Some animals are incredibly social, living and working together as one cohesive group. Alternatively, many animals are solitary, never living with and rarely interacting with others. A large body of biological research has focused on understanding the role that brains play in promoting these behavioral differences across species. Even so, it remains unclear why some brains facilitate social behavior while others do not. My dissertation aims to advance our understanding of this concept by characterizing bees’ brains and how they change over a lifetime. Most flowering plants, including many cultivated food crops, will only produce well-developed fruits and seeds if pollen is transferred from one flower to another with the help of an animal pollinator. Honey bees are the most well-known and ubiquitous but are poor pollinators of some commercially important crops, or are in poor supply during crop bloom. In such cases, farmers will employ other managed pollinators such as bumble bees or solitary bees like mason and leafcutting bees. Parkinson's Disease (PD) is a neurodegenerative disease characterized by motor (e.g. tremors) and non-motor symptoms (e.g. cognitive impairment). PD patients' change in cognitive functioning can be observed using the following classifications: cognitively intact, mild cognitive impairment (MCI), or dementia (PDD). MCI has many subtypes, one of which is MCI reversion which is defined as those with MCI at one time point reverting to cognitively intact later. While there is limited research into the utility of MCI reversion and its relationship with cerebrospinal fluid (CSF) biomarkers in PD, this study will begin to elucidate this relationship. Arginine methylation is an essential post-translational modification (PTM) in cells. Protein arginine methyltransferase 5 (PRMT5) is the primary enzyme that catalyzes symmetric dimethyl arginine (SDMA) and requires methylosome protein 50 (MEP50) for stability and enzymatic activity which are necessary for life and development. Effector proteins bind different types of PTM’s to facilitate signaling. Staphylococcal nuclease Tudor domain containing 1 (SND1) is an effector that specifically binds SDMA via its single C-terminal Tudor domain. The motivation and capacity to be social is necessary for human survival. Successful learning of complex, prosocial behavior stems from the ability to perceive and respond to visual cues, such as the body language and facial expressions, from others in our environment. This dependence on visual information to guide social interaction is especially true for humans and non-human primates. Efficient replacement of dead cells in epithelial tissue is crucial for maintaining barrier function and tissue homeostasis. Apoptotic cells can signal to neighboring cells to stimulate proliferation and compensate for cell loss and maintain overall cell numbers in normal physiology and cancer. While dying cells can transmit instructive cues to neighboring cells, the molecular mechanisms that induce cell division are not well understood. Workers in the manufacturing and construction industries are at risk of lead exposure in projects that involve removal or installation of tiles, and other individuals exposed to lead, including children may also be at risk. The concern with lead in tiles is thought to be related to the glazing process or the country where the tile was produced. This research study investigates how specific running variables and demographic characteristics impact performance, or speed, for ultramarathon runners. These variables and characteristics include biological sex, cadence, age range, total distance of the race, duration of the race, years of running experience, total number of ultramarathon races ever completed, and number of races completed in the last twelve months. An ultramarathon is any running distance over a standard marathon or 42.2 kilometers. Participation in ultramarathons has increased by 345% since 2010, with females making up 23% of participants. Food poisoning is a common term used to describe what is actually foodborne illness. Despite the fact these illnesses can become deadly, foodborne illnesses are endemic and common in the United States. Nonetheless, they are completely preventable simply by following proper food safety procedures. The Food and Drug Administration identified the most common foodborne pathogens responsible for most foodborne infections, as well as the five risk factor categories to target in order to decrease the risk of cases and outbreaks.

A major outstanding question is how the assemblage is potentially structured by mesoscale (10 - 100s km) oceanographic features such as eddies, fronts, and riverine plumes. In arid and semi-arid environments, burrowing mammals play a key role in increasing landscape heterogeneity through facilitative species interactions. The loss of burrowing mammal populations can consequently lead to negative effects cascading through the ecosystem. It is critical to understand these facilitative interactions for conservation and management. Understanding facilitative interactions may improve wildlife management tools, like translocation, which is often not very successful. This project had two primary goals: (1) to explore opportunities to further a deep-ocean vehicle’s reach using alternative pressure spheres, and (2) to implement an existing deep-ocean vehicle (lander) in active scientific research.

