Osteoarthritis is one of the most common causes of disability in adults in America. It is a progressive and degenerative disease where the articular cartilage is broken down and lost from the surfaces of bones causing chronic pain and swelling in the joints, and currently has no cure. The most commonly osteoarthritis starts from a focal lesion on the cartilage surface, which will expand on the surface and downwards through the thickness of the tissue. The current gold standard for correcting cartilage focal lesions is the osteochondral autograft/allograft transplantation (OAT), which replaces the defect with a fresh osteochondral graft. The …

Melanin is a naturally produced pigment found within the human body. Melanin is known for its ability to protect against Ultra Violet light, but also its ability to allow for mechanical protection. Making melanin a good addition to biomedical devices such as hydrogels. Silk hydrogels are weak in their load bearing capabilities but are known for their biocompatibility, biodegradability, and porosity. Tissue engineering aims to regenerate damaged and deceased tissue by combining cells with scaffold made from an appropriate biomaterial and providing a conducive environment to guide cell growth and the formation or regeneration of new tissue or organ. A uniform dispersion of reactants is necessary to achieve a complete reaction involving multi-components, especially for the crosslinking of rigid high-performance materials. In these reactions, miscibility is crucial for curing efficiency. This miscibility is typically enhanced by adding a third component, a plasticizer. For the reaction of the highly crystalline crosslinking agent hexamethylenetetramine (HMTA) with a strongly hydrogen-bonded phenol formaldehyde resin, furfural has been traditionally used as the plasticizer. However, the reason for its effectiveness is not clear. In tissue engineering, the scaffold plays a critical role in guiding and supporting cells to function and grow optimally. The electrospun nanofibrous scaffold can serve as a near ideal substrate for tissue engineering because it has high surface area and the three-dimensional interconnected porous network can enhance cell attachment and proliferation. Core-shell nanofibrous scaffolds produced with coaxial electrospinning allow bioactive molecule encapsulation to improve cell adhesion, mediate and promote the proper signaling among the cells for their functioning and growth. This dissertation has aimed to fabricate polypeptide based biomaterial and characterize physical properties. Electrospinning is used as a tool for the sample fabrication.
