hZP3 (1278 bp) was amplified from the human ovary mRNA by reverse transcription-polymerase chain reaction (RT-PCR). The hZP3 cDNA was subcloned into PSK and pREP4 expression vectors. Information about these enzymes in eukaryotic systems is mostly limited to nuclear enzymes, although our laboratory has been characterizing the biochemical and biophysical properties of the mammalian mitochondrial topoisomerases. Alzheimer's Disease (AD) is a complex neurodegenerative disorder that affects a significant portion of the human population regardless of ethnicity or gender. A mitochondrial hypothesis of AD has been proposed based on a number of studies which establish altered oxidative phosphorylation (OXPHOS) and ATP synthesis in AD tissue. Moraxella (Branhamella) catarrhalis is the third-most-frequently isolated microorganism associated with acute exacerbations of chronic bronchitis in patients during their stay at the Mountain Home VA Medical Center (MHVAMC). In order to develop a practical, epidemiologically-meaningful typing method for M. (B.) catarrhalis, we tested two methods based on analysis of chromosomal DNA for typeability, reproducibility, and ability to differentiate between unrelated strains (discriminatory power, D). M. The purpose of this study was to identified nonessential murine cytomegalovirus (MCMV) genes involved in pathogenesis in vivo. Our approach to identifyjng these genes consisted of constructing MCMV mutants, and then analyzing these mutants in vitro and in vivo. Recombinant viruses (RV) expressing the β-glucuronidase marker gene were constructed by site-directed insertion and deletion mutagenesis of the MCMV Hind III-J and -I regions of the viral genome. The v-jun oncogene was initially identified as the causative agent for fibrosarcomas in chickens. Studies show that overexpression of v-Jun proteins transforms chicken embryo fibroblasts (CEF) in vitro, and forms tumors in chickens in vivo. The mechanisms for this are not clearly defined. Conceivably, overexpression of an unregulated transcription factor would cause cell transfonnation by illicit regulation of its target genes. Seedlings of chilling-sensitive maize inbred G50 (Pioneer) survived poorly when exposed to a chilling stress. Acclimation or ABA treatment caused a dramatic improvement of chilling tolerance. Differences in gene expression between acclimated and unacclimated seedlings were investigated and three cDNA clones (designated car333, car30, and car757) were isolated, representing transcripts that were up-regulated during acclimation. Car30 and car757 were not homologous to any known sequence but car333 was 97.8% homologous to cat3, maize mitochondrial catalase. Full-length clones that encode tomato (Lycopersicon esculentum Mill.) glutamate-1-semialdehyde-2,1-aminomutase (GSAAM) and 5-aminolevulinic acid dehydratase (ALAD) were isolated and characterized. Sequence analysis showed that tomato GSAAM and ALAD clones exhibited a high level of homology to corresponding clones found in other plant species. The primary structure of GSAAM predicted a 481-amino acid precursor that comprised a 46.7 kDa, 437-amino acid mature protein and a transit peptide of 44 amino acids. In this dissertation, on the basis of the similarities in sequence and structure between GTP-binding proteins and AMPSase, it was of interest to determine whether or not the GTP-binding consensus sequences have similar functions. It will be shown that the GTP-binding consensus sequences found in AMPSase play similar roles to those of the GTPase superfamily. Saccharomyces cerevisiae is a pleimorphic ascomycete, capable of growing in the yeast form or the pseudohyphal form. The pseudohyphal form is characterized by elongated cells that possess a unipolar budding pattern and grow in chains away from the center of the colony. This change in morphology is induced by nitrogen starvation in the presence of carbon. Mycobacterium paratuberculosis transcriptional and translational signals were studied to gain insight into gene expression in this organism. To study M. paratuberculosis promoter structures, a more versatile promoter selection vector was constructed from the pKO1 parent vector. This new plasmid, pJJ2, was used to identify 11 promoter fragments from an M. paratuberculosis DNA library. A novel insulin-like immunoreactivity, adrenergic receptors (ARs) and voltage-dependent Ca[superscript]2+ channels (VDCCs) were characterized in a clonal [beta]-cell line, RINm5F, in a passage- and morphology-dependent manner. The cells exhibited two morphological features; the differentiated cells (DCs) with long neuron-like processes and the undifferentiated cells (UCs) without the processes;As morphology of the cells changed from DCs to UCs, amount of secreted insulin gradually decreased; however, insulin reacted to antiserum vs. bovine insulin (ABI) gradually increased from barely detectable amounts. In this study, DNA-protein interactions of the first kilobase region proximal to the transcription start site of the zrp2 (Zea mays root-preferential 2) gene promoter was analyzed. The intent of this research was to identify regions of the promoter and DNA-protein interactions, that are responsible for root-preferential transcription. In vitro run-on transcription assays had revealed dramatically higher levels of transcription in 3-d-old roots than in 5-d-old leaves (Held, 1993). The entire soybean Sle gene family was cloned and characterized. The five Sle genes (Sle1-5) could be divided into two classes based on sequence identity. Sle1-4 map to 4 different linkage groups. Sle1 and Sle5 are expressed weeks earlier in seed development than Sle2 and Sle3. Sle4 is a pseudogene. Maximal levels of Sle mRNA accumulate in the seeds, before desiccation had occurred, and decline rapidly upon seed imbibition. Desiccation does not induce Sle expression in seeds. By cloning and sequencing the mtDNA of cows from different maternal lineages, it was found that multiple heteroplasmic mtDNA states within the D-loop and rRNA genes of mtDNA existed extensively in Holstein cattle. Meanwhile, the nature and transmission of mtDNA were investigated. The mtDNA population in a specific animal was found to be a mixture of different genotypes. Quantitative trait loci (QTL) and QTL x environment interaction were identified for 11 agronomic traits in soybean, when measured at two locations in each of two years in Iowa. The segregating population consisted of 76 F[subscript]2:5/F[subscript]2:6 families derived from the cross between the two soybean (Glycine max (L.) The structural requirements for the function of the 3[superscript]' CCA end of E. coli valine transfer RNA in aminoacylation and protein synthesis have been investigated, by studying the activities of 3[superscript]' end tRNA[superscript] Val variants in these processes. These studies show that 3[superscript]' end tRNA[superscript] Val variants can be obtained which are functional in both aminoacylation and later steps of protein synthesis. Laser-induced fluorescence line narrowing (FLN) and non-line narrowing (NLN) spectroscopic methods were applied to conformational studies of stable DNA adducts of the 7[beta],8[alpha]-dihydoxy-9[alpha],10[alpha]-epoxy-7,8,9, 10-tetrahydrobenzo (a) pyrene (anti-BPDE). Stereochemically distinct (+)-trans-, (-)-trans-, (+)-cis- and (-)-cis adducts of anti-BPDE bound to exocyclic amino group of the central guanine in the oligonucleotide, 5'-d(CCATCGCTACC) · (GGTAGCGATGG), exist in a mixture of conformations in frozen aqueous buffer matrices. A novel cholesterol-reducing bacterium, Eubacterium coprostanoligenes ATCC 51222, was characterized and tested for its hypocholesterolemic effects in animal models. The bacterium did not require cholesterol for growth and synthesized cholesterol reductase constitutively. Lecithin was required for growth but excess lecithin in media did not improve bacterial growth and coprostanol production. Addition of pyruvate, lactose, and most reducing agents in media increased bacterial growth and coprostanol production. The bacterium was sensitive to most antibiotics but had strong resistance to tetracycline. A resting cell assay was established to evaluate the cholesterol reductase activity. Recombinant proteins of bovine rotavirus VP2, VP4, VP6 and VP7 were expressed in insect cells using baculovirus expression vectors. Expressed recombinant proteins were similar to those of rotavirus. Recombinant viral antigens expressed in insect cells reacted with polyclonal antiserum to the VMRI strain of BRV by an indirect immunofluorescence test. Expressed recombinant proteins were characterized by radioimmunoprecipitation and their molecular mass was found to be similar to that of in wild type rotavirus. These four recombinant proteins were antigenic in guinea pigs and induced antibodies to the corresponding rotavirus proteins as detected by indirect immunofluorescence test. Restriction fragment length polymorphisms (RFLPs) were employed to investigate genetic diversity and quantitative trait loci (QTL) for morphological characters on sorghum (Sorghum bicolor (L.) Moench). The objectives of this research were accomplished by studying two different sets of genotypes. One set was composed of 105 elite inbred lines (47 B- and 58 R-lines) and the other of 152 F[subscript]2:3 lines.
