Various scanning electron microscopy preparative techniques have been used by researchers to examine sectional views of dry, mature soybean (Glycine~ CL.) Merr.) seed coats. Such previously employed techniques were utilized In our preliminary investigations of seed coat structure, but often yielded unacceptable preservation. Consequently, eight preparative techniques were evaluated In an effort to define conditions required to obtain consistently high quality preservation of soybean seed coats In sectional view. Of the eight procedures tested with the cultivar Williams 82, razor sections and mechanical fractures of dry seed coats yielded the poorest definition of anatomical features. Scanning Electron Microscopy

The lung appears to be the major dose-limiting organ in radiation of the thorax. Early responses (<1 week) involve the type II pneumocyte and increased surfactant biosynthesis and secretion. Later changes, which appear to be related to the surfactant response, lead to classical radiation pneumonitis, which is often fatal. Animals which survive radiation pneumonitis develop progressive fibrosis, a late-appearing response, which reduces compliance and available air space, and is usually fatal. This study centers on the fine structural changes in the lungs of LAF₁ mice, 63 weeks following various radiation exposures (5-13 Gy). Doses which are subthreshold in evoking surfactant and pneumonitic responses precipitate fibrosis and atelectasis by 63 weeks, and involve type II pneumocyte sloughing and degeneration. Of the two major deterrents to lung irradiation (pneumonitis and fibrosis), these results suggest that fibrosis always follows pneumonitis, but pneumonitis is not a necessary preliminary step to fibrosis. Bleomycin elicits several morphological alterations characteristic of radiation, and, when combined with the latter, appears to exacerbate radiation effects.

Characterization Of Small Metallic Clusters By Electron Energy Loss Spectroscopy, M. Acheche, C. Colliex, P. Trebbia

Small spherical tin and gallium clusters (diameters in the range 2 to 80 nm), prepared with a liquid metal ion source, were analysed in a scanning transmission electron microscope (STEM) by electron energy loss spectroscopy (EELS). Both volume and surface plasmon excitations were investigated and their dependence on the cluster size interpreted by classical and quantum mechanical models. A blue shift of the volume plasmon energy with decreasing radius R of the cluster was clearly detected. The full width at half maximum (FWHM) of the peak, related to the damping of the volume plasmon excitation, showed a dependence in 1/R. …

A Floristic And Taxonomic Study Of The Wood-Rotting Aphyllophorales Of The Spruce-Fir Forest Of The Great Smoky Mountains National Park, Hack Sung Jung

An ecological survey of the wood-rotting Aphyllophorales from the spruce-fir forest of the Great Smoky Mountains National Park was accomplished with emphasis on resupinate taxa. A total of 105 species are described within 51 genera of 6 families and discussed in terms of habitats, substrates, wood rot, host trees, elevation, and distribution. Sixteen species were successfully cultured. Analytical keys for genera and species are provided, and microscopic characters are illustrated in an appendix.

Nutrient Capital Sequestration In Pioneer Plant Communities On Surface-Mine Spoil, Gary Leon Wade

Biomass production and nutrient capital sequestration of four pioneer plant communities on a surface-mine spoil were compared. A Chenopodium album-dominated community (Treatment 4) produced the greatest amount of biomass. Next were a community derived from a forest topsoil seed bank spread over mine spoil (Treatment 2), a seed bank community with common reclamation species seeded into it (Treat­ ment 3 ) , and a mix of grasses and Lespedeza commonly used in reclama­tion (Treatment 1).

Isolation And Characterization Of Temperature-Sensitive Protein Synthesis Mutants Of Escherichia Coli By Directed Mutagenesis Of The Defective Bacteriophage Lambda Fus2, Kenton L. Lohman

Mutagenesis of the defective transducing bacteriophage lambda fus2 was used to isolate a collection of temperature-sensitive mutants of E. coli in the major ribosomal protein gene cluster. Four mutants were examined in detail. Two of the mutants were resistant to the ribosomal antibiotics neamine and spectinomycin. Another mutant was defective in 50S ribosomal subunit assembly at 42(DEGREES)C. The 30S subunit proteins S17 and S19 were changed in two different mutants. Each protein migrated as a more basic species in two-dimensional gels of ribosomal proteins. Microencapsulation As An Alternative To Emulsification For Preparing As A For Use In Papermaking, William J. Corriveau

The sizing chemical ASA is emulsified to make i t compatible with the papermaking process. Unfortunately, emulsified ASA is chemically reactive with water at the elevated temperatures found in papermaking systems. As an alternative, encapsulated ASA was made and investigated for its stability towards water at elevated temperatures.

A handsheet procedure was developed to analyze emulsified and encapsulated ASA. It was concluded that the procedure gave statistically reproducible results. Investigations with the procedure revealed important variables concerning the laboratory and m ill application of ASA. Evidence indicated that retention of ASA was critical to sizing development.

One of two planted loblolly pine trees (Pinus taeda L.) and over one of three (36%) planted slash pine trees (Pinus elliottii Engelm.) on non-old-fields in East Texas have a poor quality stem. For both species, o poor quality stem is three times more likely to occur on trees with crowns in the upper canopy then in the lower canopy.

Cisplatin Ototoxicity In The Guinea Pig: An Electrophysiological, Morphological, And Biochemical Study, Sarah E. Barron

The purpose of this study was to determine changes induced by cisplatin on hair cell (HC) morphology, compound action potential (CAP), Preyer reflex (PR), and Na,K-ATPase activity in the cochlear lateral wall. Guinea pigs were treated with either 0.42, 0.83, 1.17, 1.50 mg/kg cisplatin, i.p. or sterile water for 12 consecutive days and tested on day 13. HC condition was visualized with scanning electron microscopy (SEM), CAP was initiated with a 1 volt click, and Na,K-ATPase activity was measured by the method of Fiske and SubbaRow. While visiting Pocahontas State Park in Chesterfield County, Virginia during the fall of 1984, an unfamiliar blue-flowered plant was observed growing near the upper reaches of Swift Creek Lake. This proved to be Eryngium prostratum Nuttall ex DC., a species common near bodies of water in the southeast U.S. While several standard floras include Virginia in the distribution of this plant (Fernald 1950, Gleason 1952, Gleason & Cronquist 1963, Radford et al 1968, Godfrey & Wooten 1981), the only counties for which it is recorded in Harvill et al (1981) are along the extreme southern border of the state, i.e., …

Improvements in intraocular lens implants during the past ten years have revolutionized cataract surgery. Both anterior and posterior chamber lenses are currently in wide use, but the optical superiority of the posterior chamber location has yet to be demonstrated. To do so, a more detailed analysis is required of visual function than is clinically routine. Also required is appreciation that the retina is an integral part of the eye's optical system because of its directional characteristics, which are known as the Stiles-Crawford effect. The pathologic diagnosis was supported or confirmed in three out of four cases that had an adequate cytologic specimen. The results demonstrate that adjunctive immunocytologic techniques can be used in combination with fine-needle aspiration for a variety of orbital tumors. The admission nutrition status of 135 consecutive general medical patients admitted to a single nursing unit was evaluated using a simple questionnaire and available laboratory studies. A statistical analysis of the data obtained indicates that prolonged length of stay may be related to admission nutritional status.
