Conjugated linoleic acids (CLA) have been shown to regulate many physiological functions affecting human health. These CLA can endogenously originate from trans fatty acids through the enzymatic activity of desaturase, or they are produced from dietary polyunsaturated fatty acids via the biohydrogenation process in the rumen. A minimum of seventeen CLA isomers have been identified in the intestinal contents of ruminants; however, the entire synthetic pathway for all isomers is not known. Wastewater effluents have been shown to contain a variety of anthropogenic compounds, many of which have endocrine-disrupting properties. While multiple laboratory studies have shown the effects of such compounds on an individual basis at elevated concentrations, little research has attempted to characterize the effects of exposure to environmentally relevant mixtures of endocrine disruptors. The current study examined the effects of long-term exposure to graded concentrations of wastewater effluent on the fathead minnow, Pimephales promelas, and the northern leopard frog, Rana pipiens. Changes in mating signals often result in the development of new species, thus, understanding the genetic basis of traits that confer pre-mating reproductive isolation can shed light on the speciation process. This study used interspecific hybridization between poeciliid fish, a sailfin molly (Poecilia velifera) and a shortfin molly (P. mexicana), to generate reciprocal F1 and backcross hybrids to investigate patterns of inheritance of traits that contribute to mating signal differences between these two groups of mollies. Excessive exercise may induce osteoarthritis (OA), or degeneration, of articular cartilage, which is a leading cause of lameness and decreased use in horses. The purposes of this study were to utilize a sheep model to determine the effects of circular and linear exercise on the macroscopic and microscopic characteristics of articular cartilage in the metacarpal-phalangeal (MCP) joint and to evaluate biochemical changes in serum and synovial fluid compared to non-exercised control lambs.

Biochemical endpoints are useful when determining contaminant exposure in aquatic organisms. However, behaviors can also be impaired by environmental contaminants. Links between changes in brain biochemistry and behavior have been made, and understanding these relationships could increase the utility of these endpoints in ecological risk assessments. The overall goal of this research was to determine relationships between an ecologically relevant fish behavior and brain biochemistry. Ovarian hyperstimulation used in conjunction with embryo transfer techniques is an assisted reproductive technology that aids cattle producers in reaching their reproductive, genetic, and financial goals; however, viable embryo recovery results are inconsistent, possibly because the reproductive endocrine profile is poorly understood. The soluble organic matrix (SM) extracted from the shell of the Eastern oyster, Crassostrea virginica was analyzed to identify the constituent proteins and their structures. Two acidic proteins with estimated molecular weights of 48 and 55 kD were isolated and determined to have highly related structures. Both proteins are comprised primarily of aspartate, serine, and glycine which represent nearly 90% of the total amino acids. Studies indicate that both proteins are phosphorylated and have estimated isoelectric points in the range of 3 to 5. Evidence suggests that poly-aspartate and poly-serine domains are present in the 48 kD protein. Amino terminal …

The objective of this research was to further modify and develop the Clemson Partitioned Aquaculture System (PAS) design resulting in a system design capable of yielding economic feasible production rates of marine shrimp, Litopenaeus vannamei, in excess of 45,000[kg/ha] (40,000[lb/ac]) within a 5 month culture period while minimizing impact on the surrounding environment. The operation of the pilot-scale high-rate prototype shrimp culture system showed that it is possible to combine chemoautotrophic and photoautotrophic processing of feed and waste products within the system. The focus of this study was to understand and evaluate the effect of noradrenaline (NA) on two prominent fouling marine invertebrates, the Eastern Oyster, Crassostrea virginica and the Striped Barnacle, Balanus amphitrite for the purpose of developing a novel fouling deterrence strategy.
To understand the effect of NA at a cellular level the immune cells (hemocytes) of C. virginica were chosen as representative molluscan cells. Upon stimulation with 10 micro-molar of noradrenaline ~4% hemocytes of C. virginica labeled positive for the beta-adrenergic receptor (beta-AR). Superovulation is an assisted reproductive technique used to hyperstimulate the ovaries in an attempt to proliferate desirable genetics by increasing the number of dominant follicles and oocytes available per estrous cycle. Unfortunately, the process is inefficient and costly due to FSH injection protocols, labor requirements, and animal stress. A sustained release implant as a novel FSH delivery mechanism could increase the efficiency and utility while reducing cost and animal stress. Positive density dependence (the Allee effect) has been reported in a number of species that are attracted to conspecific cues. This effect has been shown to influence the settlement of many species of marine invertebrates. Caribbean spiny lobsters are gregarious den dwellers attracted to conspecific cues. Previous studies have suggested that the benefit of conspecific attraction is a reduced predation risk by decreasing the time to find crevice shelters (the guide effect) or by cooperative group defense once sharing shelters. The role of thrombocytes, nucleated blood leukocytes, on immune responses in chickens fed diets supplemented with vitamin C has not been studied in the past. This study investigates the effects of three types of diets(control, control with corticosterone, and control with corticosterone and vitamin C) in 14-day old broiler chicks on the expression of pro-inflammatory cytokines IL-1beta, IL-6, and IL-12 in thrombocytes. Distributions of Hawaiian stream fishes are typically interrupted by waterfalls that divide streams into lower and upper segments. Larvae hatched upstream are flushed into the ocean, and must climb these waterfalls to reach adult habitats when returning back to freshwater as part of an amphidromous life cycle. Stream surveys and studies of climbing performance show that Lentipes concolor can reach fast-flowing upper stream segments, but that Awaous guamensis reaches only slower, lower stream segments. Differences between manual and machine motility analysis was determined for total motility (MOT) of bull semen samples (N=117) three ways: manual analysis on a microscope and on computer-assisted semen analysis (CASA) systems: Hamilton-Thorne IVOS and Minitube SpermVision (SV). CASA analysis includes progressive motility (PRO) and related kinematic parameters. Analysis indicated differences between MOT: 83.06 (IVOS), 75.84 (manual), 65.48 (SV), and PRO: 44.25 (IVOS) and 60.07(SV). Relationships existed between manual MOT and both IVOS MOT (r=0.32) and SV MOT (r=0.59). The relationships between IVOS and SV MOT and PRO was notable (r=0.70 for both MOT and PRO).

