In vitro propagation efforts play an essential role in conserving and restoring threatened freshwater mussel populations by circumventing the need for a fish host. Across a broad range of taxa, transformation is induced with an artificial M199 medium and rabbit serum. However, such formulation may not be sufficient in culturing critical species with more specific physiological requirements. In this study, multiple serum mixtures were tested to improve in vitro transformation of two freshwater mussel species: yellow lampmussel (Lampsilis cariosa) and tidewater mucket (Atlantaconcha ochracea). North America is home to 302 unionoid species, with approximately 53 occurring in Texas, and are considered one of the most imperiled group of organisms in North America. Several hypotheses have been proposed to explain the spatial distribution of mussels within a stream reach, but only certain hydraulic characteristics appear to be correlated with their distribution. Emerging evidence indicates that freshwater mussels may use flow refugia to remain embedded during high flow events. As the use of hydraulic variables to characterize mussel habitat becomes more widespread, it may be useful to implement sampling that captures these measures. One option may …

Native freshwater mussels (Family: Unionidae) are among the most threatened groups of freshwater fauna on Earth. Approximately 35 species have gone extinct since the 1900s and 72% of remaining species are considered endangered, threatened, or species of special concern. Unionid research can begin by establishing species presence and distributions via surveys. Objectives for this study were to 1) implement the first comprehensive unionid survey for South Dakota to assess distribution, composition, and decline, 2) estimate assemblage density and determine local versus broad scale habitat drivers of assemblage variation, and 3) determine areas of unionid conservation priority in South Dakota. The Buffalo River in Tennessee once hosted a rich population of freshwater mussels. During the 1980s, monitoring efforts demonstrated evidence of drastic declines and extirpation of entire assemblages. Increases in municipal development in the headwater tributaries and agriculture in the main stem of the Buffalo River are suspected causes for mussel community declines throughout the river. In 2011, collection data documented evidence of recovery in the lower Buffalo River.
