Major depressive disorder (MDD) is a leading cause of disability worldwide, with a lifetime prevalence rate of approximately 20%. Inadequate pharmacological treatment methods for MDD are a significant debilitating factor. Patient estimates suggest that the treatment resistance rate for pharmacological interventions is over 30%. Postmortem analyses of human tissue of individuals diagnosed with MDD have shown an increase in Poly (ADP-ribose) polymerase 1 (PARP-1) mRNA gene expression in prefrontal cortical white matter when compared to psychiatrically normal brain tissue. Recent studies have shown elevated neuroinflammation in a large subset of individuals diagnosed with schizophrenia. A pro-inflammatory cytokine, tumor necrosis factor-alpha (TNFα), has been directly linked to this neuroinflammation. This study examined the effects of two TNFα modulators (PD2024 and PD340) produced by our collaborators at P2D Bioscience, Inc., to alleviate auditory sensorimotor gating deficits and reduce microglial cell activation present in the polyinosinic:polycytidylic (Poly I:C) rodent model of schizophrenia. Auditory sensorimotor gating was assessed using prepulse inhibition and microglial activation was examined and quantified using immunohistochemistry and confocal microscopy, respectively. Caffeine is the most consumed psychoactive substance in the world, and most caffeine consumption in coffee and energy drinks is intended to produce a psychoactive effect. However, caffeine is not a primary reinforcer in preclinical paradigms – non-human species do not reliably take the drug to produce a psychoactive effect. However, caffeine is a ‘reinforcement enhancer’ in preclinical models; the effects of caffeine increase the motivation to obtain other non-drug reinforcers. Research has revealed that schizophrenics are significantly more likely to smoke cigarettes than the general population, and consume nicotine products at a much more prevalent rate. Further exacerbating this issue, it has been previously demonstrated in clinical populations that the type of antipsychotic treatment administered (typical versus atypical) may result in either an increase or a decrease of already heightened smoking behavior within the schizophrenic population. Myocardial infarction (MI), often referred to as a heart attack, is a serious health issue in the United States. There is a well-documented link between MI and major depressive disorder (MDD), with a high incidence of MDD occurring after an MI. Overlapping pathologies have been observed within the hippocampus of the brain in animal models of MI and depression. These observations suggest that pathobiological cross-talk between the heart and brain could have a role in the etiology of MDD that occurs after an MI. One of the most common childhood disorders, attention-deficit hyperactivity disorder (ADHD) places individuals at a higher risk for nicotine (NIC) dependence. Approximately 37.2% of individuals with ADHD currently smoke compared to the 18.3% of individuals with no record of mental illness. Methylphenidate (MPH; Trade name Ritalin) is the most commonly prescribed treatment for ADHD. Research regarding the synergistic effects of MPH and NIC, however, is divided. Some research indicates that MPH may enhance susceptibility to NIC effects, whereas other studies report that MPH may inhibit sensitization to NIC. Nicotine has multiple behavioral effects as a result of its action in the central nervous system. Nicotine strengthens the behaviors that lead to nicotine administration (primary reinforcement), and this effect of nicotine depends on mesotelencephalic systems of the brain that are critical to goal directed behavior, reward, and reinforcement. Nicotine also serves as a ‘reinforcement enhancer’ – drug administration enhances behaviors that lead to other drug and nondrug reinforcers. Cardiovascular disease (CVD) is the leading cause of death in the United States. Nearly half of the people diagnosed with heart failure (HF) die within 5 years of diagnosis. Brain abnormalities secondary to CVD have been observed in many discrete regions, including the hippocampus. Nearly 25% of patients with CVD also have major depressive disorder (MDD), and hippocampal dysfunction is a characteristic of both diseases. In this study, the hippocampus and an area of the hippocampal formation, the dentate gyrus (DG), were studied in a canine model of HF. Methylphenidate Conditioned Place Preference In Juvenile And Adolescent Male And Female Rats, Elizabeth D. Freeman

This investigation was an analysis of the effects of methylphenidate (MPH; trade name: Ritalin) on drug reward using the conditioned place preference (CPP) behavioral paradigm in a rodent model and underlying mechanisms of this effect. Animals were conditioned in adolescence from postnatal day (P)33-39) or P44-49 with saline, 1 or 5 mg/kg MPH. Rats administered 5 mg/kg but not 1 mg/kg MPH, resulted in a significant preference that was more robust in younger male adolescent rats. The 5 mg/kg dose of MPH also resulted in a significant decrease of the dopamine transporter in both the nucleus accumbens and striatum, revealing …

Circadian rhythm is a type of endogenous clock that controls daily behavioral patterns in most organisms. Spiders have been shown to exhibit both circadian and non-circadian rhythms in their behaviors. This rhythmicity may allow spiders to cope with diel changes in environmental conditions. Both diurnal and nocturnal behavior have different sets of costs and benefits to a species’ survival. Achaearanea tepidariorum is one species in which potential circadian rhythmicity has never been studied. Due to its foraging behavior, it was predicted that its daily activity would be arrhythmic.
