Determining the location of rare proteins in cells typically requires the use of on-sample amplification. Antibody based recognition and enzymatic amplification is used to produce large amounts of visible label at the site of protein expression, but these techniques suffer from the presence of nonspecific reactivity in the biological sample and from poor spatial control over the label. Polymerization based amplification is a recently developed alternative means of creating an on-sample amplification for fluorescence applications, while not suffering from endogenous labels or loss of signal localization. To improve the lignocellulose conversion for ABE in high solids fermentation, this study explored the feasibility of cycling the process through the cellulolytic or/and solventogenic phases via intermittent flushing of the fermentation media. Five different flushing strategies (varying medium ingredients, inoculum supplement and cycling through phases) were investigated. Flushing regularly throughout the cellulolytic phase is necessary because re-incubation at 65 °C significantly improved glucose availability by at least 6-fold. The solvents accumulation was increased by 4-fold using corn stover (3-fold using miscanthus) over that produced by flushing only through the solventogenic phase. Executive Summary This report describes best practices and tools to streamline and expedite utility relocations when they are required as part of road construction projects. As part of this effort, a research team from the Kentucky Transportation Center (KTC) conducted extensive qualitative research that involved mapping current practices at the Kentucky Transportation Cabinet (KYTC) and reviewing utility manuals from KYTC and other state transportation agencies. The KTC research team also conducted in-depth interviews with KYTC engineers and staff as well as representatives from utility companies (UCs). This report draws on data from around the United States to describe the features of a successful railway/highway at-grade crossing management program; it emphasizes the highway/trackbed structures and crossing surfaces. Developing a structurally adequate crossing system is imperative, as this will produce a smooth surface and a stable highway/trackbed that prolongs crossings’ lifespans while keeping maintenance costs low and minimizing the number of disruptions encountered by highway and railway traffic. This report documents the long-term performance of 89 railway/highway at-grade crossings in Kentucky and adjacent states that contain asphalt underlayment underneath the ballast. Asphalt underlayment increases load carrying capacity for trains and highway vehicles while enhancing the support structure’s waterproofing and confinement. The asphalt underlayment crossings studied were installed at sites that had historically exhibited poor performance and short service lives. Most of the crossing sites evaluated were 10-15 years old; however, their ages ranged up to 30 years. All of the crossings investigated have shown excellent performance since the installation of asphalt underlayments. No crossing failures, such as excessive …

Suggestions are provided for determining the most cost-effective rehabilitation procedure, techniques to insure the appropriate installation procedures are followed onsite, and instructions for post-project administration and inspection procedures. An ideal Railway/Highway At-Grade Crossing Management program involves selecting costeffective practices when designing new crossings and rehabilitating existing crossings. This report outlines two strategies to enhance KYTC’s existing program. First, it describes a process that uses decision-option diagrams to optimize the assessment and implementation of engineering solutions in order to restore desired smoothness, minimize settlement in the postconstruction phase, and foster acceptable long-term performance of crossings following their rehabilitation. Anthropogenic emissions of aromatic hydrocarbons promptly react with hydroxyl radicals undergoing oxidation to form phenols and polyphenols (e.g., catechol) typically identified in the complex mixture of humic-like substances (HULIS). Because further processing of polyphenols in secondary organic aerosols (SOA) can continue mediated by a mechanism of ozonolysis at interfaces, a better understanding about how these reactions proceed at the air–water interface is needed. This work shows how catechol, a molecular probe of the oxygenated aromatic hydrocarbons present in SOA, can contribute interfacial reactive species that enhance the production of HULIS under atmospheric conditions. The migration of Li-ions in lithium-ion battery cannot be simply described by Fick's second law; the interactions among ionic migration, field, and stress need to be taken into account when analyzing the migration of Li-ions. Using the theory of thermal activation process, the flux for ionic migration under concurrent action of electric field and mechanical stress is found to be a nonlinear function of the gradient of electric potential and the gradient of stress. Electric field can either accelerate or retard the growth of the lithiation layer, depending on polarity of the field.

The inland waterway system of the U.S. is a vital network for transporting key goods and commodities from the point of production to manufacturers and consumers. Shipping materials via the inland waterways is arguably the most economical and environmentally friendly option (compared to hauling freight by trains or railways). The inland waterway system is a vital part of the nation’s multi-modal freight network. Although less visible than other modes, inland waterways allow shippers to transport bulk commodities in a relatively cheap and environmentally-friendly method. To ensure this transportation mode remains a feasible option and accommodates growth, it must continue to be safe, efficient, and functional. This synthesis provides comprehensive perspective on the financial prospects of the inland waterways system. It analyzes current funding levels, along with proposed funding changes and reforms.

