Multiphysics modeling plays a crucial role in understanding the complexities of microwave-food interactions, especially in multi-port solid-state microwave systems where microwave parameters can be precisely and dynamically controlled. However, previous models using simplistic or manually measured oven geometries face challenges in accurately simulating the microwave heating process. This study first developed a robust 3-D scanning approach to capture precise geometric details of the oven cavity, incorporating them into multiphysics modeling for solid-state microwave heating. Furthermore, a quantitative validation approach was also developed to characterize modeling accuracy against experimental results. With an expected increase in population by 2050, the demand for more animal protein will increase. To improve the sustainability of US cattle production, producers and researchers have historically focused on improving the feed efficiency of steers, producing a more marketable beef product. However, without a successful pregnancy, there would be no marketable animal to feed. The objective of this study was to examine the physiological changes that occur during pregnancy and their impact on the rumen microbial communities and the correlation to feed efficiency status in Angus heifers. The Caddo Madtom, Noturus taylori, is a small catfish endemic to the Ouachita Mountain ecoregion in Arkansas, with habitat altered by land use practices and reservoir dams. We examined aspects of distribution, abundance, habitat, and life history of N. taylori during seasonal sampling from winter 2016 through fall 2017. Our sampling data were concordant with previous studies that suggested N. taylori is more widespread and has higher catch per unit effort in the Caddo River drainage when compared to the upper Ouachita River drainage. We did not detect N. taylori in the Little Missouri River drainage, where it is …

Physical activity has been shown to be positively associated with improved motor skill and cognitive development in young children. Intervention designs are often constructed without focus on interrelatedness of the development of motor and cognitive self-regulatory skills. Purpose: To investigate the impact of a movement-based curriculum designed to integrate both motor skill and cognitive development in preschool children. Methods: Preschool-aged children [n=34; intervention group (n=28); control group (n=6)] participated in the study. Campylobacteriosis, the foodborne disease caused by Campylobacter spp., infects one out of 10 individuals every year. C. jejuni accounts for 90% of these infections resulting in numerous postinfectious disorders including the development of colorectal cancer, Guillain-Barré syndrome, irritable bowel syndrome, and reactive arthritis. Despite its large impact on human health, the host immune response to the bacterium is largely uncharacterized. Chapter two of this dissertation addresses the development of neutrophil extracellular traps (NETs) within human and ferret campylobacteriosis. Harmful Algal Blooms (HABs) are algae undergoing prolific, unregulated growth. A well-documented HAB taxa is the cyanobacterium Microcystis spp., which induces anthropogenic, ecological, and economic consequences due to the production of toxins and biomass which results in lake hypoxia. Microcystis spp. blooms are globally distributed in freshwater systems, with climate change and the aquatic continuum serving to further exacerbate bloom distribution, duration, and frequency. Thus, there is a need to elucidate the factors driving the ecological success of Microcystis spp., and the ecological “failures” of their competitors, such as diatoms. Biomanufacturing propels the bioeconomy. Accelerating bioeconomic growth thus requires the expedited development of biomanufacturing processes that can expand the current bioproduct portfolio. Lysate-based cell-free systems provide unique advantages for simplified metabolic pathway construction. Their central metabolic pathways and transcriptional-translational (TX-TL) machineries are free from genome regulation and are amenable to direct manipulation, enabling the streamlined construction of biomanufacturing processes. Functional and quantitative metagenomic profiling remains challenging and limits our understanding of host-microbe interactions. This body of work aims to mediate these challenges by using a novel quantitative reduced representation sequencing strategy (OmeSeq-qRRS), development of a fully automated software for quantitative metagenomic/microbiome profiling (Qmatey: quantitative metagenomic alignment and taxonomic identification using exact-matching) and implementing these tools for understanding plant-microbe-pathogen interactions in maize and sweetpotato. The next generation sequencing-based OmeSeq-qRRS leverages the strengths of shotgun whole genome sequencing and costs lower that the more affordable amplicon sequencing method. Soybean cyst nematode (SCN) is the most devastating and economically important pathogen of soybean across the United States and globally. The current primary control for SCN is with the use of resistant cultivars, however, SCN populations are shifting to overcome most common sources of soybean resistance. This research was conducted to identify novel sources of genetic resistance to SCN. First, I functionally characterized two protein kinase hub genes whose function are predicted to be essential for mediating soybean susceptibility to SCN. Plant microbiomes are assembled and modified through a complex milieu of biotic and abiotic factors. Despite dynamic and fluctuating contributing variables, specific host and microbial mechanisms are likely important mediators of interactions. We combine information from large-scale datasets across diverse plant hosts with experimental genetic manipulation assays in model Arabidopsis seedlings to converge on a conserved role for metabolite production and transport in mediating host-microbe interactions. Applying a diverse set of analytical tools to microbial communities at various complexity levels will advance our knowledge of the scalability of observed phenotypes and, ultimately, help to decipher the mechanisms dictating plant–microbe interactions …