I gained a greater understanding of the limitations and design choices made for existing pressure spheres using Finite Element Analysis (FEA). My simplified FEA model predicted sphere failure for the existing 30% Fiber Glass 70% Nylon injection molded spheres at an external pressure of 3,954psi or 2,690m ocean-depth (only a 7.38% error compared to the tested minimum failure depth), so I determined it a valid model. The thesis comprises three studies. The first study in this thesis (Chapter I) focused on a six-year corn field experiment to assess the influence of conservation agricultural practices on soil properties and their long-term effects on water-use efficiency and yield. Non-tillage management did not significantly reduce soil bulk density compared to conventional tillage, as no differences were observed between non-tillage and tillage systems throughout the 6-year experiment. There was no difference in total water-use efficiency among soil management practices in 2018, 2019, 2020, and 2021. Motor unit (MU) recruitment and firing rate (MUFR) modulation are the primary factors governing force control. Differences in the anatomical distributions of α-motoneurons, muscle properties, and perception of fatigue between males and females may cause sexually dimorphic MU firing behaviors. The purpose of this dissertation was to further examine external, anatomical, and methodological factors that may influence observations of sex-related differences in neuromuscular function. In chapter 2, sex differences in how mental fatigue may influence MU firing behaviors were examined. Mental fatigue did not significantly alter MU firing behavior, maximal force, or force tracing ability in either males or females. …

The prefrontal cortex (PFC) is one of the cortical areas responsible for complex cognitive abilities, a function that is believed to arise from increased persistent activity within its microcircuits. Activity within microcircuits is regulated by parvalbumin-containing (PV), calbindin-containing (CB), and calretinin-containing (CR) inhibitory interneurons (INs). The receptor for advanced glycation end-products (RAGE) is a transmembrane cell surface protein of the immunoglobulin superfamily that acts as part of both the innate and adaptive immune system. RAGE is highly expressed in lung tissue and is therefore of interest in the pulmonary immune response. Specifically, RAGE mediates several cell-signaling responses such as inflammation and apoptosis. A few species of boreal trees form symbiotic relationships with many ectomycorrhizal (ECM) fungi that allow trees to thrive on nutrient poor soils in Canada. A small number of ECM species grow on tree seedlings raised in nurseries for reforestation of clearcut sites. The native ECM fungal community composition of a mixed spruce-fir forest in Newfoundland was determined through next generation sequencing. With the introduction of nursery seedlings, the transfer of native ECM fungi to seedlings and nursery-established ECM fungi to roots was investigated over 20 months. Thiopeptides are a family of antimicrobial peptides that are characterized for having sulfur-containing heterocycles, and for being highly post-translationally modified. Numerous thiopeptides have been identified; almost all of which inhibit protein synthesis in gram-positive bacteria. These intrinsic antimicrobial properties make thiopeptides promising candidates for the development of new antibiotics. The antimicrobial peptide micrococcin is a thiopeptide that is synthesized by the ribosome and undergoes several post-translational modifications (PTMs). Robust functional connectivity in sensorimotor resting-state network (RSN) has been linked to positive neurodevelopmental outcomes in neonates. In the current study, we aimed to map the developmental trajectory of sensorimotor RSN in awake neonates using functional near-infrared spectroscopy (fNIRS). We acquired fNIRS resting-state data from 41 healthy newborns (17 females, gestational age range=36+0 to 42+1 weeks) within the first 48 hours after birth. At the group level, we observed robust positive connectivity in numerous channel-pairs across the sensorimotor network, especially in the left hemisphere. The California sea hare (Aplysia californica) is an important biomedical model for molecular neurobiology, electrophysiology, learning, and memory due to their well-mapped and large neurons and well-characterized learning capabilities. The National Resource for Aplysia (NRA, University of Miami) maintains large stocks of live animals and relies on regular shipments of wild-caught individuals to maintain genetic diversity. Sensorimotor gating is a fundamental pre-attentive process, which can be operationally measured through prepulse inhibition (PPI) of the startle response. PPI describes the suppression of the startle response when a non-startling stimulus (“prepulse”) precedes the startling stimulus (“pulse”). As a hallmark of schizophrenia, PPI deficits are also found in other neuropsychiatric disorders and are associated with cognitive overload and attention impairments (Braff et al., 2001). However, currently-used dopaminergic antipsychotics exhibit inconsistent effects on PPI in affected individuals (Geyer et al., 2001; Frau et al., 2014; Lally et al., 2016). Atlantic Salmon (Salmo salar) and Chinook Salmon (Oncorhynchus tshawytscha) are important culturally, recreationally, economically, and are a common aquaculture species in Canada and the United States. In the Pacific Northwest, blooms of toxic algae result in millions of dollars annually in losses for salmonid aquaculture producers. Blooms of harmful cyanobacteria produce microcystins which have been linked to net-pen liver disease which has large financial impacts on net pen salmon operations. Microcystin-LR, the most toxic variant, bioaccumulates in the liver and disrupts normal cellular activity by inhibiting protein phosphatases leading to deleterious effects on growth, immune status, and liver function.
