Insulin And Glucagon Responses To Dietary Protein In Diabetic And Nondiabetic Men, Monica L. Blomstedt

Five men with non-insulin-dependent diabetes mellitus and fourteen nondiabetic men, were given test meals with soy protein, casein or additional carbohydrate in place of protein. Blood levels of insulin, glucagon, glucose and lipids were measured at fasting, one-half hour and two hours postprandial. To all meals, the nondiabetic subjects responded with a quick rise of the insulin level which decreased at two hours. In contrast, the diabetics showed a significantly lower insulin response at one-half hour (p < 0.05), but a tendency to higher levels at two hours. The same pattern was observed in the insulin to glucagon ratio. No significant difference between the meals was found in regard to insulin or glucagon responses, suggests that a sustained, although delayed postprandial insulin response, may be a precursor to the higher cholesterol and increased risk for cardiovascular disease among diabetics.

There is evidence that specific nutrients may modify hormonal balance and contribute to breast cancer etiology. To investigate the relationship between dietary nutrients and plasma reproductive hormones and to determine hormone-nutrient interrelationships, we measured plasma estrogens, androgens. progesterone and prolactin levels in 10 premenopausal Seventh-day Adventist (SDA) vegetarian and 10 premenopausal SDA nonvegetarian women. Over three days in each of three consecutive months. diet records and fasting midluteal blood samples were collected. The nonvegetarians consumed significantly more protein, total and saturated fats, oleic and linoleic acids, and cholesterol than did the vegetarians. Hormonal status of the two groups did not …

Red blood cells infected with malaria parasites experience a change in density making them lighter (less dense). This paper reports a new, rapid diagnostic technique using this change in density in the infected red blood cell. It is not only easy to use, but it is also compatible with the technological conditions found where the malaria problem is most acute - in many developing countries of the world. A small sample of blood drawn from a simple finger stick can be concentrated and examined microscopically for malaria parasites within 10 minutes. There is evidence that homocysteine may be a factor in increasing the risk of coronary heart disease (CHD). To explore this relationship further, we assessed the interrelation of dietary vitamins B-6, B-12 and folate, and plasma pyridoxal-5'-phosphate and vitamin B-12 with plasma free and protein-bound homocysteine levels. Fasting blood and three-day dietary records were obtained from nine men low-risk (LR) and five at high-risk (HR) for CHD. The HR mean systolic blood pressure, total cholesterol (TC), and TC/cholesterol ratios were significantly higher than LR levels. Groups were comparable by age, weight, height, skinfolds, exercise, and smoking history. No significant difference …

Cryptosporidiosis is a parasitic infection primarily of animals that has been recently recognized as an important disease in immunocompromised humans. Most cases to date have occurred in patients with acquired immunodeficiency syndrome (AIDS). Persons with abnormal humoral or cellular immune function, and even normal subjects, have also been affected.

To determine whether or not mice, when experimentally immunosuppressed, would develop cryptosporidiosis, a test was designed using the AKR/J strain. Eosinophils have long been associated with parasitic and allergic diseases. Methods of procurement and purification of eosinophils in the last ten years have allowed intensive investigation into their components, properties, functions, and mechanisms (Gleich et al, 1982). Eight healthy male lacto-ovo-vegetarian subjects averaging 28.0 ± 3.6 years of age (22 to 53 years) were fed a diet rich in α-linolenic acid (18:3 ω 3) for four weeks. Arterial blood pressure and body weight were recorded each week. Plasma 6-keto-prostaglandin F (6-keto- PGF ), thromboxane B2 (TXB2), total cholesterol, triglyceride, and total phospholipid fatty acid composition were analyzed at 0, 2 and 4 weeks.

Systolic blood pressure increased significantly between weeks 0 and 1 (106.5 ± 2.7 mmHg to 114.0 ± 3.7 mmHg; p < 0.003), but decreased to 107.3 t 2.6 mrHg (p < 0.02) at week 2. There was an insignificant increase in both systolic and diastolic blood pressures after four weeks. There was no significant change in either total cholesterol or triglyceride levels during the four weeks.

Schistosomiasis is a serious disease in humans. Two species of schistosome, Schistosoma mansoni and Schistosoma haematobium, organisms that cause the disease, parasitize humans in Libya. Schistosomes require aquatic snails in their life cycle. The purpose of this study was to investigate the environmental and the host relationships of the disease in the three regions of Libya where the disease is found. These relationships include:

Human serum high density lipoprotein subclasses, HDL2 and HDL3 , were isolated by preparative salt density gradient ultracentrifugation and further analyzed by electrophoresis on a 4 to 15% concentration gradient polyacrylamide gel.

The separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation for 272,000 g at 15°c in a swinging bucket rotor. High resolution concentration gradient gel electrophoresis was found to be particularly suitable for the separation of lipoproteins. The isolation of HDL by precipitation methods, heparin-manganese and dextran-magnesium have also been evaluated in this study, and were found not to completely separate …

Duck plague virus causes a hemorrhagic disease affecting the family Anatidae, resulting in 60-80% mortality in captive flocks. Plaque and neutralization assays for Anatid herpesvirus (AHV) require 5-10 days for completion and are restricted by the seasonal availability of fertile duck eggs. The enzyme linked innnunosorbent assay (ELISA) is rapid and specific and avoids the use of living cells. The Holland strain of AHV was grown on duck embryo fibroblasts and purified by banding on CsCl gradients. The resulting virus was used to raise antisera in prebled New Zealand white male rabbits. To determine the effect of folic acid supplementation on gingival healing after oral hygiene therapy, eight teeth (#3, 4, 5, 6, 11, 12, 13, 14) of sixteen human subjects with moderate levels of gingival irritation were studied. Clinical parameters included measurement of gingival fluid, pocket depth, bleeding upon probing, and plaque. Serum and red blood cell folic acid levels were determined using radioimmunoassay (RIA). Based on clinical parameters and serum folate levels, two similarly matched groups were established. The teeth were scaled and polished. Brushing and flossing instructions were given.

