A Small Intron Of Drosophila Gamma Carboxylase, Not Predictable In Silico For Mammalian Splicing Machinery, Is Spliced Improperly In Human Cells, Solmaz Moniri Javadhesari, Alireza Zomorodipour, Spideh Valimehr

The splicing of two small introns of 58 and 67 nucleotides (nts) of the Drosophila gamma carboxylase (DγC) in cultured human cells was examined. The two introns are below the minimum size, which was proposed as a requirement for an intron to be spliced in human cells. None of the well-known software examined was able to provide a proper prediction for the DγC intron 1 in a mammalian host. Machilus thunbergii Sieb. and Zucc. (Lauraceae) is a medicinal plant used to treat edema and pain in China and Taiwan. The anti-inflammatory effects of M. thunbergii were not clear. In this study, we evaluated the anti-inflammatory effects of M. thunbergii extract in LPS-stimulated RAW 264.7 cells. The cells were pretreated with various doses (12.5 μg/mL-100 μg/mL) of M. thunbergii extract and activated with LPS. The cyclin-dependent kinase (CDK) inhibitors purvalanol and roscovitine are therapeutic agents that control cell proliferation through regulating cell-cycle machinery. They also affect polyamine (PA) metabolism, which is activated in malignant tissues. Therefore, PA catabolism became a remarkable target in cancer therapies. Induction of the PA catabolic enzyme spermidine/spermine N1-acetyltransferase (SSAT) is under the control of transcription factors such as NF?B and PPARγ. Thirty different wheat genotypes, including landraces, cultivars, and related wild species, were analyzed for puroindoline genes and proteins. Full-length Pina and Pinb genes were amplified using gene-specific markers where 13 genotypes amplified both genes, while 17 genotypes had either one or none of the puroindoline genes. None of the puroindoline genes were detected in any of the Triticum durum cultivars. Genotypes with both puroindoline genes had a soft grain texture, while other genotypes had hard grains. Capsaicin possesses cytotoxic/anticancer activity and shares some common structural features, including a benzene ring and a long hydrophobic carbon tail, with the histone deacetylase (HDAC) inhibitors trichostatin A and suberoylanilide hydroxamic acid. The aims of this study were to investigate HDAC inhibitory and cytotoxic activities of a synthetic derivative of capsaicin, hydroxycapsaicin (6-hydroxy-N-(4-hydroxy-3-methoxybenzyl)-8-methylnonenamide), in colon cancer cell lines. Hydroxycapsaicin inhibited HDAC activity in vitro (IC50 = 72 µM) much more effectively than the prototype capsaicin (IC50 > 13.1 mM) and also exhibited HDAC inhibitory activity in human cells (HeLa cells). The sirtuin (SIRT) gene family is reported to regulate critical intracellular processes from aging to cellular metabolism and repair. SIRT3 knockout (SIRT3-/-) mice develop receptor positive mammary tumors starting at 13 months and SIRT3 expression is decreased in human breast cancer samples as well as several other diseases. Cyanobacteria can be used to remove nitrogenous compounds from wastewater, but a major bottleneck in the process is the separation of cyanobacterial biomass from the treated water discharge, which may cause eutrophication. The current study assessed the suitability of three periphytic cyanobacteria (Geitlerinema sp., Gloeotrichia sp., and Lyngbya sp.) isolated from shrimp ponds. These cyanobacteria were immobilized by self-adhesion to polyvinyl chloride sheets, forming mats, and were screened for their efficacy to reduce nitrogenous compounds and phosphorus. Secondary dental caries are one of the major reasons for restoration replacements. Incorporating antimicrobial properties into dental materials would limit the initiation and progression of dental caries. In the current study, dental composites having 1%, 5%, and 10% (w/w) sodium pentaborate pentahydrate were prepared and analyzed for their mechanical properties, degree of monomer conversion (DC) rate, antibacterial effects against Streptococcus mutans, and biocompatibility with human dental pulp stem cells (hDPSCs). Incorporation of boron into the composites significantly decreased flexural strength and DC in a dose-dependent manner, but the value for 1% boron-containing composite still remained within acceptable levels. Rice is one of the world's most important crops. Thus, it is necessary to improve it by genetic transformation. IR64 is a high-yielding and disease-resistant variety, but it is sensitive to abiotic stresses, mainly salinity and drought. Therefore, an efficient transformation system is a prerequisite for transforming this indica rice with a stress-tolerant gene, the potent osmolyte proline-coding P5CS gene of Vigna aconitifolia. Successful transgene integration was confirmed by polymerase chain reaction (PCR) and Southern blot analysis. A Mendelian pattern of transgene inheritance was also observed in the progeny plants. Phytoremediation is a low-cost and effective technology that removes pollutants, such as dye-containing effluents, from the environment through the use of plants. In the present study, the potential of Lemna minor L. for decolorization and degradation of the triarylmethane dye malachite green was investigated. The results revealed that the decolorization ability of the plant species is as high as 88%. The effect of some operational parameters (initial dye concentration, temperature, pH, and amount (weight) of plant) on the efficiency of biological decolorization process was determined. The current study reveals the effects of mycorrhization on heavy metal uptake and accumulation, antioxidant potential, and essential oil composition of Origanum majorana L., grown on soil polluted with industrial Cd and Pb. Two strains of Claroideoglomus claroideum (EEZ 54 and EEZ 55) were isolated from soil that is naturally rich in metals. EEZ 35 (Funneliformis mosseae) was isolated from a place with industrial contamination. Arnica montana L. (Asteraceae) is an economically important herb that contains numerous valuable biologically active compounds accumulated in various parts of the plant. The effects of carbon sources (sucrose, maltose, and glucose) at different concentrations (1%, 3%, 5%, 7%, and 9%) on growth were studied and GC-MS based metabolite profiling of A. montana hairy roots was conducted. The optimal growth and biomass accumulation of transformed roots were observed on an MS nutrient medium containing 3% or 5% sucrose. Phytoremediation of freshwater contaminated with crude oil is a technology that can restore damaged freshwater areas. Lemna minor is a small vascular plant that reproduces rapidly, is sensitive to a wide variety of pollutants, and is easy to culture. This study aimed to evaluate the phytoremediation capacity of L. minor in crude oil spiked cultures. From our prior studies concerning eugenol's biologic activity we observed the very good in vitro antifungal efficiency of this natural compound. Those positive results generated the need to supplement the available information with a comparative in vivo animal model. In this context, our current study was proposed to ascertain and compare the effects of eugenol with nystatin with a placebo control group (saline solution) by evaluating the cytohistological alterations in immunosuppressed Wistar female rats genitally infected with Candida albicans. The affinity of ß-lactamase inhibitory protein (BLIP) for TEM-1 ß-lactamase has raised hopes in the challenge of protein-based inhibitor discovery for ß-lactamase-mediated antibiotic resistance. Hemianthus callitrichoides 'Cuba' is an ornamental aquatic plant used in water gardens and phytoremediation. A protocol for its rapid and reproducible shoot organogenesis from apical shoot explants was developed. Optimum culture conditions for shoot proliferation were tested in MS media containing different concentrations of 6-($\gamma$,$\gamma$-dimethylallylamino)purine (2iP), 6-benzylaminopurine (BAP), 1-phenyl-3-1,2,3-thiadiazol-5-yl urea (TDZ), and gibberellic acid (GA3). Plant growth regulators significantly (P ≤ 0.001) affected the diameter, area, and fresh weight of shoot clumps.
