Post-treatment discomfort following endodontic procedures can be caused by inflammation due to injudicious use of intracanal medicaments. Camphorated paramonochlorophenol, metacresyl acetate, and prednisolone in combination is advocated to reduce the inflammatory potential of the germicide and thus reduce the occurrence of post-operative pain. It is imperative that a medicament retain antimicrobial activity while reducing inflammatory potential.
This investigation was undertaken toward determining on an in vitro quantitative bactericidal versus bacteriostatic basis by use of the broth dilution test tube method whether camphorated paramonochlorophenol, metacresyl acetate, and prednisolone in combination retains antimicrobial activity comparable to selected other commonly used nonspecific germicides. …

The prime motivating factor in the present study was the desire to show significant differences in radio-sensitivities within six strains of Trypanosoma avium. A seventh strain was later added for the last two irradiation treatments. Gross morphology and differences in growth curves associated with varying temperatures suggested that the strains being cultured were not the same. With these differences in mind, the strains were subjected to different dosage levels of gamma irradiation.

The purpose of this study was to investigate the ultrastructure of Tilletia caries teliospores. Specimens were prepared for viewing in the electron microscope by freeze-etching and thin-sectioning techniques. Thin-sectioning techniques gave excellent results with the teliospore walls and the lipid bodies within the teliospore but failed to adequately fix the membranes and internal organelle systems. Micrographs of Tilletia caries teliospores prepared using thin-sectioning show three major wall layers and two partition wall layers and abundant lipid bodies. The freeze-etch method of spore preparation proved superior to the thin-sectioning used in this study to reveal the internal organelle systems of the …

The purpose of this investigation was to evaluate the possible taxonomic significance of the "rodlet" patterns and spore wall configurations of conidia from monosporous cultures of Penicillium digitatum Saccardo and Aspergillus nidulans Eidam var. echinulatus Fennell & Raper. Freeze-etching the conidiospores from single spore isolates of these fungi revealed that spore wall patterns and configurations vary much too greatly to be of taxonomic importance on the strain and subspecies level. Environmental factors and genetic influences are discussed as possible mechanisms causing the variations of the spore wall configurations. Basidiospores of the Basidiomycete Psilocybe mutans have been found to germinate only in the presence of water extracts of animal dung. The chemical nature of the factor present in the dung that induces germination is not known, nor has it ever been isolated· Experiments showed that extracts of bile salts cause the basidiospores to germinate, indicating that since bile salts are found in dung, they may be the factor which stimulates the spores to germinate. Investigations of the properties of the factor in dung show that the factor has solubility properties that are similar to those of the bile acids. …

A bacteriological procedure for the removal of the intestinal flora of Triatominae is described. Immersion of Triatoma protracta adults and fifth instar nymphs in 5% aqueous formaldehyde for three hours was sufficient to free the integument of these insects of viable bacteria without penetration into the alimentary tract.
Twenty-two adults and 17 fifth instar nymphs of Triatoma protracta protracta were collected from Neotoma lodges in southern California. Eleven of the 17 nymphs molted into adults in the laboratory. With the use of a 2% peracetic acid sterilized custom built plastic germ-free isolator the Triatoma were dissected and various sections of …

A Comparative Survey Of Soil Microfungi From Three Vegetation Types In Cedar Valley, Utah, Brent T. Wahlquist

Microfungi were isolated from soil samples by the soil plate method. Soil samples were collected at depths of 0-1", 1-4", and 4-8" both between and beneath the canopy of the dominant vegetation in stands of greasewood, sagebrush, and pinyon-juniper. Salinity, pH, and percent organic matter were measured for each soil sample. A total of 148 strains of fungi representing 141 species and 46 genera were isolated and identified from these alkaline soils, Few strains, including some Aspergilli, occurred only in the more saline greasewood soils, while many strains, including several Penicillia, occurred in both sagebrush and piny-on-juniper soils but were …

Transferable Drug Resistance Among Enterobacteriaceae Isolated From Human Urinary Tract Infections, Donna G. Sieckmann, Norman D. Reed, Carl E. Georgi

Fifteen sulfonamide-resistant cultures isolated from urinary tract infections in eastern Nebraska were screened for transferable drug resistance by three methods. Seven of the 15 resistant cultures could transfer resistance of varying levels to two or more chemotherapeutic agents. Transfer of drug resistance occurred without accompanying transfer of chromosomal traits and required cell to cell contact. In mixed culture, the number of drug-resistant recipients increased exponentially, reaching a plateau 2 hr after mixing. Spontaneous or artificial elimination of resistance was found to be a rare event. In addition, several drug-sensitive isolates from urinary tract infections were shown to be competent recipients …

A study was conducted in Rich County, Utah to determine the extent of avian tuberculosis in livestock, poultry and wild birds. Sensitivity was elicited in cattle, swine and chickens by tuberculin testing.
The Mycobacterium avium organism was obtained from tissues and specimens of cattle, swine, goats, chickens, magpies and sparrows.
Methods of eradication of avian tuberculosis and the relationship of avian tuberculosis to mammalian tuberculosis are discussed.

