Group B Streptococcus (GBS) can be found in the vaginal and genitourinary tract of females, as well as the genitourinary tract of males, where it behaves as a commensal organism. However, as an opportunistic pathogen, GBS has the capability to infect the immunocompromised, making it a major threat to neonates and fetuses. The pathogen can be passed from mother to baby either in utero or during birth. We developed High Resolution Melt Analysis (HERMA) as a rapid and reliable molecular diagnostic assay for the detection and identification of the main bacterial species recovered from the blood and lesions of bacterial chondronecrosis with osteomyelitis (BCO) from the lame broilers at the University of Arkansas poultry research farm. Also, the present study confirms that raising young birds on suspended wire flooring has been proved to successfully induce lameness attributable to BCO with birds being more susceptible to bacteraemia than those that were raised on litter flooring. The productivity and compounds of diatoms, especially oils, support aquatic food chains and human medical and industrial needs. The urinary tract contains a community of bacteria called the urinary microbiota (urobiota) thatmay be relevant to health; the genomic component of the urobiota is the urinary microbiome (urobiome). Urinary bacteria have been associated with both asymptomatic states and disease conditions, such as urinary tract infection (UTI), overactive bladder (OAB), and urge urinary incontinence (UUI). In the United States, municipal solid waste (MSW) landfills remain a potential mining source of recoverable materials, including but not limited to critical, precious, and rare earth metals found in electronic waste. The hemlock woolly adelgid (HWA), Adelges tsugae Annand (Hemiptera: Adelgidae), is an invasive insect that is causing mortality of eastern hemlock trees, Tsuga canadensis, and Carolina hemlock trees, Tsuga caroliniana, across the eastern United States. To protect these ecologically important tree species, a neonicotinoid insecticide, imidacloprid, is commonly used. Imidacloprid is an effective treatment and can remain effective against HWA for four to six years but long-term (≥ one year after application) non-target effects of imidacloprid on forest ecosystems are not well-studied. These bacteria are of particular concern due to their high survivability, low infectious dose, and high adaptability. Cases of shigellosis from Shigella sonnei are becoming a more prevalent issue in the U.S. as the bacteria continues to develop higher resistance to today’s strongest antibiotics. Although molecular approaches can identify members of microbial communities in the environment, genomic information does not necessarily correlate with environmental phenotype. Understanding functional roles can be done by cultivating representative species, yet the culturablility of bacteria from caves remains low, at 0.02%, limiting our understanding of microbial community interactions and processes. We have investigated several factors influencing culturability of bacteria from a single sample location in Maxwelton Sink Cave, WV, USA. Extended incubation of inoculated plates showed a significant increase in colony counts from two to four weeks, indicating that extended incubations increase culturability. Naturally found in soil and water environments arsenic is toxic to many organisms, carcinogenic to humans, and poses a significant public health risk. Yet communities of bacteria found thriving in arsenic ridden environments have evolved mechanisms to tolerate and exploit both oxidation states of this heavy metal (arsenite and arsenate). As the site of an old arsenic mine, Black Mountain Open Space Park in San Diego, California has yielded concentrations of arsenic in the soil between 111-14,800 ppm. In an effort to better understand the role that various microbes may play in toluene production, bacteria from a toluene-producing enrichment culture derived from contaminated groundwater at a Superfund site were cultivated and isolated on low nutrient solid media. A total of 14 solid medium formulations containing varying pH ranges, carbon sources, solidifying agents, and incubation gas headspaces were used to obtain 278 isolates in pure culture. Isolated bacteria, identified using partial 16S rRNA gene sequences, were most closely related with the genera Anoxybacillus, Azospira, Bacillus, Bradyrhizobium, Cellulosimicrobium, Micrococcus, and Propionicimonas.

