Mechanical stimulation has been imposed on living cells using several approaches. Most early investigations were conducted on groups of cells, utilizing techniques such as substrate deformation and flow-induced shear. To investigate the properties of cells individually, many conventional techniques were utilized, such as AFM, optical traps/optical tweezers, magnetic beads, and micropipette aspiration. In specific mechanical interrogations, microelectro- mechanical systems (MEMS) have been designed to probe single cells in different interrogation modes. This paper details an investigation into methods and designs of 3D printing a microfluidic system capable of droplet emulsion using NinjaFlex filament. The specific field in which this paper’s experiment is rooted is dubbed “BioMEMS,” short for bio microelectromechanical systems. One prominent research area in BioMEMS is developing a “lab on a chip.” Essentially, the goal is to miniaturize common lab processes to the micro scale, rendering it possible to include these processes in a small chip. The objective of this research work is to develop a centrifugal microfluidic system for general purposes based on microfabrication technologies including SU-8 photolithography, polydimethylsiloxane (PDMS) casting. The main contribution of this research is to integrate a flyball governor system into the polymer based centrifugal microfluidic platform. This work presents the modeling, design, and testing of an underwater microactuation system. It is composed of several thermomechanical in-plane microactuators (TIM) integrated with a ratchet system to provide long displacements and high forces to underwater microelectromechanical systems (MEMS). It is capable of actuating a 200µN load 110µm. It is a two-layer silicon MEMS device fabricated with a MEMS fabrication process, PolyMUMPS. This work also shows the development of an elliptic integral model to analyze the compliant fixed-guided beams in the TIM and gives new insight into the buckling behavior, reaction forces, and displacement of the beams. A novel high flow rate cell capture design was introduced to overcome the limitations of the current technologies or methods for rare target cell capture.
