Mitochondria are primarily appreciated for the generation of adenosine triphosphate (ATP), a chemical store of energy required by all cells. These organelles, however, also play key roles in apoptosis, autophagy and shaping cytosolic calcium (Ca2+) signaling via Ca2+ uptake into the mitochondrial matrix. This Ca2+ uptake is mediated chiefly via the mitochondrial Ca2+ uniporter (MCU), an inner mitochondrial membrane protein that oligomerizes to form a Ca2+ selective pore. MCU is regulated by several protein binding partners, including the recently identified MCU regulator-1 (MCUR1). MCUR1 stabilizes a higher order MCU heterocomplex through interactions with MCU …

There is a growing body of research evaluating head impacts in sport, but studies have not evaluated youth football in Canada. It is important to evaluate football in Canada as the rules and field size are different than American football, and this may influence the impact exposures. The purpose of this study was to quantify the head impact exposures in youth Canadian football and determine if a season’s worth of impacts could cause impairments in executive function. Players (n=50) had their head impacts recorded using an impact measuring device (GForceTracker) throughout all contact practices and games. A subset of players …

How are cholangiocarcinoma cells different from non-malignant cholangiocytes?
All of the hallmarks of cancer apply- those reported in this dissertation include cell proliferation, migration, apoptosis, and evasion of growth suppression. My studies began with testing how apoptosis might be regulated through embelin, a small molecule reported to sensitize cells to apoptosis by blocking XIAP. My data however revealed that embelin reduced the proliferative capacity in cholangiocarcinoma cells, but did not increase cell death. Malignant cells exhibit dysregulation of microRNA processing and expression. Hence, my studies seeking ways that cholangiocarcinoma eludes apoptosis transitioned to the oncomiR miR-106b, which is overexpressed in …

The Hippo pathway is conserved regulator of organ size through control of proliferation, apoptosis, and stem-cell self-renewal. In addition to this important function, many of the canonical signaling members have also been shown to be regulated during mitosis. Importantly, Hippo pathway components are frequently dysregulated in cancers and have attracted attention as possible targets for improved cancer therapeutics. Further exploration of Hippo-YAP (yes-associated protein) signaling has revealed new regulators and effectors outside the canonical signaling network and has revealed a larger non-canonical network of signaling proteins in which canonical Hippo pathway components crosstalk with important cellular homeostasis and apoptosis signaling …

Synthetic polymers have been extensively explored for improved delivery of anticancer agents. Polymers can be designed as either carriers of existing drugs or as polymeric drugs with intrinsic pharmacological activity. Advantages of such polymeric drugs include common positive features of polymer-drug conjugates, such as targeted delivery of drugs, altered pharmacokinetic and biodistribution, improved drug safety, but also favorable pharmacological activities due to multivalent receptor-ligand interactions.

Single-cell sequencing enables the rapid acquisition of genomic and transcriptomic data from individual cells to better understand genetic diseases, such as cancer or autoimmune disorders, which are often affected by changes in rare cells. Currently, no existing software is aimed at identifying single nucleotide variations or micro (1-50bp) insertions and deletions in single-cell RNA sequencing (scRNA-seq) data. DNA damage is frequently induced in cells by both endogenous and exogenous agents. DNA damage, particular double strand breaks (DSBs) may lead to genomic instability, and the progression of cancer, aging, neurodegeneration, and other human diseases. The cell employs two major DSB repair pathways, including homologous recombination (HR) and Non-homologous end joining (NHEJ), but the detailed mechanisms of DSB repair remain to be further revealed.

As critical mediators of cell-to-cell communication, gap junctions (GJs) are comprised of membrane channels that directly link the cytoplasm of adjacent coupled cells thereby allowing for the passage of ions, small metabolites, and secondary messengers. Each channel is formed by the apposition of two connexons from adjacent cells, each composed of six connexin (Cx) proteins. Each GJ channel functions to promote signal propagation and synchronization of cells and tissues in organs. N-methyl-D-aspartate receptors (NMDARs) are ligand-gated ion channels consisting of two GluN1 subunits and two other subunits from among GluN2A-2D and GluN3A-3B subunits. NMDARs play critical roles in synaptic plasticity, learning and memory, and higher brain function such as cognition and perception. Dysfunction of NMDARs (hyper-function and hypo-function of NMDARs) are related to various diseases, including stroke, schizophrenia, Alzheimer’s disease, and others. However, to date, NMDARs antagonists have mostly failed in clinical trials due to adverse effects.

Understanding the degree to which species distributions are controlled by climate is crucial for forecasting biodiversity responses to climate change. Climatic equilibrium, when species are found in all places which are climatically suitable, is a fundamental assumption of species distribution models, but there is evidence in support of climate disequilibria in species ranges. Long-lived, sessile organisms such as trees may be especially vulnerable to being outpaced by climate change, and thus prone to disequilibrium. Black soldier flies (BSF), Hermetia illucens (Linnaeus) (Diptera: Stratiomyidae) consume decaying organic waste as larvae (BSFL) and can be used for recycling a variety of biogenic wastes. BSFL can also be processed into value-added animal feeds, including those used in aquaculture. An overarching goal of this project was to obtain additional insights into BSF biology to improve their rearing and handling in future mass-production facilities serving Maine aquaculture.

