The capacity to selectively target DNA sequences within a complex mixture is a useful feature for genomic studies. Several methods aimed at undertaking this feat have utilized biotinylated oligonucleotides and streptavidin beads to capture DNA but have low efficiencies, require PCR, or cannot be combined with high-throughput sequencing. The work presented here developed a protocol referred to as selected enrichment through capture (SEC) that uses a biotinylated “capture primer” to target and concentrate specific DNA sequences at high efficiencies, eliminates the use of PCR, and can be combined with high-throughput sequencing. The objective of this study was to design an electron collimation system for Elekta Infinity accelerators with significantly reduced applicator weights, while maintaining acceptable in-field beam flatness and out-of-field leakage dose. The design was restricted to 6-20 MeV beams for 6x6 to 25x25 cm2 applicators at 100 cm SSD with the MLCi2 treatment head. In Aim1, measured off-axis profiles showed that the current Elekta collimation system met dose flatness (4%) and IEC leakage dose specifications in the patient plane for custom 7, 13, and 20 MeV beams at MBPCC. Place-based concentration effects can be detrimental to individual health outcomes and overall community health. These useful properties are related to the coexistence and coupling of several degrees of freedom, including charge, lattice, orbital and spin. I investigate the effects of the issuance of the PCAOB Part II report on audit fees and audit quality. The PCAOB replaced the peer review auditor program with an independent inspection of audit firms. Upon completion of each inspection, the PCAOB issues inspection reports that include a public portion (Part I) of identified audit deficiencies, and most include a nonpublic portion (Part II) of identified quality control weaknesses which is not disclosed publicly unless the audit firm does not sufficiently complete remediation during the next 12 months. During the latter half of the twentieth century there was a marked shift in the way that many scholars approached analysis of late-nineteenth-century tonality. This shift in approach was motivated by the behavior and interaction of harmonic and melodic entities encountered in the music of the great composers of the nineteenth century, such as Schubert, Schumann, Brahms, Mahler, Wagner, and Strauss. Multiple NSF (National Science Foundation)-funded MARGINS Source-to-Sink cruises were conducted in the Gulf of Papua (GoP), Papua New Guinea (PNG), from 2003 through 2005 to better understand how sediment is created, transported, and deposited. Although much work has been done on the data collected during these cruises, palynological analysis has never been conducted on the hundreds of available cores. The first phase of this project (Chapters 2 and 3) examines the connection between modern depositional regimes in the GoP and species assemblages recovered. Examining a broad range of texts,“‘Superbly Sterile:’ Queer Reproduction in Victorian Literature and Culture,” argues that Thomas Hardy’s final naturalist novels, popular nineteenth-century vampire narratives, and the fiction of Oscar Wilde queered the bildungsroman (or novel of development) through characters who failed or refused to progress along sexual and maturational timelines. Where these texts’ critics have tended to read them as cautionary tales about homosexuality or predatory female sexuality, this dissertation contends that they also presented alternate forms of kinship and reproduction. Edwardsiella ictaluri is a gram negative bacterium that is the causative agent of enteric septicemia of catfish. In 2011, this bacterium was identified as the causative agent of massive death in zebrafish populations in U.S. In this project, we found that isolates of E. ictaluri from zebrafish comprise a unique strain that differs from the type strain of E. ictaluri phenotypically as well as genetically. Also, strains of E. ictaluri from zebrafish are non-infectious in channel catfish Ictalurus punctatus by immersion. Chapter 1 outlines the synthesis of an array of pheromone analogs that are derivatives of the dideoxy sugar ascarylose. In 2012, excavations were conducted at Witz Nabb’ and Killer Bee the last remaining above sea level features Paynes Creek National Park, Belize. Salt is a basic biological necessity that was in limited supply at inland Maya cities. The ancient Maya of coastal Belize produced by fire enhanced evaporation of salt enriched brine. Survey and excavation at inundated salt works in a shallow lagoon in Paynes Creek National Park provide extensive evidence of this technique in the form of briquetage, the remains of pots used in the fire evaporation. Protocols for patterning nanostructures of polymers were developed to enable scanning probe microscopy (SPM) studies of surface properties at the molecular level. A chemically selective surface for patterning polymers was generated by combining particle lithography with organosilane immersion. Poly(N-allyl glycine), a biocompatible and backbone degradable polypeptoid, was grown on Si(111) at confined amine sites using surface-initiated polymerization (also known as the “graft from” approach). This case study, with four elementary school students as participants, examined the use of books and bibliotherapy when experiencing a traumatic incident. Utilizing fiction books, I explored how literature could be a powerful tool for overcoming life-altering events and circumstances, such as losing a pet, going through a divorce, or dealing with a grandparent who is suffering from Alzheimer's, with a bully in school, or the death of a loved one. This dissertation examines the paleoecological records from the Shark River Estuary in the southwestern part of the Everglades National Park (ENP), Florida, with primary goal of reconstructing the Holocene history of the coastal mangrove ecosystem in the Florida Coastal Everglades. Roughly 15 meters of sediments were collected from 4 study sites and subjected to loss-on ignition, palynological, and X-ray fluorescence analyses. According to the literature, the earliest communities of Rhizophora mangle (red mangrove) occurred prior to 8,000 cal yr BP in the south-central area of the Belize Barrier Reef Platform. Atmospheric droplets, such as fog, which can have an exceptionally high surface area-to-volume ratio, also have a large range of physical and chemical properties (e.g. temperature, pH, ionic strength, and chemical composition) based on location, time of day, and emission source. The purpose of this research study was to determine the support and resources needed by first generation college students and how community mentor programs could bridge the gaps in existing support. A purposeful sampling method was used to select three first generation college student participants who had personal experiences in community mentor programs. All three of the first generation college student participants are now college graduates. Two of the three participants currently work with a local community mentor program.
