Scanning Microscopy

The occurrence and cation content of bacteria in a eutrophic freshwater lake (Rostherne Mere, Cheshire, UK) were investigated over a one year sampling period in relation to cation changes in the lake surface water and phytoplankton.

Scanning electron microscope examination of trawl-net and filtered samples demonstrated bacterial association with Anabaena, Aphanizomenon and diatoms. Direct counts of associated and unassociated bacteria showed that increases in bacterial population relate to population decline of major algal constituents.

Scanning Electron Microscopy

Electron microscope autoradiography and X-ray microanalysis have been used for the detection and quantitation of cations in the bacterium Pseudomonas tabaci. These techniques differ in the information they provide (relating to either cation uptake or in situ levels), their applicability to different cations, their sensitivity and their spatial resolution. With uptake of 63Ni2+, high resolution autoradiography (involving gold latensification and physical development) demonstrated a high degree of cation localisation to the central nucleoid area (glutaraldehyde-fixed cells) and within this to the constituent chromatin (acetic-alcohol preparations).

Streptococcus and Staphylococcus were collected from patients hospitalized at the Oregon Burn Center, Emanuel Hospital, Portland, Oregon, between October 27, 1980 and January 1, 1981. Forty-five isolates of Staphylococcus and twenty-one isolates of Streptococcus, Lancefield Group D, were included in the investigation.

Lake Mead is a deep, subtropical, moderately productive, desert impoundment with a negative heterograde oxygen profile occurring during; the summer stratification. investigations of the Boulder Basin of Lake Mead by the University of Nevada were initiated in November 1971. The primary objective of the study was to determine what effects industrial and sewage effluent from the Las Vegas metropolitan area, discharged into Las Vegas Bay, have had on the water quality and limnological conditions of Boulder Basin. Data from the 1975-76 period are presented in detail, with earlier data included in the summaries and discussions.

Thirteen bacterial cultures were isolated from the North arm of Great Salt Lake during the months of January and February of 1973. Eleven isolates were gram-negative pleomorphic rods which lysed in hypotonic solution. The remaining two were gram-positive cocci. All isolates and one known strain of Halobacterium salinarium were subjected to examination of morphological, cultural, physiological, and biochemical characteristics. A numerical taxonomic analysis was applied to the compiled characters to compute a coefficient of similarity for each individual isolate as compared to all other isolates. It is the purpose of this study to determine whether the drug atropine has an effect on bacteria found normally inhabiting the human body, most specifically the gastrointestinal system. Atropine is one of the oldest drugs in medicine. It is derived from the roots and leaves of solanaceous plants such as henbane and Jimson weed. Atropine is often termed a belladonna alkaloid because it can also be found in the deadly nightshade plant, Atropa belladonna. During bacteriological studies of the Logan City Sewage Oxidation Ponds, Logan, Utah, a group of bacteria was discovered which could lyse killed cells of Escherichia coli. The present research effort was an attempt to determine whether two isolates from this earlier study might play a role in the reduction of coliforms and other bacteria in the oxidation ponds. It was also desired to characterize partially the lytic enzymes produced by the two isolates and to determine as far as possible the organisms' identity.

This study developed from observations made during an earlier study on Lake Mead which was reported in CHE-70, Water Quality Study of Lake Mead. Results from that study indicated that poor-quality water was flowing into the Las Vegas Bay reach of Boulder Basin, Lake Mead. On July 10, 1968, while observing the effects of ultra-violet light on a plate culture of Sarcino lutea, o contaminant was found growing in the form of a colony surrounded by a clear zone of inhibition where S. lutea had failed to grow. A smear from this contaminant colony was found to contain a Gram positive, chain-ond spore-forming bacillus. The organism was isolated and its antagonism against S. lutea was reshocked.
