Many chloroviruses replicate in Chlorella variabilis algal strains that are ex-endosymbionts isolated from the protozoan Paramecium bursaria, including the NC64A and Syngen 2-3 strains. We noticed that indigenous water samples produced a higher number of plaque-forming viruses on C. variabilis Syngen 2-3 lawns than on C. variabilis NC64A lawns. These observed differences led to the discovery of viruses that replicate exclusively in Syngen 2-3 cells, named Only Syngen (OSy) viruses. As one of Earth’s most valuable pollinators, bees provide important pollination services to wild plant species and crops alike (Winfree 2010). But, in recent years, bee populations appear to be declining due to a variety of anthropogenic drivers (Potts et al. 2010). Quantifying the extent of bee population decline is difficult because there is currently a lack of a standardized protocol for how best to survey bee populations (Tepedino & Portman 2021). Influenza C virus (ICV) is increasingly associated with community-acquired pneumonia (CAP) in children and its disease severity is worse than the influenza B virus, but similar to influenza A virus associated CAP. Despite the ubiquitous infection landscape of ICV in humans, little is known about its replication and pathobiology in animals. The goal of this study was to understand the replication kinetics, tissue tropism, and pathogenesis of human ICV (huICV) in comparison to the swine influenza D virus (swIDV) in guinea pigs. Current methods of vaccination against swine Influenza A Virus (IAV-S) in pigs are infrequently updated, induce strain-specific responses, and have a limited duration of protection. Here, we characterize the onset and duration of adaptive immune responses after vaccination with an adenoviral-vectored Epigraph vaccine. In this longitudinal study we observed robust and durable antibody responses that remained above protective titers six months after vaccination. We further identified stable levels of antigen-specific T cell responses that remained detectable in the absence of antigen stimulation. Protists are known to increase plant growth through two main mechanisms: the microbial loop and the alteration of the root microbiome. The microbial loop is a nutrient recycling method in which protists provide inorganic nitrogen ions to the plant. Alteration of root microbiome leads to the removal of plant pathogens and shifting communities towards plant growth-promoting bacteria (PGPB). This study aimed to elicit which mechanism could produce the largest boost in shoot weight for Medicago truncatula. Antibiotic treatment failure is a public health crisis, with a 2019 report stating that roughly 35,000 deaths occur in the United States yearly due to bacterial infections that are unresponsive to antibiotics (1). One complication in the treatment of bacterial infection is antibiotic persistence which further compromises our battle to effectively treat infection. Bacterial persisters can exist in clonal bacterial cultures and can tolerate antibiotic treatment by undergoing reversible phenotypic changes. The American Cancer Society estimates that in 2023, 1,958,310 new cancer cases and 609,820 cancer deaths will occur in the United States [16]. A promising therapeutic option that has been supported by recent clinical trials is the use of oncolytic viruses to treat malignant tumors. The mechanism of action of existing treatments, such as chemotherapy, radiotherapy, and surgery, differs from that of oncolytic virus therapy because oncolytic viruses are able to affect cancer cells with specificity, minimizing side effects. ESKAPE pathogens (Enterococcus faecium, Staphylococcus aureus, Klebsiella

pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter

spp.) are seeing a growing resistance to multiple classes of antibiotics. Misuse and overuse of antibiotics have played directly into the resistance observed, and the problem is growing exponentially. Antibiotic resistance is partially due to several intrinsic factors limiting the drug's uptake. These include efflux pumps, increased biofilm production, and reduced cell wall permeability in the resistant bacteria. Fusarium graminearum is a devastating pathogen of wheat that causes Fusarium head blight (FHB) and contaminates the grain with the mycotoxin deoxynivalenol (DON). Resistance to FHB is quantitative and it is important to identify additional genes conferring resistance against it. The goal of this thesis was to examine if the constitutive expression of two sorghum phenylpropanoid pathway genes, SbCCoAOMT (encoding caffeoyl-CoA O-methyltransferase) and SbC3’H (encoding p-coumarate 3-hydroxylase), in the moderately-susceptible spring wheat CB037 can provide Type-I and Type-II resistance to F. graminearum and determine the underlying mechanisms of the enhanced resistance. Staphylococcus aureus is a bacteria that can exist asymptomatically, but can be dangerous due to it’s individual toxin profile. These various toxins, however, seem to be mutually exclusive from another in some cases. Samples of S. aureus often have superantigen SELX and some samples have Staphylococcal Enterotoxin (SE) A, B, or C. Through the use of Airtable and various methods of data organization and comparison on the strains we’ve collected on campus, we found that SELX exhibits an inverse relationship with SE- A/B/C, and that SE- A/B/C also exhibits exclusivity between the three toxins. Candida auris is a fungus that is creating a significant public health threat, causing severe healthcare associated infections. Commonly misidentified in laboratories, C. auris is extremely resistant to most antifungal medications. C. auris infections are most commonly identified through a culture of the blood or other bodily fluid, but can be misidentified for other Candida strains. The most reliable way to accurately identify C. auris is through matrix-assisted laser desorption/ionization time-of-flight mass spectroscopy (MALDI-TOF MS). Crimean-Congo Hemorrhagic Fever (CCHF) is a tick-borne virus. It was first identified in Crimea in 1944 and was later detected in Congo in 1969. It is endemic where it is mainly located in regions such as Africa, Europe, and Asia. CCHF is primarily transmitted through the bite of an infected tick, however direct contact with bodily fluids can also be another route of transmission. Staphylococcus aureus is a commensal bacterium found in the nares, skin, and mucosal membranes. In most cases, it causes no harm, but there are instances where it can become pathogenic and produce toxins that have superantigenic activity. Staphylococcal enterotoxin A (SEA), when ingested, is the causative agent responsible for classical food poisoning symptoms such as emesis, nausea, and others. When SEA enters the bloodstream, it can cause toxic shock due to mass T cell proliferation and cytokine storm. Acinetobacter was discovered in 1911 by Martinus Beijerinck. Acinetobacter baumannii didn't receive its scientific name until 1986. A. baumannii is now commonly referred to as "Iraqibacter" due to a rise in infections among US military personnel returning from Iraq and Afghanistan. After the Iraq War began in 2003, the frequency of cases began to rise, especially among patients in intensive care units. Samples taken from the CSP community were previously isolated and characterized as Staphylococcus aureus. In this study, the DNA from those positive samples were purified through a genomic prep and ran through a polymerase chain reaction for the SEB toxin gene. Staphylococcus aureus was isolated from bacteria swabs collected from participants at Concordia University St. Paul for this study. Isolated Staphylococcus aureus was then run through genomic prep to gather DNA from each strain. Collected DNA was amplified using PCR with specific primers to the targeted sec gene. The amplified sec gene was then transferred to gel electrophoresis, establishing whether the original strain contained the sec gene.

Chop, DelRio, and GrandSlam are phage with a Siphoviridae morphotype isolated from soil in Arkansas using the host Gordonia terrae 3612. All three are temperate, and their genomes share at least 96% nucleotide identity. These phage are assigned to cluster DI based on gene content similarity to other sequenced actinobacteriophage.

This work aims to investigate the antidermatophyte action of Azelaic acid (AzA) and evaluate its efficacy upon entrapment into transethosomes (TEs) and incorporation into a gel to enhance its application. Compared to microbiomes on other skin sites, the bacterial microbiome of the human hand has been found to have greater variability across time. To increase understanding regarding the longitudinal transfer of the hand microbiome to objects in the built environment, and vice versa, 22 participants provided skin microbiome samples from their dominant hands, as well as from frequently and infrequently touched objects in their office environments. Additional longitudinal samples from home environments were obtained from a subset of 11 participants. Wild Atlantic salmon in the Gulf of Maine (GOM) is a Distinct Population Segment (DPS) that has been listed since 2000 as endangered by the U.S. Fish and Wildlife Service (USFWS) and the National Oceanic and Atmospheric Administration (NOAA). The complex two-membrane organization of the envelope of Gram-negative bacteria imposes an unique biosynthetic and topological constraints that can affect translocation of lipids and proteins synthesized on the cytoplasm facing leaflet of the cytoplasmic (inner) membrane (IM), across the IM and between the IM and outer membrane (OM).