Chytridiomycosis, a cutaneous fungal disease caused by two related fungi, Batrachochytrium dendrobatidis (Bd) and Batrachochytrium salamandrivorans (Bsal), is a major cause of amphibian population declines worldwide. Bd has a broad distribution, covering every continent except for Antarctica. Meanwhile, Bsal is currently restricted to Europe and Asia. Treatment options for both Bd and Bsal remain a major area of concern for wild populations. Mixed pastures are known to produce greater biomass yields and higher nutritive value than monocultures. Intercropping biologically N fixating legumes, along with the use of intercropping warm-season annual grasses, such as crabgrass [Digitaria ciliaris Retz.], provides the potential to aid tall fescue [Schedonorus arundinaceus (Schreb.) Zoysiagrasses (Zoysia spp. Willd.) are commonly used on golf course fairways and tees in addition to residential and commercial lawns due to lower input requirements relative to bermudagrass (Cynodon spp.). This has led to increased interest in using zoysiagrass for golf course putting greens; however, zoysiagrass establishment from sprigs is prolonged compared to bermudagrass. A series of experiments were conducted in glasshouses in Knoxville, TN in 2022 to evaluate the effects of environmental conditions and management practices on the establishment of Prizm zoysiagrass from sprigs. The first thesis chapter employs two single-stage data envelopment analysis (DEA) models under varying returns to scale assumptions to provide technical efficiency estimates for 26 dairy farms in Tennessee, Kentucky, and North Carolina participating in the UT Dairy Gauge program. These analyses reveal how efficiently farms utilize inputs (cows, feed, labor, forage) to generate farm income relative to other sample farms. Under the assumption of constant returns to scale (CRS) 7 farms are estimated to be technically efficient, while 13 farms display comparative efficiency under an assumption of variable returns to scale (VRS). Integral membrane proteins are prolific targets for the design, development, and delivery of pharmaceuticals. In fact, over 60% of all currently available drugs target these proteins to accomplish their therapeutic effect. However, integral membrane proteins remain the least characterized class of all proteins, accounting for only ~2% of all solved protein structures. One of the primary reasons for this low number of solved protein structures is that many membrane proteins lose their native conformation when extracted using conventional methods (e.g. detergents), convoluting accurate structure determination. The Forest Inventory and Analysis (FIA) conducts Timber Products Output (TPO) studies to assess timber resource use across the United States. The TPO studies gather information through surveys of primary wood processing facilities to quantify the utilization of roundwood by geographic location, tree species, and timber product type. However, not all responding mills provide information on tree species utilized. As a result, a portion of the reported volume falls into the “unknown/unclassified” softwood or hardwood category, which account for up to 74% of total hardwood/softwood southern volumes in some cases. For several years, conversations about the absence of racial and ethnic diversity in ecology and evolutionary biology (EEB) and the conservation movement have been growing in scope. Critics argue that the overwhelmingly white demography of EEB departments and conservation organizations deprive both of a necessary diversity of perspective and, more importantly, deprive people of color and other minoritized groups of a voice in the study of and advocacy for their lived environments. Food insecurity affected 10.2% of United States households in 2021. While food pantries offer free food to households in need, prior research suggests that stigma may prevent households from utilizing this valuable resource. The purpose of this study is to investigate a potential additional barrier to food pantry used posed by the near-to best if used by dates (BUBD) commonly found on foods donated to foodbanks. Phytophthora nicotianae, a hemibiotrophic oomycete, is the causal agent of disease of over 300 hosts, notably tobacco, tomato, citrus, strawberry, and ornamentals. Black shank of tobacco is a devastating disease that can result in up to 100% yield loss if left untreated, and buckeye rot of tomato compromises the marketability and yield of fruit impacted by P. nicotianae. Plants are stationary organisms that are charged with overcoming a multitude of biotic and abiotic stresses. The eukaryotic translation initiation factor 2 (eIF2) is responsible for charging the P-site of the forming 80S ribosome with the initiator methionyl-tRNA. Thus, eIF2 is a protein of utmost consequence in the growth and development of organisms. The eIF2 protein is also a mediator of global translational regulation in the eukaryotic integrated stress response (ISR), where the α-subunit is phosphorylated by protein kinases, such as the kinase GCN2. Poultry houses might have high levels of airborne pathogens, including Escherichia coli (E. coli) and avian influenza (AI), which may be transmitted through the air and pose risks of infection.
