Background

Intramuscular fat (IMF) and intramuscular connective tissue (IMC) are often seen in human myopathies and are central to beef quality. The mechanisms regulating their accumulation remain poorly understood. Here, we explored the possibility of using beef cattle as a novel model for mechanistic studies of intramuscular adipogenesis and fibrogenesis.

Methods

Skeletal muscle single-cell RNAseq was performed on three cattle breeds, including Wagyu (high IMF), Brahman (abundant IMC but scarce IMF), and Wagyu/Brahman cross. Sophisticated bioinformatics analyses, including clustering analysis, gene set enrichment analyses, gene regulatory network construction, RNA velocity, pseudotime analysis, and cell-cell communication analysis, were performed to elucidate …

Aquaculture is a large part of the food production sector which is greatly expanding. One of the largest losses in aquaculture is due to pathogens. Current solutions for protecting farmed finfish from pathogens can be very expensive with variable efficiency. Current disease prevention strategies include vaccination. Types of vaccines include immersion vaccines, feed vaccines, and injectable vaccines. The most popular solution is oil-based injectable vaccines due to its protection. However, the oil-based adjuvant used in most of these formulations causes adverse reactions in the fish including reduced growth. The equine racing industry is one of the main proponents in Kentucky’s economic infrastructure. In this industry there has come a need to investigate the nature of the proximal sesamoid bone (PSB). Breakdowns involving the PSBs are the leading cause in racehorse deaths in the industry, with still little known about what causes this bone to fracture. This study seeks to shed insight by investigating the internal structure of the PSB. Using microCT scanning, the internal porous structure was captured. From there, noticeable differences in the pores were noticed and quantified using fast Fourier transform (FFT) analysis.

We numerically study solutions to a length-structured matrix model for fish populations in which the probability that a fish grows into the next length class is a decreasing nonlinear function of the total biomass of the population. We make conjectures about the convergence properties of solutions to this equation, and give numerical simulations which support these conjectures. We also study the distribution of biomass in the different age classes as a function of the total biomass.

This research focused on developing biosensing method and biosensing device for rapid detection of pathogens in poultry: Salmonella Typhimurium and avian influenza virus H5N1. The first part of the dissertation reports an original research on the development of a portable biosensing device for Salmonella detection. The device was designed and constructed based on a previously developed optical biosensing method, using immuno-magnetic nanoparticles to specifically capture target bacteria, and immuno-quantum dot beads to label the target bacteria for fluorescence detection. The pronuclear (PN) injection technique was first established in mice to introduce foreign genetic materials into the pronuclei of one-cell stage embryos. The introduced genetic material may integrate into the embryonic genome and generate transgenic animals with foreign genetic information following transfer of the injected embryos to foster mothers. Following the success in mice, PN injection has been applied successfully in many other animal species. The engineering of floating media biofilters has been optimized over the years. The backwashing process has made them more energy and water efficient. Likewise, moving bed bioreactors (MBBR) are gaining interest and popularity because they are relatively affordable to build. The release of the Final Guidance from the US Food and Drug Administration on the commercialization of genetically engineered animals has sparked renewed discussion over the ethical, consumer, and regulatory implications of transgenesis in animal agriculture. Animal welfare critiques have focused on unexpected phenotypic effects in animals used in transgenic research, rather than on the health and welfare implications of the intended productivity enhancement. Unless breeding goals are redefined to reflect social concerns, the occurrence and magnitude of undesirable side effects may increase and consumer confidence in the nascent technology may be undermined.

The kinetics of milk coagulation are complex and still not well understood. A deeper understanding of coagulation and the impact of the relevant factors would aid in both cheese manufacturing and also in determining the nutritional benefits of dairy products. A method using confocal microscopy was developed to follow the movement of milk fat globules and the formation of a milk protein network during the enzyme-induced coagulation of milk.
