Curcumin is a bright orange compound with myriad applications for human health and wellness. Curcumin occurs naturally in the plant Curcuma longa (commonly known as turmeric) but must be extracted from the roots in an environmentally unfriendly fashion to obtain commercially relevant amounts of the compound. In addition, extraction of curcumin from turmeric spice yields a mixture of various curcuminoids, presenting an issue for isolating it in its pure form and complicating its use in clinical settings.

Small, biologically active molecules with unique properties and applications are potential solutions to a wide range of threats to global health including infectious agents and neurodegenerative disease. Experimental studies on a class of oligomeric p-phenylene ethynylenes (OPEs) have shown potential both as bioactive antimicrobials and fluorescent sensing agents for tracking amyloid-β (Aβ) aggregates found in Alzheimer’s Disease (AD). A second type of small molecule with potential applications in AD therapy, curcumin, has been found to interfere with Aβ fibril growth. Radiotherapy is used as a primary treatment for many cancers, including lung cancer. Although radiotherapy has proven to be an effective cancer treatment, its use is heavily limited due to the peripheral toxicity to healthy tissue. In this work, the antioxidant, curcumin, was tested as a radioprotectant to reduce radiation damage to healthy cells. Curcumin has been limited in use due to its poor bioavailability. In order to avoid problems associated with free curcumin delivery, curcumin poly(beta-amino ester) (CPBAE) was synthesized.

Skin cancer is the most prevalent cancer diagnosis worldwide. Squamous cell carcinoma (SCC) is one of the most common diagnoses. Fortunately, these cancers are rarely fatal if detected and treated early on. However, current treatment options can be painful, disfiguring and can require long-term treatment courses, resulting in poor patient compliance and cancer progression. Since SCC begins as precancerous lesions, an opportunity exists for early preventative interventions which this work aims to address. Researchers have been trying to fight cancer with synthesis of new bioactive compounds but many of these novel drugs have low solubility in water and it is difficult to deliver them into a patient's body. One way of solving this particular problem is to use nanoscale drug delivery systems. In this dissertation, we describe using an ultrasonic assisted layer-by-layer encapsulation process to prepare anti-cancer drugs with 50∼200 nm particle size with designed coating to achieve sustained release and target delivery.
