Turkish Journal of Medical Sciences

Urothelial carcinomas (UC) are the most common cancer type of the urinary system. The early diagnosis and treatment of UC are very important. The aim of this study was to evaluate the importance of the nuclear matrix protein 22 (NMP22) in early diagnosis of UC and to present its variation in cystitis and urothelial dysplasia (UD). Twenty patients with UC, 20 patients with UD and 20 patients with cystitis diagnosed by urinary cytological examination were enrolled in this study. In addition, 20 healthy people constituted the control group. Urine samples were collected from all patients and control subjects for NMP22 measurement …

Turkish Journal of Medical Sciences

The aim of the present study was to determine the lengths of humerus segments in the Turkish population and compare these with the data from other countries' population for use in forensic and archeological cases. For this purpose one hundred and twenty (56 left side and 64 right side) male adult dry Caucasians' humerus bones were taken to measure the morphometric properties of humerus segments. Six segments on the articular surface of the humerus (Maximum height of the humerus (MHH), and the distances between the articular segment of the humeral head and the greater tuberosity (H1), caput humeri and collum …

Reversal Of Endotoxin-Induced Hypotension By Inhibition Of Inducible Nitric Oxide Synthase Activity Is Associated With Improved Oxidative Status In Rat Heart, Aorta And Mesenteric Artery, Bahar Tunçtan, Belma Korkmaz, Hati̇ce Yildirim, Lülüfer Tamer, Uğur Ati̇k, C. Kemal Buharalioğlu

Turkish Journal of Medical Sciences

Overproduction of reactive oxygen and nitrogen species leads to oxidative stress and decreased total antioxidant capacity, which is responsible for high mortality from several diseases such as endotoxic shock. Nitric oxide (NO) produced by inducible NO synthase (iNOS) during endotoxemia is the major cause of vascular hyporeactivity, hypotension and multiple organ failure. We investigated whether NO-mediated oxidative stress in rat heart, aorta and mesenteric artery is involved in the attenuation of endotoxin-induced hypotension by inhibition of iNOS. In conscious male Wistar rats, injection of endotoxin (10 mg/kg, i.p.) caused a gradual fall in mean arterial pressure (MAP) for 4 hours …

Turkish Journal of Medical Sciences

The aim of the retrograde filling in apical resection operation is to obtain an effective and hermetic apical sealing. According to the development of technology many materials are now used in apical resection operations for retrograde fillings. An in vitro dye leakage study was performed to test the sealing ability of IRM, MTA, amalgam and zinc-phospate cement materials. Forty single rooted extracted human teeth were used in this study. After cleaning and shaping, all the roots were obturated with zinc-phospate cement and gutta-percha. Teeth were randomly divided into four groups. After root-end resections of the teeth in all groups, 3 …

Turkish Journal of Medical Sciences

Acrylic resin dentures may have cytotoxic effects on oral tissues. However, there is sparse data about the cytotoxic effect of fiber-reinforced acrylic resin materials. The aim of this in vitro study was to determine the cytotoxicity of a heat-polymerized acrylic resin denture base material reinforced with glass, carbon fibers and unreinforced acrylic resin denture base material. 100 acrylic resin discs were assigned to 5 experimental groups (n=20) and 1 control group. Group NOF did not receive any fiber. Roving type glass (Vetrolex) and carbon fibers (Type Tenox J) were introduced into the acrylic resin specimens of Group RGF and Group …

Primary Structure Of The Carbohydrate-Binding Modules In Various Cellulolytic, Thermophilic, Anaerobic, Ethanol-Producing Isolates, Melek Özkan, Gülay Özcengi̇z

Turkish Journal of Biology

In the present study, the carbohydrate-binding module (CBM) coding sequences of the cellulosomes of 13 thermophilic, cellulolytic, anaerobic, ethanol-producing bacterial isolates having some variations in their growth and cellulose degradation capacities were amplified by PCR and then sequenced. The sequence analysis of the amplicons revealed that CBMs of 7 of the isolates including the isolate 7-9-1 with the highest capacity of cellulose degradation in solid medium have 100% identity in both nucleotide and amino acid sequences to CipB of Clostridium thermocellum in the compared regions. On the other hand, CBM of the isolate 7-1-2, also having a high cellulolytic activity, …

Ethidium Bromide Spot Test Is A Simple Yet Highly Accurate Method In Determining Dna Concentration, Murat Kasap, Si̇nem Torol, Gülçi̇n Gacar, Fatma Budak

Turkish Journal of Medical Sciences

A commonly used method to measure the amount of DNA in nucleic acid preparations is to use UV irradiation. However, if the DNA sample contains impurities, measuring UV irradiation is misleading. A quick way to estimate the amount of DNA in such samples is to analyze spot densities created by UV-induced fluorescence emitted by ethidium bromide. The amount of DNA is proportional to the amount of fluorescence. In many references, the spot density analysis is recommended for rough estimation of DNA concentration and is done by visual examination and comparison of the densities of each spot with the densities of …

The Oncocytic Variant Of Papillary Thyroid Carcinoma, Reşi̇t Doğan Köseoğlu, Nurper Onuk Fi̇li̇z

Turkish Journal of Medical Sciences

Papillary thyroid carcinoma (PTC) has many subtypes. A rare subtype consisting of papillary projections covered by cells oncocytic in character has been defined in recent years. Oncocytes with characteristic nuclear features of PTC are seen in the thyroid aspiration cytology of this subtype. In the cytological diagnosis, this variant could be confused with other oncocytic lesions of the thyroid. We discuss here the cytological and histopathological features of the oncocytic variant of PTC diagnosed in the aspiration biopsy and thyroid resection material in a 42-year-old female patient.

Genetic Transformation Of Citrus Paradisi With Antisense And Untranslatable Rna-Dependent Rna Polymerase Genes Of Citrus Tristeza Closterovirus, Bayram Çevi̇k, Richard F. Lee, Charles L. Niblett

Turkish Journal of Agriculture and Forestry

Protein and RNA-mediated forms of pathogen-derived resistance (PDR) have been developed against many viruses in different plants. However, no resistance has been reported against Citrus tristeza virus (CTV), a closterovirus, in Citrus species transformed with coat protein genes or other sequences of CTV. The successful use of replication-associated genes in RNA-mediated resistance in other crops prompted the use of the RNA-dependent RNA polymerase (RdRp) gene of CTV for the development of RNA-mediated PDR in Citrus. The RdRP gene was amplified from CTV isolate DPI3800 from Florida and used to generate antisense (RdRp-AS) and untranslatable (RdRp-UT) constructs with point mutation consecutive …
