Rising sea levels and saltwater intrusion in aquifers pose significant challenges for South Florida agriculture, leading to increased groundwater salinity and potential crop losses. Utilizing salttolerant crop species presents a potential solution for saline soils and regions with active saltwater intrusion. However, the effects of soil salinization through groundwater alone remains less studied. This research investigates the impact of short-term, below-ground saltwater stress on the growth, survival, and overall health of commonly grown cherry tomatoes (Solanum lycopersicum). Plants are exposed to a wide variety of environmental stress in the wild and have developed an equally diverse set of adaptations to tolerate them. The evolutionary processes that have led to this functional diversification, and the specific genes and physiological mechanisms involved, are of immense interest to both evolutionary biologists and crop breeders. In this dissertation I investigate adaptation to different types of environmental stress in two economically important grass species, seashore paspalum (Paspalum vaginatum Sw.) and rice (Oryza sativa L.).

Salinity is the major threat to global wheat production, particularly in arid and semiarid areas. Breeding salt-tolerant cultivars is one feasible solution, while the presence of genetic variation is a prerequisite for genetic improvement. To detect genetic variation for salt tolerance in wheat, a total of 150 wheat genotypes were tested for seedling-stage tolerance response at 300 mM NaCl in hydroponic culture. Significant differences (P ≤ 0.001) were identified in wheat for seedling traits, and 20 salt-tolerant and 5 susceptible genotypes were selected on the basis of root and shoot weights. The aim of the current study was to investigate the effects of glycinebetaine (GB) on oxidative stress and some key growth parameters in maize (Zea mays L. 'DK 647 F1') grown under saline conditions. Maize seedlings were grown in pots containing perlite. The experiment was designed in a 2 × 2 factorial arrangement with 2 levels (0 and 100 mM) of sodium chloride (NaCl) and 2 levels (25 and 50 mM) of GB sprayed onto leaves of maize seedlings. Soil salinity limits plant productivity and quality. We evaluated the response of 12 aspen (Populus tremula) families to salt stress in two different soils irrigated for 4-weeks with 0, 80 or 160 mM saline solution. Easily measurable characteristics such as shoot height, leaf number, dry mass as well as the distribution of sodium (Na+) ions were measured in 5-month-old aspen seedlings raised in controlled greenhouse conditions on two different soils. Growth among families varied significantly, and the interaction between family and soil type was significant. Na^+/H^+ antiporters have been suggested to play important roles in salt tolerance in plants. English cordgrass (Spartina anglica) is a gramineous halophyte with very strong salt tolerance. It possesses salt glands in its stems and leaves, through which excess salt can be excreted. In this study, a vacuolar Na^+/H^+ antiporter homologous gene (temporarily named SaNHX1) was isolated from Spartina anglica by RT-PCR and rapid amplification of cDNA ends (RACE). In this study the salt tolerance of stock (Matthiola tricuspidata) was investigated. For this purpose, a control and 4 different concentrations of NaCl + CaCl_2 were applied to plants grown under greenhouse conditions. Na, Cl and electrical conductivity (EC) analyses were conducted in soil, plant and leached samples. Yield, plant growth parameters and plant dry matter were also determined. All plants survived until the end of the experiments. Treatments affected the ion content and EC of leached and soil samples significantly. The salt tolerance of some barley genotypes and their cross populations were evaluated at seedling stage in a greenhouse, and at the generative stage in a saline field in line x tester analysis. Promising crosses were determined through comparison of heterotic effects. Tokak-157/37 x LIGNEE-640, Quantum x MENTA "S" and Kaya-7794 x MENTA "S" were found to be superior cross combinations with respect to salt tolerance in both greenhouse and field conditions. The salt tolerance of eight barley genotypes and their 15 F1 hybrid populations were investigated in line x tester analysis for seedling traits in greenhouse and generative stage characteristics in saline fields. The data indicated that there was sufficient genetic variability among genotypes for salt tolerance in barley. The MENTA "S" genotype for general combining ability and Tokak 157/37 x ZARZA "S", Quantum x No.56000/MISC-233 and Kaya-7794 x ZARZA "S" crosses for specific combining ability had positive and significant estimates both in greenhouse and field conditions.
