In vitro propagation efforts play an essential role in conserving and restoring threatened freshwater mussel populations by circumventing the need for a fish host. Across a broad range of taxa, transformation is induced with an artificial M199 medium and rabbit serum. However, such formulation may not be sufficient in culturing critical species with more specific physiological requirements. In this study, multiple serum mixtures were tested to improve in vitro transformation of two freshwater mussel species: yellow lampmussel (Lampsilis cariosa) and tidewater mucket (Atlantaconcha ochracea). Fishes of the family Goodeidae are considered to be among the most imperiled taxa in the world. The goal of this project was to provide a foundation for development of sperm repositories for goodeids and to use them as a model to assist conservation programs for imperiled freshwater live-bearing fishes. Development of such repositories is challenging because of unusual reproductive characteristics of live-bearing fishes, such as sperm bundles, internal fertilization, and the bearing of live young. The North Central Regional Aquaculture Center has designated the yellow perch (Perca flavescens) as a high priority species for culture. The demand for this species is high and it is estimated that the market could readily consume 50 to 100 million pounds per year. Tank culturing of yellow perch has several advantages over pond culture and this method has been growing in popularity, but is currently held back by problems in larval development. Remote Site Incubators (RSIs) were developed for the incubation of salmonid eggs directly at a field site to enhance success and imprinting of young. These have been employed in the re-establishment of Arctic Grayling in Montana and are being laboratory tested for possible reintroduction of this species in the Big Manistee River, MI. Arctic Grayling, Rainbow and Brook Trout eggs, obtained from state hatcheries, were reared in a laboratory using flow through lake water to assess egg and fry survival using RSIs. This research attempts to connect patterns in growth and migration of an anadromous species. The goal of this research was to understand habitat movements and growth of juvenile alewives (Alosa pseudoharengus) in the Penobscot Estuary and Bay through the use of otolith microchemistry, otolith growth increments, and a laboratory stable isotope turnover study. Understanding the connection between growth and movement of juvenile alewives may lead to more accurate and sophisticated conservation and restoration methods for anadromous species.

The sea lamprey (Petromyzon marinus) has survived throughout evolution for hundreds of millions of years. It is considered an invasive species to the Great Lakes that has caused dramatic changes in the ecosystem for fish communities resulting in the collapse of a fishing industry that was previously valued at billions of dollars. Hypoxia inducible factors (HIFs) are transcription factors and the master regulators of oxygen-dependent gene expression in animals. The focus of this thesis is the distribution of HIF protein in tissues of the fish Fundulus heteroclitus and F. grandis, two widespread species that occur in naturally hypoxic waters. Polyclonal antibodies against HIF-1α, HIF-2α, and HIF-3α were tested on proteins made in vitro and on extracts made from several tissues of normoxic and hypoxic fish. Little is known about the physiology or biochemistry of hypoxia (reduced levels of oxygen) tolerance during development in vertebrate embryos. In most species, relatively brief bouts of severe hypoxia are lethal or teratogenic. An exception to such hypoxia intolerance is the annual killifish Austrofundulus limnaeus, in which populations persist in hypoxic environments. This species inhabits seasonal ponds in Venezuela, surviving through the dry season in the form of diapausing embryos. Embedded in the pond sediment, embryos of A. limnaeus are routinely exposed to hypoxia and anoxia (lack of oxygen) as part of their normal development. Here, we exposed embryos to …

Embryos of the annual killifish Austrofundulus limnaeus display a remarkable tolerance to anoxia during development, most notably during embryonic diapause. Little is known about the metabolic or enzymatic changes that accompany this state of anoxia tolerance. This study examined the metabolic changes associated with exposure to anoxia by measuring the activity of the enzyme phosphoenolpyruvate carboxykinase (PEPCK), and by profiling the concentration of 31 metabolites ranging from amino acids to citric cycle intermediates at 4 different developmental stages, diapause 2 (DII), 4 days post diapause (dpd), 12 and 22 dpd. Green Salamanders (Aneides aeneus) are the only salamanders in West Virginia to exhibit an arboreal lifestyle. The focus of this study was to determine the cellular anatomy of the distal digit structures and how these structures may influence climbing ability. Comparative histology, comparative morphometrics, and comparative osteology of Green Salamanders, Cumberland Plateau Salamanders (Plethodon kentucki), and Slimy Salamanders (Plethodon glutinosus), were also used to determine if Cumberland Plateau Salamanders had the potential to compete with Green Salamanders for arboreal habitats. The objective of this study was to characterize membrane potentials in systemic arteries of Pacific hagfish, Eptatretus stouti, Rainbow trout, Onchorhyncus mykiss, and Sea lamprey Petromyzon marinus. Previous studies have characterized membrane potentials in piscine tissue. However, these studies utilized non-vascular tissues such as cardiac muscle and skeletal muscle (69, 161). Characterization of membrane potentials in fish vasculature is without precedent in the literature. The hypothesis of this study was that membrane potentials in fish vascular smooth muscle differ little between species when comparable vasculature is evaluated.

Populations of Desmognathus quadramaculatus were compared for genotypic and phenotypic differences across 5 states in the Southern Appalachians. Series consisting of 10 salamanders each were collected from West Virginia, Virginia, Tennessee, North Carolina and Georgia. Salamanders were assessed for genotypic differences by using allozyme (proteins with polymorphic loci) electrophoresis staining for 13 loci of the muscle, liver and stomach tissue. Nei’s genetic distance was used to score genetic variation between populations. Phenotypic differences were assessed by comparing 10 external morphological measurements between populations. Mahalanobis’ distance was used to score morphological differences between populations. Both genetic and morphological differences were correlated …

The blacknose dace, Rhinichthys atratulus, is a cyprinid minnow common to shallow streams throughout North America. It has been shown to be sensitive to anthropogenic stress and a study of the genetic nature of R. atratulus populations may yield information about their habitats. In West Virginia, two subspecies of R. atratulus are present, R. a. atratulus and R. a. obtusus, which are separated by the Allegheny divide. In north central West Virginia, R. a. atratulus was observed in the Cheat River drainage, an area previously thought be within the range of R. a. obtusus. Fish of the R. a. atratulus …
