Extracellular vesicles (EVs) in bovine milk confer beneficial physiologic effects to consumers. Industrial processing treatments may affect the amount or bioactivity of EVs intrinsic to bovine milk. We investigated how the content and concentration of EVs were affected by homogenization and thermal processing of raw bovine milk. Raw milk was processed by homogenization, low-temperature (LT) heat, or pasteurization [high-temperature short-time (HTST) and ultra-high-temperature (UHT)] in a pilot processing facility. EVs were isolated from the raw and processed bovine milk using differential ultracentrifugation and quantified using a nanoparticle tracking analyzer. The aim of this research paper was to evaluate the effect of a slow-release milk replacer on health and behaviour of neonatal dairy calves. The main objective of this thesis is to improve the understanding and awareness of methodologies to decrease phytoestrogens in conserved legumes without sacrificing forage nutritive value. In chapter 1, we discussed the main factors influencing each stage of hay production and our current understanding of the hay microbiome dynamics. The primary objective of haymaking is to dry forage enough (80-85% DM) to inhibit the growth of undesirable microbes and halt residual plant enzymatic activity that causes nutrient losses. Dairy producers play a central role in evaluating and seeking treatment or care for animal pain. The primary aim of this study is to examine dairy producers’ emotional states and professional quality of life and analyze the relationship between these variables and their perception of pain in dairy cattle. Dairy farm owners and managers of 65 southeastern U.S. herds participated in a survey that included demographic information, the Depression, Anxiety, and Stress Scale, a modified Professional Quality of Life Scale, and 23 items requiring participants to evaluate various painful conditions in cattle.

The objective of this thesis was to determine how pregnant, lactating Holstein cows on pasture interact with shade and sprinklers when offered a choice or no-choice between methods. Authors hypothesized that cows would use shade more then sprinklers, but sprinklers would be more effective at cooling cows. Additionally, authors predicted cows using shade more would have decreased lying bouts and steps and an increase in time lying, compared to cows choosing sprinkler. Results indicate cows will use both methods when offered alone or together. However, cows will choose to be exposed or in shade at lower temperature humidity index values, …

Black seed oil concentrations of 0%, 0.1%, 1% and 5% were added to milk inoculated with kefir grains and incubated at 25°C for 22 h. The pH and microbial count indicated 1% black seed oil caused low inhibition (P > 0.05) of fermentation, but 5% black seed oil caused significant inhibition of the kefir microorganisms (P < 0.05).

Fetch cows, dairy cattle that do not voluntarily enter automated milking systems (AMS), increase labor costs, and decrease efficiency. The objective of this study was to describe fetch cows based on fetch lists and herd-management software data. Outcomes and Impact Quarterly
A needs assessment was conducted by the Utah State University Dairy Extension Team to identify the most urgent issues faced by Utah dairy farmers. The assessment found that the top five priority needs were implementing new technology, improving public perception of water use, managing risk, succession planning, and reducing feed costs. The results will guide dairy Extension programming in Utah. Several workshops and events have already been planned to address these needs. Overall, the assessment will help USU Extension provide research-based educational programs to meet the needs of the dairy industry in Utah.

This report presents findings from a pilot survey conducted among undergraduate and graduate students (N = 410) at the University of Nebraska-Lincoln about students’ perspectives on technology usage, consumption, and sustainability in dairy production systems. An interdisciplinary research team developed the survey instrument and report. The main purpose of this pilot study was to create and administer survey items to support further research on experiential education and outreach opportunities related to robotics in small-scale dairy production and rural economic development. Japanese encephalitis virus (JEV), a zoonotic flavivirus, is principally transmitted by hematophagous mosquitoes, continually between susceptible animals and incidentally from those animals to humans. For almost a century since its discovery, JEV was geographically confined to the Asia-Pacific region with recurrent sizable outbreaks involving wildlife, livestock, and people. However, over the past decade, it has been detected for the first time in Europe (Italy) and Africa (Angola) but has yet to cause any recognizable outbreaks in humans. Our objective was to understand dairy employees' perceptions and educational needs at the onset of the COVID-19 pandemic. A bilingual (English and Spanish), anonymous survey targeted at dairy employees was circulated nationwide via university and allied industry media outlets. Responses (n = 63) from 11 states were received (May–Sep. 2020). Respondents worked in herds ranging from 50 to 40,000 animals in size. Dairy managers (33%) responded mostly to the English survey (52%), whereas entry-level workers (67%) chose the Spanish format (76%). Survey results highlighted different perspectives, educational needs, and preferred sources of information between English- and Spanish-speaking dairy workers. Overall, …