It integrates several abiotic features and leads to strong, repeatable patterns of community structure, including changes in the abundance and richness of numerous taxa. However, the influence of elevational gradients on microbes is understudied relative to plants and animals. Despite efforts to decrease the global health burden of malaria, infections with Plasmodium species continue to cause over 200 million episodes of malaria each year which resulted in 405,000 deaths in 2018 [1]. One complication of malaria is increased susceptibility to invasive bacterial infections. Plasmodium infections impair host immunity to non-Typhoid Salmonella (NTS) through activities of heme oxygenase I (HO-I) )-induced release of immature granulocytes and myeloid cell-derived IL-10. The Atlantic horseshoe crab (Limulus polyphemus) is a common marine aquarium species and model organism for research. There is potential monetary and conservation value in developing a stable captive population of horseshoe crabs, however, one major impediment to achieving captivity is a lack of knowledge regarding captive diseases. The bacterial microbiome is an essential component of many corals, although knowledge of the microbiomes in scleractinian corals far exceeds that for octocorals. This study characterized the bacterial communities present in shallow water Caribbean gorgonian octocorals over time and space, in addition to determining the bacterial assemblages in gorgonians exposed to environmental perturbations. We found that seven shallow water Caribbean gorgonian species maintained distinct microbiomes and predominantly harbored two bacterial genera, Mycoplasma and Endozoicomonas. Over two million tons of feather waste is generated annually by the poultry industry, the majority of which goes into landfills due to the difficulty of degrading its major component keratin. Although a portion of feather waste is eliminated via incineration or chemical treatment, the use of Feather Degrading Bacteria (FDB) has been proposed as a cheap and eco-friendly alternative. Recent evidence has suggested that the presence of light (and in conjuction, periphytic algae) stimulates the growth and production of bacteria on decaying plant litter. However, the current method of measuring bacterial biomass, flow cytometry, drastically underestimates the bacterial biomass levels associated with litter samples. To determine and correct for this underestimation, the present study determined conversion factors using direct counting methods via epifluorescence microscopy. Infection with Y. pestis typically begins with deposition of Y. pestis into the dermis (bubonic plague) or respiratory tract (pneumonic plague). The gut microbiome plays an essential role in the overall health of organisms. However, the presence and abundance of these microbes may be altered by environmental factors, such as exposure to pesticides. The goals of this project were to understand 1) the prevalence of pesticide residues in honey bees in eastern Washington, and 2) the impact of pesticides to the honey bee gut microbiome. Rothia mucilaginosa has been found at high abundance on oral leukoplakia (OLK). The ability of clinical isolates to produce acetaldehyde (ACH) from ethanol has not been investigated. The objective of the current study was to determine the capacity of R. mucilaginosa isolates recovered from OLK to generate ACH. Analysis of R. mucilaginosa genomes (n = 70) shows that this species does not normally encode acetaldehyde dehydrogenase (ALDH) required for detoxification of ACH. Algal blooms are growing rampantly in lacustrine systems due to an increase of phosphorus, a nutrient for algae. Phosphorus is being released into the environment causing overpopulation and eutrophication which damages the ecosystem. The availability of phosphorus is affected by the solubility of iron which is regulated through iron reducing and iron oxidizing bacteria. Phosphorus adheres to insoluble Fe(III), which prohibits algae from utilizing it, while phosphorus does not attach to soluble Fe(II) and therefore it remains available to algae. The rise of infections caused by antibiotic resistant bacteria, compounded by a reduction in antibiotic discovery and development, jeopardizes human health. Historically, antibiotics derive from secondary metabolites produced by soil microbes in pure culture, but recent genetic evidence suggests that microbes can produce more secondary metabolites than are currently observed. The modified crowded plate technique directly identifies antibiotic-producing soil microbes that were co-plated with a target pathogen. Here, this technique was refined by testing the effect of a D-alanine auxotrophic target pathogen rather than a prototrophic pathogen as well as investigating conditions most conducive to antibiotic production. Part 1 of this experiment was to explore the bacterial types within the surface water on the campus of Gardner-Webb University, located in Boiling Springs, NC. Two water samples were collected from four locations on campus, including the boiling spring, Lake Hollifield, and an adjacent creek. Using the Zymo Research “Quick-DNA Fungal/Bacterial Miniprep Kit,” the bacterial DNA within these samples was isolated and sent to Psomagen, Inc. for analysis.
