Polymers have deep roots as drug delivery tools, and are widely used in clinical to private settings. Currently, however, numerous traditional therapies exist which may be improved through use of polymeric biomaterials. Through our work with infectious and oxidative stress disease prevention and treatment, we aimed to develop application driven, enhanced therapies utilizing new classes of polymers synthesized in-house. Applying biodegradable poly(β-amino ester) (PBAE) polymers, covalent-addition of bioactive substrates to these PBAEs avoided certain pitfalls of free-loaded and non-degradable drug delivery systems. An atomic force microscope (AFM) was used to measure the steric forces of lipopolysaccharides (LPS) on the biofilm-forming bacteria, Pseudomonas aeruginosa. It is well known that LPS play a vital role in biofilm formation. These forces were characterized with a modified version of the Alexander and de Gennes (AdG) model for polymers, which is a function of equilibrium brush length, L, probe radius, R, temperature, T, separation distance, D, and an indefinite density variable, s. Gene therapy involves the use of nucleic acids, either DNA or RNA for the treatment, cure, or prevention of human diseases. Synthetic cationic polymers are promising as a tool for gene delivery because of their high level of design flexibility for biomaterial construction and are capable of binding and condensing DNA through electrostatic interactions.
Our lab has developed a novel polymer (poly (polyethylene glycol-dodecanoate) (PEGD), a polyester of polyethylene glycol (PEG) and dodecanedioic acid (DDA). The goal of this dissertation was to parse the roles of physical, mechanical and chemical cues in the phenotype plasticity of smooth muscle cells (SMCs) in atherosclerosis. We first developed and characterized a novel synthetic hydrogel with desirable traits for studying mechanotransduction in vitro. This hydrogel, PEG-PC, is a co-polymer of poly(ethylene glycol) and phosphorylcholine with an incredible range of Young’s moduli (~1 kPa - 9 MPa) that enables reproduction of nearly any tissue stiffness, exceptional optical and anti-fouling properties, and support for covalent attachment of extracellular matrix (ECM) proteins. Atom level computer simulations of the arabinan and cellulose interface were performed to better understand the mechanisms that give arabinan-cellulose composites (ArCCs) their strength with the goal to improve man-made ArCCs. The molecular dynamics (MD) software LAMMPS was used in conjunction with the ReaxFF/c force field to model the bond between cellulose and arabinan. A cellulose nanocrystal with dimensions 51 x 32 x 8 Å was minimized with various weight percent of water, 0%, 3%, 5%, 8%, 10%, and 12%. After the system was equilibrated for at least 100,000 femtoseconds, an arabinan molecule composed of 8 arabinose rings was added …

The purpose of this project was to create a unique SCBA (self-contained breathing apparatus) for a firefighter named Chris Gauer. This prototype consists of a SCBA headgear connected to a polycarbonate-formed stoma mask with a medical-grade sanitary silicone hose.

Encapsulation of a model anti-cancer drug, 5-Fluorouracul (5-FU) into biocompatible core-shell nanofibers of polycaprolactone (PCL) nanofibers was fabricated using a coaxial electrospinning process. Our work aims to solve these issues using a novel method of fabrication of fibers featuring confinement of drugs within a biodegradable core-shell structure, thereby permitting sustained release of drugs to specific sites of treatment, such as tissues affected with tumor cells. The coaxial electrospinning was performed using a sheath polymer solution consisting of a 14 wt% PCL solution and a 5 wt% solution of 5-FU as the core solution. An imbalance of NO is closely associated with many serious diseases such as neurological disorders, cardiovascular diseases, chronic inflammations and cancers. Herein two chemiluminescence-based devices (a real-time NO measurement device and a controllable NO delivery device) were developed to facilitate the NO quantitative study and obtain information for NO related drug design.

Sterilization of tissue engineered scaffolds is an important regulatory issue and is at the heart of patient safety. With the introduction of new biomaterials and micro/nano structured scaffolds, it is critical that the mode of sterilization preserve these built-in features. Conventional sterilization methods are not optimal for engineered polymeric systems and hence alternate systems need to be identified and validated. PCL is polyester with a low melting point (heat labile), susceptible to hydrolysis and is popular in tissue engineering. Polylactic acid (PLA) is becoming an increasingly important biopolymer for packaging applications. PLA brittleness limits its applicability. This study evaluated PLA properties with increasing amounts of added polyethylene glycol (PEG) plasticizer. Oxygen transmission rate (OTR) of cast films was determined using the newly available Dynamic Accumulation (DA) method. Arrhenius temperature sensitivity of OTR and polymer Permeability was also determined. With the aim of creating a biodegradable scaffold for tympanic membrane (TM) tissue regeneration, core-shell nanofibers composed of a poly(caprolactone) shell and a poly(ethylene glycol) core were created using a coaxial electrospinning technique. In order to create fibers with an optimal core-shell morphology, the effect of relative humidity (RH) on the core-shell nanofibers was systematically studied, with a FITC-BSA complex encapsulated in the core to act as a model protein. Magnesium has been identified as a promising biodegradable implant material because it does not cause systemic toxicity and can reduce stress shielding. However, it corrodes too quickly in the body. Titanium, which is already used ubiquitously for implants, was chosen as the alloying element because of its proven biocompatibility and corrosion resistance in physiological environments. Thus, alloying magnesium with titanium is expected to improve the corrosion resistance of magnesium.