We explore and quantify the coupled dynamics of a pair of micron scale cantilevers immersed in a viscous fluid that are also directly tethered to one another at their tips by a spring force. The spring force, for example, could represent the molecular stiffness or elasticity of a biomolecule or material tethered between the cantilevers. We use deterministic numerical simulations with the fluctuation-dissipation theorem to compute the stochastic dynamics of the cantilever pair for the conditions of experiment when driven only by Brownian motion. High-quality HfO2 cannot be grown directly on Si substrate using atomic layer deposition (ALD), and an interfacial oxide layer is needed. Traditionally, interfacial oxide layer is formed either in SC1 solution (2 NH4OH: 4 H2O2: 200 H2O) or by ozonated water spraying. A highly hydrophilic SiO2 interfacial layer was in-situ formed in the ALD chamber using 1 cycle of ozone and water. The HfO2 deposited on this interfacial layer showed great growth linearity. The gate leakage current is comparable to that formed using chemical oxide as the interfacial layer. …

Conventional semi-infinite analytical solutions of correlation diffusion equation may lead to errors when calculating blood flow index (BFI) from diffuse correlation spectroscopy (DCS) measurements in tissues with irregular geometries. Very recently, we created an algorithm integrating a Nth-order linear model of autocorrelation function with the Monte Carlo simulation of photon migrations in homogenous tissues with arbitrary geometries for extraction of BFI (i.e., αDB ). Background
As part of the fungal endophyte genomes project, we maintain genome browsers for several dozen strains of fungi from the Clavicipitaceae and related families. These genome browsers are based on the GBrowse software, with a large collection of in-house software for visualization, analysis, and searching of genome features.
Although GBrowse supports serving multiple data sources, such as distinct genome assemblies, from a single GBrowse instance, there are advantages to maintaining separate instances for each genome. Due to the localized surface plasmon (LSP) effect induced by Ag nanoparticles inside black silicon, the optical absorption of black silicon is enhanced dramatically in near-infrared range (1,100 to 2,500 nm). The black silicon with Ag nanoparticles shows much higher absorption than black silicon fabricated by chemical etching or reactive ion etching over ultraviolet to near-infrared (UV-VIS-NIR, 250 to 2,500 nm). The maximum absorption even increased up to 93.6% in the NIR range (820 to 2,500 nm). The high absorption in NIR range makes LSP-enhanced black silicon a potential material used for NIR-sensitive optoelectronic device.

Organic and/or inorganic fertilizers are used to promote crop growth yet the rate of fertilizer application is often based on the nitrogen needs of the crop and not the phosphorus needs, especially when utilizing animal manures with low N-to-P ratios as compared to plant N:P uptake. Because of this, soil phosphorus levels in many areas of the world are in excess of crop needs. Excessive levels of phosphorus in water bodies such as streams and lakes can result in a condition known as eutrophication. This report documents an analysis of traffic crash data in Kentucky for the years of 2009 through 2013. A primary objective of this study was to determine average crash statistics for Kentucky highways. Rates were calculated for various types of highways and for counties and cities. Difference criteria were used for exposure.
Average and critical numbers and rates of crashes were calculated for various types of high ways in rural and urban areas. This project involved an evaluation of the codes which relate to roadway contributing factors. This included a review of relevant codes used in other states. Crashes with related codes were summarized and analyzed. A sample of crash sites was inspected to evaluate the accuracy and consistency of the data being evaluated.
The results of the study are recommendations for a coding format to enable accurate and consistent data concerning the roadway as it affected the crash as well as examples of areas of training to improve the accuracy and consistency of the data.

Complex engineered nanoparticles (CENPs), which have different core and surface components, are being developed for medicinal, pharmaceutical and industrial applications. One of the key challenges for environmental health and safety assessments of CENPs is to identify and quantity their transformations in biological environments. This study reports the effects of in vivo exposure of citrate-coated nanoalumina with different rare isotope labels on each component. Anecdotal evidence suggests that historically African American communities on the fringes of cities and towns in North Carolina have been systematically denied access to municipal drinking water service. This paper presents the first statistical analysis of the role of race in determining water access in these fringe areas, known as extraterritorial jurisdictions. Using publicly available property tax data, we quantified the percentage of residences with municipal water service in each census block in Wake County (the second-largest by population in North Carolina). Metal oxidation at high temperatures has long been a challenge in cermet solar thermal absorbers, which impedes the development of atmospherically stable, high-temperature, high-performance concentrated solar power (CSP) systems. In this work, we demonstrate solution-processed Ni nanochain-SiOx (x < 2) and Ni nanochain-SiO2 selective solar thermal absorbers that exhibit a strong anti-oxidation behavior up to 600 °C in air. The thermal stability is far superior to previously reported Ni nanoparticle-Al2O3 selective solar thermal absorbers, which readily oxidize at 450 °C. Lipid extraction from Scenedesmus sp. microalgae using hot compressed hexane (HCH) was investigated. Extraction performance was evaluated near the critical point of hexane and was compared with that of hexane extraction performed at room temperature and pressure, and the Bligh and Dyer extraction method. Experimental data showed that HCH significantly improves the lipid yield and rate of lipid extraction compared to the use of hexane at ambient conditions. High yields of biodiesel-convertible lipid fractions were rapidly achieved at the critical point of hexane, at a level comparable to that of the Bligh and Dyer method.

The mechanical properties of ultrathin films synthesized by atomic layer deposition (ALD) are critical for the liability of their coated devices. However, it has been a challenge to reliably measure critical properties of ALD films due to the influence from the substrate. In this work, we use the laser acoustic wave (LAW) technique, a non-destructive method, to measure the elastic properties of ultrathin Al2O3 films by ALD. Understanding the long-term effects and possible toxicity of nanoceria, a widely utilized commercial metal oxide, is of particular importance as it is poised for development as a therapeutic agent based on its autocatalytic redox behavior. We show here evidence of acute and subacute adverse hepatic responses, after a single infusion of an aqueous dispersion of 85 mg/kg, 30 nm nanoceria into Sprague Dawley rats. Light and electron microscopic evidence of avid uptake of nanoceria by Kupffer cells was detected as early as 1 hr after infusion.
