Human carboxylesterases (CESs) are enzymes that are responsible for the metabolism of many important pharmaceuticals. Although CESs are key players in the hydrolysis of many ester-containing drugs, they remain understudied. Our group hypothesizes that this is primarily because there are few methods capable of reporting activity in live cells. Here, I report a new series of fluorogenic chemical tools to study the CES activity of one of the two major CESs in humans, CES1, in live cells. MCP-Me, MCP-Et, and MCP-iPr utilize the same carbonate group of a previously developed chemical tool in our group, FCP-1, and work to mimic …

Neurodegenerative diseases such as Alzheimer's and Parkinson's are the main causes of age-related dementia. These diseases can be due to neuronal cell death and/or impairment of neurite outgrowth. Giant oyster mushroom (GOM), Pleurotus giganteus, is used as a nootropic to improve cognitive function. GOM can also be used to prevent the onset of dementia. The underlying mechanism behind the medicinal property of GOM is unclear. Previous studies have shown that GOM has a high concentration of uridine. In this study, I examined the effects of uridine on neurite outgrowth in the Neuro-2a (N2a) neuroblastoma cell line. We also examined …

Pollutants, as a result of wastewater treatments, have been shown to have negative impacts on aquatic ecosystems. To better understand the possible consequences caused by effluents on ecosystems, it is important to examine ecotoxicology data. One of the most commonly used species for water quality testing is the fathead minnow, Pimephales promelas. Ecotoxicology can then be taken one step further to understand the effects of pollutants on a molecular level. Previous research had identified effluents as causes for abnormal minnow fin morphology. As one of the major organs, the liver plays vital roles in the homeostasis of an individual. Being able to identify master regulator genes, genes whose product can affect the activation or deactivation of other genes, will enrich our understanding of hepatic function and liver disease. Previously, our lab used genome-wide microarray data to identify several transcription factors that may play a key role in liver expression. Aedes albopictus, Asian tiger mosquitoes, are vectors of a wide number of human viral diseases including, West Nile virus, Dengue virus, Chikungunya virus and (recently) Zika virus. A large body of evidence has suggested that microbiomes of mosquito midguts are closely associated with specific mosquito life processes such as nutrition, reproduction, aggregation and defense against toxins. In this study we characterize the bacterial flora of the midguts of adult female Ae. albopictus collected from woodlots and residential areas in Champaign and Coles Counties of Central Illinois (40 samples in each category). Anthocyanins in the leaves of carnivorous plants are suggested to play a role inprey capture. In this study, we investigated the role of red leaf coloration (an indicator of anthocyanins)on prey capture using Pinguicula planifolia (Chapman’s Butterwort). Overall,red leaves had less prey (i.e., Collembola) than green leaves, suggesting that red colorationdoes not enhance prey capture for Chapman’s Butterwort. However, the frequent presenceof Collembola on leaves suggests that this plant species could be relying on other cues toattract prey (e.g., olfactory cues).

Cyperus fedoniae is described as a new species. It is known from a single collection from the lowlands of southern Venezuela. It is a member of section Incurvi. It is related to C. dichromeniformis and C. inops, from which it can be distinguished by its smaller size and features of spikelets and achenes. MitoNEET was discovered through interactions with a labeled and photoactive derivative of pioglitazone (pio), a drug used to increase peripheral insulin sensitivity. Its unique coordination of a [2Fe-2S] cluster by three cysteine residues (Cys-72, Cys-74, and Cys-83) and one histidine (His-87) gives this cluster both stability and the ability to be donated to acceptor proteins. These qualities allow mitoNEET to participate in a diversity of biological functions. Functions of mitoNEET and the consequences of pioglitazone (pio) treatment in human hepatocellular carcinoma (HepG2) cells cultured in glucose or galactose-based medium were examined by respiration and proliferation studies. Fish larvae develop a fin fold that will later be replaced by the median fins. I hypothesize that fin fold reabsorption is part of the initial patterning of the median fins, and that caspase-3, an apoptosis marker, will be expressed in the fin fold during reabsorption. I analyzed time series of larvae in the first 20-days post hatch (dph) to determine timing of median fin development in a basal bony fish - sturgeon- and in zebrafish, a derived bony fish. Fish are known to use vision in many essential behaviors, including foraging, intraspecific communication and predator avoidance. Turbidity is one of the many environmental factors potentially affecting vision quality. I examined the behavior of interacting conspecific fish in varying levels of turbidity to determine how this environmental variable affects vision and behavior. Experiments were designed to observe how longear sunfish, Lepomis megalotis, reacted to visual cues - in this case a conspecific fish in a glass jar - in the presence or absence of turbidity. My hypothesis is that sunfish will display territorial behavior when introduced to another fish …

First discovered in the 1980's, late embryogenesis abundant (LEA) proteins now play an important role in the modern paradigm of anhydrobiosis. These proteins are intrinsically disordered in the hydrated state, and gain more defined secondary structure motifs (e.g. beta-sheets and alpha-helices) during the process of dehydration. This gain in secondary structure likely plays a key role in the ability of LEA proteins to protect the machinery important to life sustaining processes within the cell during severe water stress. In the face of a changing climate, questions regarding sub-lethal effects of elevated habitat temperature on the physiology of ectotherms remain unanswered. In particular, long-term responses of ectotherms to the warming trend in tropical regions are unknown and significantly understudied due primarily to the difficulties in specimen and community traceability. In freshwater lakes employed as cooling reservoirs for power plants, increased physiological stress from high water temperature can lead to an increase in mortality, reduce growth, and potentially alter the community structure of fishes. Throughout this study, we employ this highly tractable system to assess how elevated thermal regimes can …
