Alzheimer’s disease is a progressive brain disorder affecting more than 10 percent of Americans over the age of 65 (1). This disease destroys memory and thinking skills and is a leading cause of dementia. Research in our lab has predicted hundreds of bacterial genes that influence nine different traits in the fruit fly, Drosophila melanogaster. As a practical alternative to creating site-directed mutants for each of the predicted genes, we created an arrayed transposon insertion library using a strain of Acetobacter fabarum DsW_054 isolated from fruit flies. Creation of the Acetobacter fabarum DsW_054 gene knock-out library was done through random transposon insertion, combinatorial mapping and Illumina sequencing. Successful mapping of transposon insertion was achieved for 6418 mutants with hits within 63% of annotated genes within Acetobacter fabarum DsW_054. Insertion sites were verified in …

Systemic Lupus Erythematosus (SLE) is an autoimmune disease affecting 20 to 250 individuals per 100,000 worldwide. Symptomology includes dermatological manifestations such as discoid lesions, acute cutaneous rashes, and oral and nasal ulcers, along with musculoskeletal, pulmonary, and renal complications. Abnormal T and B lymphocyte function and apoptosis, immune complex clearance, complement function, and nucleosome processing are typical of disease pathophysiology. SLE is the result of both environmental and genetic factors, which together create the conditions leading to disease onset and progression. Phosphate is an essential compound for life. Escherichia coli employs a signal transduction pathway that controls the expression of genes that are required for the high-affinity acquisition of phosphate and the utilization of alternate sources of phosphorous. These genes are only expressed when environmental phosphate is limiting. The seven genes for this signaling pathway encode the two-component regulatory proteins PhoB and PhoR, as well as the high-affinity phosphate transporter PstSCAB and an auxiliary protein called PhoU. As the sensor kinase PhoR has no periplasmic sensory domain, the mechanism by which these cells sense environmental phosphate is not known. This paper …

Human-bear interactions are an important consideration of bear biology, as interactions can lead to destruction of property as well as injury or death for both human and bear. Successful analysis of why these interactions occur can lead to appropriate preventative measures and mitigation of further conflict. Bryce Canyon National Park (BRCA) is comprised of relatively poor bear habitat, but a black bear population exists on the Paunsaugunt Plateau, on which the park occupies the eastern edge. Common oat (Avena) has held a significant place within the global crop community for centuries; although its cultivation has decreased over the past century, its nutritional benefits have recently garnered increased interest for human consumption. No published reference sequences are available for any of the three oat subgenomes. Here we report a quality sequence assembly, annotation and hybrid optical map of the A-genome diploid Avena atlantica Baum and Fedak. The assembly is composed of a total of 3,417 contigs with an N50 of 11.86 Mb and an estimated completeness of 97.6%. This genome sequence will be a valuable research tool …

An important obstacle to a functional cure for HIV/AIDS is the persistence of viral reservoirs found throughout the body in various cells and tissues. Reservoirs can be latently infected cells, or in the case of follicular dendritic cells (FDC), non-infected cells that trap infectious virus on their surface through immune complexes (HIV-IC). Although several strategies have been employed to target and eliminate viral reservoirs, they are short-lived and ineffective. BACKGROUND: Chronic ankle instability (CAI) patients have varying levels of mechanical and sensorimotor impairments that may lead to disparate functional movement patterns. Current literature on landing biomechanics in a CAI population, however, considers all patients as a homogeneous group. Introduction: Irish dance has evolved to become more athletically demanding, thus making the art form very hard on dancers' bodies. Irish dancers must land from difficult moves without letting their knees bend or heels touch the ground, causing large amounts of force to be absorbed by the body. Past studies have found dancers landing with a range of 4.5–6 times body weight, potentially causing high amounts of overuse injury. Yelling in society is a common occurrence. Parents yell at their children, bosses yell at their employees and coaches yell at their athletes. Yet, in many cases the yelling coach exhibits unkind, loud in nature, and very personal statements. The purpose of this study was to better understand former athlete’s perspectives regarding their thoughts and experiences of their coaches yelling at them. For this study yelling will imply saying loud, unkind, personal comments towards the athlete. For this study 124 former athletes were surveyed regarding their experiences with a yelling coach. This dissertation explores the differential effects of an ungulate community (cattle, mule deer, and American elk) on aspen (Populus tremuloides) regeneration pre- and post-fire disturbance. It's first chapter examines the differential effects of cattle, mule deer, and American elk on aspen regeneration across variable topography, climate and forest type; aspen dominant to conifer dominant subalpine forests. We found that each ungulate species, if sufficiently numerous, can cause aspen regeneration failure. Also, high elevation, southern aspect and high winter snow pack increase resilience to ungulate herbivory. Chapter two compares the efficacy of four methods of estimating ungulate impact on …