Using species distribution models (SDMs) in Natural History Collections (NHCs) can influence how humans implement conservation changes in flora and fauna communities and ecosystems. Microplastics are small plastics resulting from degradation or the dumping of raw material and can lead to deleterious impacts on the coastal marine environment and its organisms. The aim of this study was to investigate the differences in nutrient limitation within the beach and marsh environments at Waties Island, SC. Conducting the experiment on Waties Island allowed most physical factors to be held as constant, meaning the marsh and beach environments would serve as the only variables. Experiments were performed in the winter, spring, summer and fall of 2018 to determine temporal changes in limitation to phytoplankton and cyanobacteria growth. Maturase K (MatK) is the only group II intron encoded protein in the chloroplast of land plants. Maturases are prokaryotic enzymes that catalyze formation of the lariat structure needed for intron removal from precursor RNAs. The chloroplast maturase MatK, is a descendant of prokaryotic maturases, however, unlike its prokaryotic relatives, MatK is thought to catalyze excision of, not only its own intron, but also the introns of other group II introns in the plastome. Corn (Zea mays) is frequently infected by a soil fungal pathogen Aspergillus flavus. The fungus produces aflatoxins, which cause liver cancer. Maize inbred lines that are resistant to infection by A. flavus have been developed, and these inbred lines provide excellent models for studying molecular mechanisms of maize resistance to the fungus. MicroRNA-like RNAs (milRNAs) recently identified in A. flavus had been found to be correlated with aflatoxin production conditions, suggesting that the milRNAs might play a role in the regulation of aflatoxin production. N-methyl-D-aspartate (NMDA) receptors are a subtype of glutamate receptors that are widely expressed in the brain, where they mediate critical functions. While the actions of the predominate GluN2 NMDAR subunits, GluN2A and GluN2B are relatively well understood, the function of GluN2C and GluN2D in the telencephalon is largely unknown. To better understand the possible role of GluN2C and GluN2D subunits, we used fluorescence in situ hybridization together with multiple cell markers to define the distribution and type of cells expressing GluN2C and GluN2D mRNA. Lysine demethylase 3A (KDM3A) is an enzyme that specifically catalyzes the removal of 2 or three methyl groups from lysine 9 of histone 3 (KH3). It belongs to the family of histone demethylases that contain the Jumonji C (JmjC) domain, which means cruciform in Japanese. KDM3A also belongs to a family of hydroxylases that are alpha-ketoglutarate dependent. The systematic evolution of ligands by exponential enrichment (SELEX) is a powerful method for the development of high affinity RNA ligands toward and infinite array of target molecules. SELEX is based upon the generation of a randomized population of RNA or DNA molecules followed by a target molecule that selects high affinity ligands from the randomized population followed by the subsequent amplification of the selected molecules. Hemodynamics and shear forces are associated with pathological changes in the vascular wall and its function, resulting in the focal development of atherosclerosis. Flow complexities that develop in the presence of established plaques create environments favourable to thrombosis formation and potentially plaque rupture leading to stroke. The carotid artery bifurcation is a common site of atherosclerosis development. Recently, the multi-directional nature of shear stress acting on the endothelial layer has been highlighted as a risk factor for atherogenesis, emphasizing the need for accurate measurements of shear stress magnitude as well direction. In the Western Gulf region of the United States cold-tolerant eucalyptus have been explored as pulpwood feedstock. However, non-native plantations may alter understory species diversity, modifying environmental conditions and soil characteristics. Few studies have compared eucalyptus plantations with other ecosystems to understand the impacts of converting these land uses on understory vegetation in the United States. Three plantations were selected: (1) slash pine (Pinus elliottii) established in 2008, (2) slash pine established in 2013, and (3) and Camden white gum (Eucalyptus benthamii) established in 2013. Gastropod immunobiology has benefitted from investigations focused on the planorbid snail Biomphalaria glabrata, intermediate host for the human parasite Schistosoma mansoni. Though such concentrated efforts have elucidated fascinating aspects of invertebrate immunity, they have not provided full knowledge regarding the evolution of immune function among other gastropod species. This dissertation presents the importance of making strategic choices regarding which organisms to select for comparative immunology. Asymmetric cell division (ACD) is important for cell fate and tissue patterning. Many aspects of ACD in plants, however, are still unclear. The process of ACD can be broadly broken down into three phases: (1) cell polarization, (2) division plane establishment and maintenance, and (3) cell division and cytokinesis. Stomatal development in maize has proven to be a useful model for understanding the ACD mechanism. Previous studies have identified several proteins important for all three phases of ACD.