Bacterial cellulose (BC) is a natural hydrogel made of nanofibers. This material has been used in commercial products, including wound dressings. BC can be modified and optimized for improved performance in multiple applications. This work will focus on producing and characterizing resorbable cellulose, a composite for bone applications, and a composite for a synthetic venous valve leaflet.

Investigations into the use of gold nanorods (Au-NRs) for biological applications are growing exponentially due to their distinctive physicochemical properties, which make them advantageous over other nanomaterials. Au-NRs are particularly renowned for their plasmonic characteristics, which generate a robust photothermal response when stimulated with light at a wavelength matching their surface plasmon resonance. In tissue engineering, the scaffold plays a critical role in guiding and supporting cells to function and grow optimally. The electrospun nanofibrous scaffold can serve as a near ideal substrate for tissue engineering because it has high surface area and the three-dimensional interconnected porous network can enhance cell attachment and proliferation. In microsurgery, the human hand imposes certain limitations in accurately positioning the tip of a device such as scalpel. Any errors in the motion of the hand make microsurgical procedures difficult and involuntary motions such as hand tremors can make some procedures significantly difficult to perform. This is particularly true in the case of vitreoretinal microsurgery. The most familiar source of involuntary motion is physiological tremor. The peridynamic method is used here to analyse the effect of van der Waals forces on the mechanical behaviour and strength and toughness properties of three-dimensional nanofibre networks under imposed stretch deformation. The peridynamic formulation allows for a natural inclusion of long-range forces (such as van der Waals forces) by considering all interactions as ‘long-range’. We use van der Waals interactions only between different fibres and do not need to model individual atoms. Fracture is introduced at the microstructural (peridynamic bond) level for the microelastic type bonds, while van der Waals bonds can reform at any time. We conduct statistical …

To assess corrosion rates of metal alloy bio-absorbable stents an experimental set-up was designed to mimic the coronary artery environment. The artery was modeled using 4mm diameter Tecoflex tubing and the metal alloy stents were inserted into the tubing using a catheter. As is the case in cardiac surgery, the catheter with the stent and a microballoon were maneuvered to the desired position. The microballoon was then slowly inflated to expand the stent and compress it against the tubing walls. The catheter and microballoon were then withdrawn. Microfluidics refers to manipulation, precise control, and behavior of fluids at the micro and nanoliter scales. It has entered the realm of science as a way to precisely measure or mix small amounts of fluid to perform highly controlled reactions. Glass and polydimethylsiloxane (PDMS) are common materials used to create microfluidic devices; however, glass is difficult to process and PDMS is relatively hydrophobic. However, most research has not focused on the copolymer itself in regards to how long it can be used effectively and if varying the concentrations of polylactic acid (PLA) and polyglycolic acid (PGA) affect the resulting properties. Femtosecond laser machining is a direct-write lithography technique by which user-defined patterns are efficiently and rapidly generated at the surface or within the bulk of transparent materials. When femtosecond laser machining is performed with tightly focused amplified pulses in single-pulse mode, transparent substrates like fused silica can be surface patterned with high aspect ratio (>10:1) and deep (>10 μm) nanoholes. Treatments of bone injuries and defects have been largely centered on replacing the lost bone with tissues of allogeneic or xenogeneic sources as well as synthetic bone substitutes, which in all lead to limited degree of structural and functional recovery. As a result, tissue engineering has emerged as a viable technology to regenerate the structures and therefore recover the functions of bone tissue rather than replacement alone. Research was conducted to observe bacterial growth on the surface of metals in a static bioreactor. Metal and non-metal samples were subjected to bacterial exposure (1 day and 9 days). The metal samples were surface treated prior to bacterial exposure. The microstructures of the surface treated samples were analyzed by optical microscopy. After exposure, the microstructures of the samples were analyzed by scanning electron microscopy (SEM). The analysis suggested that microbial attachment on the surface was related to the underlying microstructure of steel. Bone defects are a prevalent problem in orthopedics and dentistry. Calcium phosphate-based coatings and nanocomposites offer unique solutions towards producing scaffolds with suitable physical, mechanical and biological properties for bone regeneration.
We developed a novel method to synthesize hydroxyapatite (HA) particles with high aspect ratio using sol-gel chemistry and hydrothermal treatment. We obtained tunable pure-phase carbonated-HA in the form of micro/nanorods and nanowires (diameters 25-800 nm). To mimic the structure of bone, HA nanowires were homogenously mixed within poly(ε-caprolactone) (PCL) to produce nanocomposites with improved mechanical properties as determined by uniaxial tensile testing.