In this study we determined the impact of increasing dietary whole cottonseed (WCS) on rumen fermentation, nutrient digestibility, and microbial community composition. This study contributes novel information to the dairy community deepening the understanding of how including different levels of WCS can affect the rumen environment. This research was conducted in continuous culture fermenters. Treatments included a control diet without WCS, or the control diet plus 5, 10, or 15% (dry matter) WCS. Although traditional ryegrass pastures are still commonly used, they require intense management and input and do not perform well during the summer months. Multispecies systems have been recognised as more sustainable, needing less fertiliser and being tolerant to drought and heat. Atrial Fibrillation (AF) is the most common cardiac arrythmia in adults with likely sex-specific risk factors. Female sex hormones may be important in modulating risk for atrial fibrillation. We hypothesize that pregnancy and progesterone (P4), a hormone found in high levels during pregnancy with abrupt withdrawal immediately after parturition, modulates AF susceptibility in female goats. Cardiac specific TGF-?1 transgenic female goats and age-matched wild-type (WT) female goats were utilized. Pacemakers were implanted in all animals for continuous arrhythmia monitoring and AF inducibility. AF inducibility was evaluated using 5 separate 10 s bursts of atrial pacing (160 - 200 ms). In …

Cheesemaking in Nepal is full of love, passion, and creativity. What began as a Swiss development project in the 1950s has now grown into a full-fledged industry, providing jobs, creative outlets, and cultural expression for many. Nepal’s Dairy Development Corporation owns several factories throughout the country, setting the tone for the industry by establishing the standard price for milk and process for cheesemaking. Research suggests that androgens increase skeletal muscle growth by modulating polyamine biosynthesis. As such, the objective of this study was to investigate effects of anabolic hormones, polyamine precursors, and polyamines relative to proliferation, protein synthesis, and the abundance of mRNA involved in polyamine biosynthesis, proliferation, and protein synthesis in C2C12 and Sol8 cells. Cultures were treated with anabolic hormones (trenbolone acetate and/or estradiol), polyamine precursors (methionine or ornithine), or polyamines (putrescine, spermidine, or spermine). Consumption of the total Western diet (TWD) in mice has been shown to increase gut inflammation, promote colon tumorigenesis, and alter fecal microbiome composition when compared to mice fed a healthy diet, i.e., AIN93G (AIN). The objective of this retrospective observational study was to determine whether preweaning calf behaviors and milk replacer (MR) intake from an automatic calf feeder were associated with age at first calving and first-lactation performance. Calves were housed in groups with access to an automatic calf feeder for 7 wk with a maximum milk allowance of 1,800 g/d of MR (12 L/d). Outcomes of interest included age at first calving (n = 137), first-lactation mature-equivalent 305-d (ME305) milk yield (n = 132), and first-lactation ME305 energy-corrected milk (ECM) yield (n = 132). Automated monitoring devices have become increasingly utilized in the dairy industry, especially for monitoring or predicting disease status. While multiple automated monitoring devices have been developed for the prediction of clinical mastitis (CM), limitations in performance or applicability remain. The aims of this study were to (1) detect variations in reticuloruminal temperature (RRT) relative to an experimental intramammary challenge with Streptococcus uberis and (2) evaluate alerts generated automatically based on variation in RRT to predict initial signs of CM in the challenged cows based on severity of clinical signs and the concentration of bacteria (cfu/mL) in the infected quarter separately. …

Dairy cows are predisposed to diseases during the postpartum period. Dystocia has been associated with increased risk for disease, which is likely the result of increased tissue trauma and stress during the prolonged parturition. To attenuate the inflammatory response seen in dystocic animals and improve well-being, we assessed the effects of a glucocorticoid, dexamethasone administered within 12 h after calving. Dystocia was defined as a difficult birth resulting in a prolonged calving (≥70 min after the amniotic sac appears) and was monitored through 3 video cameras in the close-up dry-cow pen. Long-term gains in dairy cow production efficiency have already been delivered through genetic selection. Brewer’s spent grains (BSGs), a byproduct from beer production, are generated in excess globally. Most often, they are sold or given to proximate cattle farmers for use as feed. However, spent grain can also be used as a medium for fungal cultivation. Given that certain fungal species have the capability to degrade lignin and produce protein, the cultivation of fungi on spent grains may serve to enhance the nutritional profile of the grains for their use as cattle feed. Listeria monocytogenes (Lm) is a foodborne pathogen of major safety concerns. This bacterium can resist/adapt to environmental stressors and persist through forming robust biofilms despite dairy plants following stringent surface cleaning protocols. Surface roughness is considered a major factor controlling bacterial attachment and biofilm formation. Food contact surfaces like stainless steel (SS) have low surface roughness for easy cleaning. On the other hand, non-contact surfaces like floors need a rougher surface to ensure employee safety. The newly proposed physically adjusted NDF system (paNDF) system aims to maintain a favorable rumen environment (e.g., pH) by taking into account both the physical and chemical composition of diet. Thus, the objective of this study was to determine the effects of water-soluble carbohydrates (WSC) and non-forage NDF inclusion in a paNDF system on production performance, rumen pH and fermentation profile, rumen bacterial abundance, feeding behavior and nutrient digestibility in late-lactating Holstein cows.
