Objective The nuclear factor-κB (NF-KB)/RelB transcription factor plays an essential role in development of some dendritic cell (DC) subsets in mice. In this laboratory, immature myeloid DC are produced in vitro in a stroma-dependent murine spleen long-term culture (LTC) system. In LTC, DC differentiate from hematopoietic progenitors maintained within the stromal cell matrix. Expression and function of RelB in development of LTC-DC has been investigated, with a view to assessing the relationship between DC produced in this system and other known subsets of DC. The dendritic cell (DC) population in spleen comprises a mixture of cells including endogenous DC progenitors, DC precursors migrating in from blood and bone marrow, and DC in different states of differentiation and activation. A role for different microenvironments in supporting the dynamic development of murine DC of different types or lineages is considered here. Recent evidence for production of DC dependent on splenic stromal cells is reviewed in the light of evidence that cell production is dependent on cells comprising an endothelial niche in spleen. Background: Many important high throughput projects use in situ hybridization and may require the analysis of images of spatial cross sections of organisms taken with cellular level resolution. Projects creating gene expression atlases at unprecedented scales for the embryonic fruit fly as well as the embryonic and adult mouse already involve the analysis of hundreds of thousands of high resolution experimental images mapping mRNA expression patterns. Challenges include accurate registration of highly deformed tissues, associating cells with known anatomical regions, and identifying groups of genes whose expression is coordinately regulated with respect to both concentration and spatial location. Background
Due to the increased use of genetic modifications in crop improvement, there is a need to develop effective methods for the detection of both known and unknown transgene constructs in plants. We have developed a strategy for detection and characterization of unknown genetic modifications and we present a proof of concept for this method using Arabidopsis thaliana and Oryza sativa (rice). The approach relies on direct hybridization of total genomic DNA to high density microarrays designed to have probes tiled throughout a set of reference sequences.

The conditional expression of hairpin constructs in Drosophila melanogaster has emerged in recent years as a method of choice in functional genomic studies. To date, upstream activating site–driven RNA interference constructs have been inserted into the genome randomly using P-element–mediated transformation, which can result in false negatives due to variable expression. To avoid this problem, we have developed a transgenic RNA interference vector based on the phiC31 site-specific integration method.

The over 300 million ha of public and private rangelands in the United States are characterized by low and variable precipitation, nutrient-poor soils, and high spatial and temporal variability in plant production. This land type has provided a variety of goods and services, with the provisioning of food and fiber dominating through much of the 20th century. More recently, food production from a rangeland-based livestock industry is often pressured for a variety of reasons, including poor economic returns, increased regulations, an aging rural population, and increasingly diverse interests of land owners. The U.S. EPA has identified pathogen indicators as a leading cause of impairments in rivers and streams in the U.S. Elevated levels of bacteria in streams draining the agricultural watersheds cause concern because they indicate the potential presence of pathogenic organisms. Limited understanding of how bacteria survive in the environment and are released from fecal matter and transported along overland flow pathways results in high uncertainty in the design and selection of appropriate best management practices (BMPs) and in the bacterial fate and transport models used to identify sources of pathogens.

A Mini-Fluidics system was designed to facilitate the muscle growth of the Zebra Fish embryos. This experimental device is made of peristaltic pump, inflow/outflow manifolds, fluid storage tank, series of valves and flexible pipes and the main plate with six mini channels. These closed channels provide pathways for an extremely laminar flow. The Zebra Fish embryos are placed in the channels and exposed to the forces present in the fluid.

Many studies have shown that soil disturbance facilitates establishment of invasive, non-native plant species, and a number of mechanisms have been isolated that contribute to the process. To our knowledge no studies have isolated the role of altered soil compaction, a likely correlate of many types of soil disturbance, in facilitating invasion. To address this, we measured the response of seeded non-native and native plant species to four levels of soil compaction in mesocosms placed in an abandoned agricultural field in the Methow Valley, Washington, USA. Whether mucosal immunization is required for optimal protective CD8 T cell memory at mucosal surfaces is controversial. In this study, using an adoptive transfer system, we compare the efficacy of two routes of acute lymphocytic choriomeningitis viral infection on the generation, maintenance, and localization of Ag-specific CD8 T cells in tissues, including the vaginal mucosa. Surprisingly, at day 8, i.p. infection results in higher numbers of Ag-specific CD8 T cells in the vaginal mucosa and iliac lymph node, as well as 2-3x more Ag-specific CD8 T cells that coexpress both IFN-gamma and TNF-alpha in comparison to the intranasal route of …