Synapses can be altered due to experiences in a process called synaptic plasticity, which causes memory formations. Synapses can be strengthened through methods known as long-term potentiation (LTP) or weakened through long-term depression (LTD). Stresses can cause changes by altering synapses through either LTP or LTD. Rats were used to study the effects of post-traumatic stress disorder (PTSD)-like symptoms and a prophylactic treatment using pharmaceuticals. The kit fox (Vulpes macrotis) is a species of conservation concern in western North America. Recent methods for monitoring populations of kit fox include using lures and remote cameras in an occupancy-modeling framework and habitat modeling to predict areas of occupancy. In chapter one, we tested the optimal lure and movement procedure for scent stations to maximize visits and detection of foxes, thereby improving estimates of occupancy. In species that require parental care, each parent can either care for their offspring or leave them in the care of the other parent. For each parent this creates three possible parental care strategies: biparental care, uniparental (male or female) care, and uniparental desertion by either the male or female. The burying beetle, Nicrophorus orbicollis, typically exhibits biparental care of offspring, and thus provides a unique system that allows us to compare the fitness benefits of these parental care strategies in an unconfounded way. Physical activity impacts health and disease in multiple body tissues including the intervertebral discs. Fluid flow within the disc is an indicator of disc health that can be observed using diffusion weighted magnetic resonance imaging. We monitored activity levels of 26 participants, age 35 to 55 yrs, using Actigraph accelerometers for four days to evaluate vigorous-intensity activity, moderate to vigorous-intensity activity, and sedentary time. Participants underwent structural and diffusion weighted magnetic resonance imaging to evaluate intervertebral disc health and fluid flow. Successful management of bighorn sheep depends on understanding the mechanisms responsible for population growth or decline, habitat selection, and utilization distribution after translocations. We studied a declining population of desert bighorn sheep in the North San Rafael Swell, Utah to determine birthdates of neonates, demographics, limiting factors, population size, probable cause of death, production, and survival. We documented 19 mortalities attributed to a variety of causes including cougar predation (n = 10, 53%), bluetongue virus (n = 2, 11%), reproductive complications (n = 2, 11%), hunter harvest (n = 1, 5%), and unknown (n = 4, 21%). The men's steeplechase event requires participants to jump over thirty-five 0.914-meter-tall obstacles, 4 rigid barriers and 1 fixed barrier followed by a 3.66-meter-long water pit per lap, over a 3000-meter distance. This study investigated the effect of increasing running velocity, through a range of 5.33 m/s to 6.66 m/s, on takeoff and landing ground reaction forces, for males during steeplechase hurdling using a force plate embedded under a track surface. Teacher Perceptions Of School Breakfast Programs In Utah, Emily Breanne Krueger

The objective of this study was to identify differences in teacher perceptions of benefits, challenges, and preferences to different School Breakfast Program (SBP) service models. A survey instrument was developed, pilot tested, and then distributed electronically to K-12 teachers throughout the state of Utah, who were part of the Utah Education Association. Demographics and factors influencing SBP models in Utah were gathered. Frequencies and ANOVA tests were performed and a significance level of <0.01 was chosen to protect for multiple comparisons. Results indicated that traditional breakfast was the most preferred model with a mean score of 2.80 and breakfast in the classroom was the least preferred model by teachers with a mean of -1.32. Children not going hungry was identified as the greatest benefit (95.4%, n=352) to SBP and food waste was identified as the greatest challenge (45.8%, n=168). This study concluded that increased awareness and education amongst teachers regarding different models of SBP service could lead to increased efficiency, increased participation in SBP, decreased costs, decreased food waste, and increased academic performance and health benefits for students.

Introduction (Context): The purpose of this study was to measure and correlate pectoralis minor length (PML) and acromiohumeral distance (AHD) in male and female collegiate swimmers, overhead athletes, and a control group. Methods: Participants underwent assessment of pain related to impingement syndrome with special tests (painful arc, external rotation resistance, empty can, and Neer's impingement test), as well as range of motion, measurement of PML, and measurement of subacromial space via ultrasound. A hallmark of type 2 diabetes (T2D) is β-cell dysfunction and the eventual loss of functional β-cell mass. Therefore, mechanisms that improve or preserve β-cell function could be used to improve the quality of life of individuals with T2D. Studies have shown that monomeric, oligomeric and polymeric cocoa flavanols have different effects on obesity, insulin resistance and glucose tolerance. We hypothesized that these cocoa flavanols may have beneficial effects on β-cell function. The alpaca is a small South American camelid (SAC) that is an important production animal in Peru, especially among the highly impoverished communities of the high Andes, and raised for its fiber and meat. Alpacas are highly reliant on the microbes within their digestive tracts to digest the plant material they consume; volatile fatty acids (VFAs) are released as a byproduct of this microbial fermentation and used as a major source of energy by the alpaca.