The purpose of this study was to obtain information on the morphologic changes that occur in cellular cementum exposed to periapical inflammation.

Ten single rooted human teeth were collected. There were five specimens of normal cellular cementum and five specimens of diseased cellular cementum, which had been exposed to periapical inflammation. Each specimen was fractured longitudinally through the apical foramen, leaving the organic component intact. The cross-sectional morphology of cellular cementum was then examined using an ARM Model 1000 scanning electron microscope. Energy dispersive x-ray spectrometry was also conducted on samples representative of the two test groups.

Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the 125IUdR release assay. The Balb/c mouse response to the growing HSV-induced tumor, treated with radiation and , was measured by survival rate, tumor growth and immune cytotoxicity of spleen and peritoneal exudate cells (PEC) as determined by 125IUdR release assay. The possible effects of ablastin, a reproduction-inhibiting antibody which is produced during the infection of the blood parasite, Trypanosoma musculi in the mouse, were investigated by fractionating the parasite into a supernatant fraction (designated as Group A test mice) and a pellet fraction (Group B test mice). Human Herpes simplex virus type 2 (HSV 2)-transformed Balb/c mouse cells (H238 cells) are malignant in Balb/c mice. The immune stratus of mice bearing H238 cell-induced tumors was studied and changes in immune response with time and with altered diet of the mice were measured.

In the experiments dealing with the status of immunity with time after tumor cell injection, mice were fed Purina Laboratory Chow (P.L.C.), injected with H238 cells, and sacrificed three to eight weeks post-injection. Duck plague is a hemorrhagic, necrotic disease found exclusively in the order Anseriformes, family Anatidae (ducks, geese, swans). The etiologic agent has been reported to be a herpes-like virus. The Holland strain of duck plague was found to induce cytopathic effects in cell culture (perinuclear vacuolation followed by lysis). A simple reproducible plaque assay for quantitating the virus was developed. Under conditions of the plaque assay, one-hit kinetics were observed. The host range of duck plague virus in cell culture was found to include the order Galliformes, family Phasianidae (pheasants, quails and partridges). Both dibutyryl cyclic AMP and trypsinized concanavalin A have been shown to inhibit growth in certain cell lines but little testing of these substances has been done using human tumor cell lines. Further, there are at this time no published experimental attempts to correlate the effects on growth with a particular phase of the cell cycle in any cell line.

The purpose of this study was to evaluate microscopically the effect of topically applied 2% chlorhexidine gluconate on the healing of gingival wounds in dogs.

One week after the teeth were scaled and polished, seven experimental wound sites were prepared in the facial gingiva adjacent to the four premolars on each side of the maxillary arch in six young Labrador dogs. Adults of the blood fluke Schistosoma mansoni were perfused from the hepatic portal system of CF1 female mice sixty to seventy days post infection. The worms were then incubated in Eagle’s minimal essential medium with glutamate and 0.03 mg/100 ml of nonlabelled aspartic acid. To this medium was added 10 μc of either NaH1403, glucose-UC14 or aspartate-4-C14. The worms were found to fix carbon from the NaH1403 and to incorporate the glucose and aspartate. A series of experiments involving Listeria monocytogenes was undertaken to study first the fecal shedding of Listeria by conventional mice challenged intraperitoneally with Listeria and second the fecal shedding and ability to survive of germ free mice with similar challenge. In addition, mites infesting Listeria infected mice were cultured for their possible contamination by Listeria.

The administration of the antibiotic, Chloramphenicol, to pregnant rats just prior to, and for four days after delivery, diminishes the incidence of the so-called "Wasting Disease" in neonatally thymectomized rats.

A total of 50 rats were divided into the following groups:

  1. Thymectomy only---nonmedicated 17 rats
  2. Thymectomy only---medicated 12 rats
  3. Thymectomy-adrenalectomy---nonmedicated 5 rats

In four separate experiments mouse embryo cell monolayers infected with polyoma virus and uninfected control mouse embryo cells were maintained in Eagle’s Minimum Essential Medium for periods of time ranging from 0 to 168 hours. The cell layers were removed from the glass by trypsinization. Samples of the cells were counted in a Coulter Counter and the remainder of the cells frozen after centrifugation, in a button. The literature dealing with the larva of Pacific Coast brachyuran crabs is slight. The prominent papers, those of Aikawa (1927, 1928, 1929, 1933, 1937), deal with a variety of genera and offer information useful in determin- ing family relationships. A single paper by Hart (1935) also deal with a number of brachyuran larvae found in the Pacific Ocean. The early embryology closely parallels that of other nematode species.

The mature embryos remain alive in saline at 35°C as long as 5 days. Dead embryos can be distinguished from living ones by virtue of the rapid staining by methylene blue by the former. Parturient female thelazias release embryos into saline, and embryos are found regularly in tear secretions of infected laboratory rabbits.

The purpose of this work was to determine the extent and effects of infestation of the eye worm of mammals, Thelazia californiensis, and to elucidate through experimentation or observation the mode of transmission.

A survey of literature revealed that thelaziasie (infestation by Thelazia species) may cause serious pathological changes in the eye and its adnexa, sometimes resulting in blindness. It was found that reports of thelaziasis in the United States were (with two exceptions) of cases in California. It was further found that T. californiensis has been reported in a wider variety of host than has any of other …