Aquatic fungi in-Powell's Slough were.studied in 1967 & 1968. Collections were made and water temperature, oxygen content & pH values were measured at weekly intervals. For collections, wire baskets containing substrata were submerged in water for one month. After collecting, substrata were brought to the laboratory. Identification was based on the keys of Sparrow, 1960. Aquatic fungi identified included seven orders, nine families, fifteen genera, and thirty species, from thirty-three collections of fifteen types of substrata. The rate of protein and ribonucleic acid (RNA) synthesis was examined during the outgrowth of spores of Bacillus cereus T in a chemically defined medium. RNA synthesis started 2.5 min after the initiation of germination, and protein synthesis after 4 min. Addition of a complete amino acid supplement and uracil supported high rates of RNA and protein synthesis throughout outgrowth. To determine the relationship between the rate of protein (k) and RNA synthesis, the kinetics of formation of various classes of RNA were followed during outgrowth. Complementarity studies were conducted between bacteriophage P4/2 DNA and the DNAs of its naturally lysogenic host Salmonella potsdam strain 4 (SP4) and its sensitive host Salmonella potsdam strain 2 (SP2). Calf thymus DNA was used as a control.
Nitrocellulose membranes were used to adsorb varying concentrations of unlabeled DNA, using P4/2 DNA, SP4 DNA, SP2 DNA, and calf thymus DNA. A constant amount of P32 -labeled P4/2 DNA was added to each membrane and the hybrids were allowed to form. The non-lysogenic strain and several lysogenic strains of S. potsdam were tested biochemically and antigenically to detect any possible phage related property.
Fermentation reactions and citrate utilization tests were identical for all strains.
Tubes agglutination tests showed the non-lysogenic and lysogenized strains of SP2-S to be similar, and different from the naturally lysogenic strains SP3, SP4, SP9, and SP10. Absorption tests confirmed the SP2-s strains, either non-lysogenic or lysogenized, to possess an additional antigen SP3 and SP4 did not possess. SP9 and SP10 may contain small amounts of this antigen. From variation may explain the antigenic difference between these strains. …

Addition of KH2P04 salts to Fraser’s Medium produced higher titers of Salmonella potsdam bacteriophage P3/2, but it failed to maintain stable titers under normal cold storage conditions. The addition of ethylenediaminetetraacetate (EDTA) to the medium also produced high titers and helped to maintain phage viability indefinitely in storage. The presence of KH2P04 with the EDTA did not increase the titers nor better the storage properties and was discontinued in favor of EDTA alone. This provided for the accumulation of the necessary amounts of P3/2 for DNA studies.

Cercariac of Schistosoma mansoni were studied for glycogen utilization, uptake of glucose from solution, lactic acid production, and respiration.
Using the anthrone method of determination of carbohydrate, the amount of glycogen used by a single cercaria in 4 hours is determined as 43.2 x 10-4 μg ± 6.6 x 10-4 μg.
Glucose was not observed to have been used from solution under the conditions of the experiment, even in the presence of trehalose. The determinations were made by the Glucostat method.

A mutant of Salmonella potsdam SP2 which had been grown in mixed culture with either of three F+ strains of Escherichia coli was unable to serve as a donor of chromosomal genes when mixed with another mutant of S. potsdam. Therefore, there was no evidence of F transfer in these crosses.
To promote the formation of conjugal pairs, the filter mating technique of Matney and Achenbach was used. With this method, no recombinants were detected when six different Hfr strains of E. coli were mated with various mutants of SP2. Hfr K1 of Salmonella typhimurium transmitted loci for the …

The purpose of this study was to evaluate the ultrastructural aspects of the host-pathogen relationship between the fungus Sphaerotheca pannosa (Wallr.) Lev. and a susceptible horticultural variety of floribunda rose "Eutin". Attention was given to the penetration of hyphal tubes, and their subsequent development into: haustorial elements in epidermal cells of rose. In addition, changes in the morphology of infected epidermal cells were described. Thin sections of rose leaves infected with S. pannosa were examined in a Hitachi electron microscope, model HS-7, after glutaraldehyde-acrolein fixation, OsO4-uranyl acetate staining, and embedding in Epon plastic. The microflora of Cane Branch of Beaver Creek in McCreary County, Kentucky, which drains an area that was strip-mined between 1955 and 1959, was studied and compared with that of Helton Branch which drains a comparable area where there has been no mining. Differences include: the establishment of Ferrcbacillus ferrooxidans, for which procedures were developed for direct colony isolation from the stream; fewer saprophytic bacteria; more numerous and more diversified filamentous and unicellular fungi; and characteristic differences in algal flora. Representatives of 42 genera of filamentous fungi were identified. Of these, 21 were isolated only from Cane Branch. Representatives of …
