This project report provides detailed information on designing and building a DIY Incubator. The device is indicated for growing and maintaining BSL-1 mammalian or bacterial cell culture with a temperature control in the range of 25-45°C ± 1°C. The device is made fully programmable via an Arduino, and is structurally designed to fit two T75 cell culture flasks at a time. The device is able to return to a specified temperature set point within 10 minutes and maintains 5% CO2 partial pressure conditions. Histology often requires a tissue specimen to be embedded so that it may be sectioned, stained, and mounted on a microscope slide for viewing. One common method of tissue embedding for rapid histology is freezing, since freezing allows tissue to be stored without the need for fixing. Frozen tissue is often embedded in a medium such as Optimal Cutting Temperature (OCT) compound so that it can be sectioned using a cryostat. Photo-crosslinkable polymeric biomaterials have emerged in the field of biomedical research to promote tissue regeneration. For example, scaffolds that can be crosslinked and hardened in situ have been known to make suitable implant alternatives. Since injectable and photo-crosslinkable biomaterials offer the advantage of being minimally invasive, they have emerged to compete with autografts, a current highly invasive method to repair diseased tissue. A series of novel photo-crosslinkable, injectable, and biodegradable nano-hybrid polymers consisting of poly(ε-caprolactone fumarate) (PCLF) and polyhedral oligomeric silsesquioxane (POSS) has been synthesized in our laboratory via polycondensation. Especially, being able to image and detect human diseases at the cellular and molecular level allows people to diagnose diseases at early stages and to study the molecular mechanisms behind various diseases. We systematically vary the controllable laser parameters, namely, wavelength, intensity, and exposure time of the laser on single cells to clearly establish laser parameters that allow negligible cell damage with significant cellular position control. To accomplish this goal, embryonic day 7 (E7) chick forebrains neurons were cultured with control and test cells selected one hour after seeding to allow attachment, at which time test cells were subjected to the laser. Cells were imaged at 4, 12, 24, and 36 hours after laser exposure to evaluate viability and functionality by using the Live/Dead Viability Cytotoxicity Kit (Molecular Probes L3224) and measuring …