Concentrations of polybrominated diphenyl ethers (PBDEs), and organochlorine pesticides (OCPs; DDTs, HCHs, CHLs, and HCB) were measured in human breast milk samples collected across Massachusetts, USA, in 2004. Seventeen PBDE congeners were found in the samples, ranging in concentration from 0.06 to 1910 ng g(-1) lipid wt. BDE-47 (2,2',4,4'-tetraBDE), BDE-99 (2,2',4,4',5-pentaBDE), and BDE-100 (2,2',4,4',6-pentaBDE) were the major congeners detected in breast milk samples. Overall mean (+/-SD) concentrations of DDTs, HCHs, CHLs, and HCB were 64.5 +/- 75, 18.9 +/- 19, 32.4 +/- 36, and 2.3 +/- 2.2 ng g(-1) lipid wt, respectively. Recent studies have suggested that the structural Fe(III) within phyllosilicate minerals, including smectite and illite, is an important electron acceptor for Fe(III)-reducing microorganisms in sedimentary environments at moderate temperatures. The reduction of structural Fe(III) by thermophiles, however, has not previously been described. A wide range of thermophilic and hyperthermophilic Archaea and Bacteria from marine and freshwater environments that are known to reduce poorly crystalline Fe(III) oxides were tested for their ability to reduce structural (octahedrally coordinated) Fe(III) in smectite (SWa-1) as the sole electron acceptor. Anthropogenic nitrous pollutant emissions in China significantly increased during the last decades, which contributed to the accelerated nitrogen (N) deposition. In order to characterize spatial pattern of nitrogen deposition, we employed the kriging technique to interpolate sampling data of precipitation chemistry and ambient air concentration from site-network observations over China. The estimation of wet deposition in China was limited to aqueous NO3− and NH4+, while ambient NO2 was the only species involved in the predicted dry deposition fluxes. To obtain wet deposition fluxes, precipitation concentration was multiplied by 20-year mean precipitation amounts with a resolution of 10 × 10 km. …

Long-term management practices are needed to increase dryland C storage and improve soil quality. We evaluated the 21-yr effects of combinations of tillage and cropping sequences on dryland crop biomass (stems + leaves) returned to the soil, residue C, and soil C fractions at the 0- to 20-cm depth in a Dooley sandy loam (fine-loamy, mixed, frigid, Typic Argiborolls) in eastern Montana. While foraging, animals often exploit group members to obtain food. One way to describe this behaviour is with the producer-scrounger (PS) model, where scroungers use social interaction to obtain food discovered by producers. Mexican jays (Aphelocoma ultramarina) are a groupforaging species with a linear dominance hierarchy. We studied interactions in a free-living foraging group to determine (1) if foraging interactions can be explained with the PS model, (2) if these roles are consistent and (3) if dominance or relatedness affects joining frequency. Many secondary compounds are typically present in unpalatable shrubs on arid and semi-arid rangelands. However, the relationship between intake by livestock and concentration of individual chemicals has been examined for very few of these compounds. Four experiments were conducted to examine effects of individual volatile compounds on intake of alfalfa pellets by lambs. Forty-five lambs (9 lambs/treatment) were individually fed alfalfa pellets with eugenol, α-terpineol, terpin-4-ol, or methyl eugenol applied at one of five concentrations. Pleistocene glacial cycles are thought to have played a major role in the diversification of temperate and boreal species of North American birds. Given that coalescence times between sister taxa typically range from 0.1 to 2.0 Myr, it has been assumed that diversification occurred as populations were isolated in refugia over long periods of time, probably spanning one to several full glacial cycles. In contrast, the rapid postglacial range expansions and recolonization of northern latitudes following glacial maxima have received less attention as potential promoters of speciation. The purine salvage pathway of Anopheles gambiae, a mosquito that transmits malaria, has been identified in genome searches on the basis of sequence homology with characterized enzymes. Purine nucleoside phosphorylase (PNP) is a target for the development of therapeutic agents in humans and purine auxotrophs, including malarial parasites. The PNP from Anopheles gambiae (AgPNP) was expressed in Escherichia coli and compared to the PNPs from Homo sapiens (HsPNP) and Plasmodium falciparum (PfPNP). The potential benefits of their use have not been adequately investigated, especially in the developing world.