In this study, I estimated the population size of migrating Alabama shad below Jim Woodruff Lock and Dam (JWLD) in the Apalachicola River located in the central panhandle of Northwest Florida using mark recapture and relative abundance techniques. The number of marked fish was adjusted for tag loss, emigration and mortality. The population size of migrating Alabama shad near JWLD was estimated at 26,029 (95% C.I. = 15,174 - 49,040) in 2005 and as 972 (95% C.I. = 270 - 9,720) in 2006. In this study, I investigated the potential for temperature to serve as a mechanism for maintenance of reproductive isolation in a sympatric population of Atlantic and Gulf striped bass in the Apalachicola and Ochlockonee rivers, Florida. Adult striped bass (n= 66, mean weight = 8.3 kg, mean total length = 780.6 mm) were collected from February to May below Jim Woodruff Lock and Dam, on the Apalachicola River, and below Jackson Bluff Lock on the Ochlockonee River, Florida. Age and growth of Alabama shad Alosa Alabamae were estimated by examining scales and otoliths from 203 adult fish collected on their spawning run in the upper Apalachicola River from 2005 and 2006. Ages of Alabama shad ranged from 1 - 4 years. Scales and otoliths both gave similar age estimates. Age distributions from this study differed from those of previous studies for both males and females. Female Alabama shad, on average, were found to be larger than males at age 2 and age 3. Alabama shad demonstrated a positive correlation of length to fecundity, with fecundity estimates ranging from …

Osteoarthritis is a leading cause of chronic lameness in horses. Many treatment options for osteoarthritis have limited effectiveness and are expensive. Treatments that stimulate cartilage development may promote cartilage healing. Bovine supramammary lymph nodes are available at no cost and contain bioactive growth factors that could promote cartilage healing. The purpose of this study was to measure the proliferative response of equine chondrocytes to bovine lymph node proteins. Chondrocytes were harvested from adult horses. Experiments were conducted to evaluate whether or not the lymph node extract could be used as a substitute for bovine growth serum in cell media. This suspended sediment, even in the absence of adsorbed contaminants, may be a significant stressor to aquatic organisms. Understanding speciation remains a holy grail of evolutionary biology. One useful approach is studying the evolutionary mechanisms important in population divergence to infer the mechanisms important in speciation. This method is especially useful when closely related species can be compared to determine whether intraspecific differences parallel interspecific differences. I studied population divergence in two species of Mexican sailfin mollies, Poecilia velifera and P. petenensis. In the present study, five microsatellite loci are used to examine genetic diversity and determine genetic similarities among 66 radiated tortoises (Geochelone radiata) located in ten different zoos and institutions in the United States. These tortoises, representatives of one of the world's most endangered species, were originally collected in Madagascar and are being used as founders in the development of an organized breeding program, the radiated tortoise Species Survival Plan (SSP). In North Carolina, Precis coenia that emerge during the Summer months exhibit a ventral hindwing (VHW) with well-defined reddish-brown and brown pattern elements on a light tan background. During late Summer and early Fall, however, individuals begin to appear with poorly defined or obscured pattern elements on a dark reddish-brown background. The present study shows that the Fall (rosa) color morph can be induced by either low rearing temperatures or short daylengths. The effect of such conditions seems to be cumulative throughout the larval life, although animals are much more sensitive during the last 24 hours of larval …
