Evidence suggested there might exist in brain a contractile protein: 1) actomyosin contains Nacetyl aspartic acid; brain contains a high concentration of the free compound, 2) actomyosin functions as a Mg dependent ATPase; brain contains a Mg dependent ATPase, 3) actomyosin-like contractile proteins have been described in cells, membranes, and tissues other than muscle. Therefore whole rat brains were extracted with 0.6M KCl and the extract diluted to O.lM KCl. The precipitate exhibited superprecipitation (SP) properties which required both Mg and ATP. SP was inhibited by p-chloro-mercuribenzene sulfonic acid (10- M) but not by ouabain (10 M). The protein purified …

(1) Compartmentation of glutamate metabolism in brain cortex previously observed only in vivo, has now been demonstrated in vitro. This was shown by using [U-14C]aspartate and [U-14C]glutamate as tracer substrates. (2) Preparation and maintenance of the slices at 0° resulted in reversible inhibition of glutamine synthesis. Preincubation at 37° for 10 min or preparation of the slices at room temperature partially overcame this inhibition. (3) Transfer to fresh medium after preincubation had an added stimulatory effect on glutamine synthesis. (4) Incubation in high K+ medium (27 mM) altered the relative specific activity of glutamine. Guinea pig brain cortex slices incubated in media containing U-C14 aspartic acid form glutamine of higher specific activity than glutamic acid (Fed. Proc. -25, 713 (1966) ). Glutamic acid and glutamine from such experiments were enzymatically decarboxylated and the o/o of label in C-1 ranged from 37 to 42o/o. These values exceed the maximum predicted from the known operation of the citric acid cycle viz. 33o/o. When l-C14 acetate was injected into mice or incubated with brain slices the aspartate isolated from brain had more than 50o/o of the label in C-4. From the known operation of the citric acid …

The determination of urinary 3-methoxy-4-hydroxyphenylethylene glycol (HMPG), a major metabolite of epinephrine and norepinephrine by gas-liquid chromatography and electron capture detection is described. A purified extract of HMPG was treated with trifluoroacetic anhydride to form the highly sensitive HMPG-trifluoroacetate derivative. Chromatography was performed on a 6% QF-I- 0065 column at 158°. Recovery was found to be 55·4% using [3H]HMPG. Two sets of eight identical samples demonstrated a standard deviation from the mean of 6.9% and 5.7%. Thirty-five normal human urine specimens assayed by this procedure gave a mean HMPG excretion of 0.86 #gfmg creatinine (range 0.25-1.49). Six urine samples from …

The presence, in brain, of metabolic pools of glutamic acid have been demonstrated in vivo but not in tissue slices. Brain tissue slices prepared in the cold in Ringer's-bicarbonate containing 1% glucose and stored in an ice bath go not show compartmentation when incubated at 37 (10-20 min.) in a similar medium which contains tracer quantities of C aspartic acid, U.l.; the specific activity of glutamine remains considerably below that of glutamic acid. Transglutaminase is an enzyme which catalyzes the Ca++-activated incorporation of amines into proteins. More detailed studies of the action of this enzyme showed that the incorporated amines displaced ammonia, and that only L-glutamine and not asparagine residues in the protein or peptide are substrates for this enzyme. This enzyme will not act on free glutamine, hence is not to be confused with glutaminase or transamidases which will act on free glutamine as a substrate. A young Metasequoia tree was debarked; its branches and heartwood were removed. The branches, sapwood and heartwood were separately ground to 8o mesh and air-dried. The moisture and lignin contents of these were determined. "Milled-wood" lignin was isolated from the sapwood sawdust, and was characterized by means of its ultraviolet and infrared absorption spectra. As a means of determining the empirical formula and the number of certain functional groups present in this lignin, an acetylated derivative and a phenylhydrazone were prepared. The use of gas-liquid chromatography (G.L.C.) for the analysis of amines of biological interest is currently under investigation in our laboratory. An intensive study of the chromatographic properties of a number of derivatives was presented by VandenHeuvel et al. (1), and this laboratory in a short communication reported on the possible use of heptafluorobutyrylated amine derivatives (2). As was noted in this early communication, sensitivity considerations must maintain priority in any proposed assay because of the extremely small quantities of these compounds present in biological fluids. Until the discovery of the major product of catecholamine n1etabolism, vanillylmandelic acid (VMA), by Annstrong in 1957 (1), biochemical studies of epinephrine and norepinephrine metabolism were dependent upon determination of the small quantities of these materials excreted unchanged in the urine. The methods designed to measure VMA have proved adequate to detect the gross abnormalities in metabolism usually associated with catecholamine-producing tumors (2-7), but more exact procedures are required for quantitative metabolic studies. Present spectrophotometric and electrophoretic methods are nonspecific in that they yield values for VMA that are higher than those obtained by chromatographic and isotope dilution techniques (5). …

When N -phenylsydnone is hydrolyzed by acid, an internal oxidation- reduction reaction takes place with the formation of phenylhydrazine, formic acid, and carbon dioxide. The mechanism shown below was suggested for this reaction by Baker and Ollis. Aside from the nature of the end products of this hydrolysis, the strongest evidence cited by Baker and Ollis in support of their mechanism is the paper by Kenner and Mackay who reported the isolation of a-acylhydrazines when the hydrolysis was carried out in benzene with stoichiometric quantities of water and hydrochloric acid. The rate of fixation of carbon dioxide in vivo into aspartate, glutamate, oxoglutarate, and glutamine of cerebral cortex and liver in the absence and presence of elevated concentrations of tissue ammonia was studied. The specific activities of the isolated metabolites were related to the specific activity of the tissue CO2. It was attempted to obtain an approximate steady state level of tissue 14CO2 by maintaining a constant isotope concentration in the venous and arterial blood through continuous intravenous administration of NaH14CO3. 1. Radicinin, a metabolite of the formula C12H1205 was isolated from the carrot plant pathogen Stemphylium Tadicinum and described in earlier communications. Hansen isolated a metabolite from S. radicinum that inhibited the growth of Lepidium sativum and named it stemphylone. Carbon dioxide fixation in brain was studied in cats to which NaHC14O3, with and without ammonia, was administered by intracarotid infusion. Glutamic and aspartic acids, glutamine, glutathione, and γ-aminobutyric acid were isolated from blood, brain, and liver, and their specific activities were determined. The data indicate a significant incorporation of CO2 into the amino acids of the cerebral cortex, presumably by way of the citric acid cycle. Without simultaneous ammonia infusion, the specific activity of aspartic acid is 3 times that of glutamine, whereas in the presence of ammonia the ratios of specific activity of both compounds are closer to …

In experiments reported some years ago the enzyme-catalyzed exchange of the amide group of glutamine and asparagine with 15N ammonia was demonstrated but an exchange of the amide groups of protein bound dicarboxylic amino acids could not be explored owing to the unfavorable experimental conditions1 . The synthesis of a phosphatidyl tripeptide has been accomplished. It was prepared by (a) phosphorylation of D-α,β-distearin with phenylphosphoryl dichloride and pyridine, (b) esterification of the resulting distearoylL- α-glycerylphenylphosphoryl chloride with N-carbobenzoxy-L-serylglycylglycine benzyl ester in the presence of lutidine, and (c) removal of the protective groups by catalytic hydrogenolysis. The O-( distearoyl-L-α-glycerylphosphoryl)- L-serylglycylglycine was obtained in an over-all yield of 12 per cent. The synthesis of N-carbobenzoxy-L-serylglycylglycine benzyl ester, for which two procedures have been reported, has been simplified considerably by condensing N -carbobenzoxy-L-serine with glycylglycine benzyl ester by means of N ,N' -dicyclohexylcarbodiimide. The conditions for the growth of Sterad and isolation of radicinin are described. The formula C12H1205 wasas established for this compound by preparation and analyses of several derivatives. Degradation and spectrophotometric studies were employed to establish the molecular formula. Studies on the mode of action of radicinin revealed that it caused an increase in the rate of dehydrogenation of isopropyl alcohol by FIB, but that it had no effect on the quantity or degree of desaturation of the fat synthesized by FIB
