Prenatal opioid exposure has increased as a consequence of both increased Opioid Use Disorder (OUD) and prescription use for treatment of pain and OUD. However, human studies on prenatal opioid exposure are complicated by several confounding variables and thus, use of an animal model can lessen these confounds. Illicit use of opioids produces fluctuating blood levels whereas prescribed use of opioids results in stabilized blood levels of opioids. Neuroimaging studies have revealed consistent abnormalities in functional connectivity within specific neural networks that may serve as biomarkers for major depressive disorder (MDD). It is important to find inexpensive, non-invasive techniques that target these biomarkers to make diagnosis easier and more objective. EEG microstates are quasi-stable potential topographies that are thought to reflect the quasi-stable network activity of the underlying neural generators. MDD has been shown to alter features of the four canonical EEG microstates (A, B, C, D) with some conflicting results. The current study employed a Pavlovian conditioning paradigm to investigate the influence of pre-stimulus unconscious threat anticipation on subsequent fear responses in associative learning. Additionally, we explored the differences in pre-stimulus anticipation among individuals with varying levels of anxiety. Electrodermal response (EDA) was chosen as a primary measure due to its robustness in fear conditioning paradigms. Moreover, the State-Trait Anxiety Inventory (STAI) was utilized to assess anxiety levels in participants. Aerobic metabolism is known to generate damaging ROS, particularly hydrogen peroxide. Reactive oxygen species (ROS) are highly reactive molecules containing oxygen that have the potential to cause damage to cells and tissues in the body. ROS are highly reactive atoms or molecules that rapidly interact with other molecules within a cell. Intracellular accumulation can result in oxidative damage, dysfunction, and cell death. Due to the limitations of H2O2 (hydrogen peroxide) detectors, other impacts of ROS exposure may have been missed. Humans have a fundamental need to form and maintain social connections, and thus experiencing social exclusion is extremely distressing as it threatens this basic human need. Individuals who are socially excluded often respond aggressively, not only towards their ostracizers but also towards innocent bystanders, with ostracism being implicated in extreme acts of violence such as school shootings. However, individual differences in behavior exist within the context of social exclusion as not everyone responds aggressively after being ostracized. Research on human morality is at a crossroads, with one side claiming that moral judgment is the result of rational inference and the other side claiming that it is the result of emotion-laden intuition. This study investigated whether emotion drives moral judgment by manipulating a core component of the experience of emotion: physiological arousal. The sample consisted of 77 undergraduate students at Brooklyn College (57% women, 43% men; mean age = 20.1). One group of participants was led to believe their heart was beating quickly, and another group slowly, while they read and evaluated a series of text vignettes depicting …

The placenta is responsible for regulating the fetal growth and environment in preparation for the anticipated external life. To succeed, it continuously adapts and weaves in a variety of neuroendocrine signals from the maternal organism. If a mother was exposed to trauma and her physiology sustained a change in the functioning of the immune, autonomic and/or endocrine systems, the placenta may anticipate different external environments for her children as compared to children of mothers without trauma exposure. The main purpose of this dissertation is to characterize noninvasive neuroimaging biomarkers of neurovascular injury in moderate-severe traumatic brain injury (msTBI) as potential treatment targets for neuromodulation therapy. In Chapter 1, I aim to introduce msTBI and two known sequelae of neurovascular damage in msTBI: reduced cerebral blood flow (Ware et al., 2020; Julliene et al., 2016) and altered low frequency fluctuations in blood oxygen level-dependent signal (Zhou et al., 2021; Palacios et al., 2013). Successful regulation of fear memories is a fundamental tenet to the exposure-based therapies often employed by mental health professionals for individuals with PTSD, phobias, and other anxiety disorders. Traumatic brain injury (TBI) is a significant health concern. Around 74 million people sustain a traumatic brain injury worldwide. The damage caused by TBI produces two types of injury; primary and secondary injuries. Primary injury is caused within milliseconds and is irreversible. Secondary brain injury is delayed and produced by molecular, cellular, and structural disruption after the initial injury. However, approximately a third of individuals with anxiety do not respond to current treatment approaches, highlighting the need to identify additional potential therapeutic mechanisms. Safety learning is one such mechanism, but methodological challenges and a dearth of research have prevented the field from advancing the understanding of the role of safety learning in the etiology and remediation of anxiety disorders. Post-traumatic stress disorder (PTSD) is a trauma-related disorder characterized by intense fearful memory formation. Women are twice as likely as men to develop PTSD, indicating there may be sex differences in the underlying neural circuits. Given that serotonin (5-HT) dysfunction is implicated in PTSD, and 5-HT modulates fear learning, we investigated whether there are sex differences in the modulation of fear learning by 5-HT. We administered the selective serotonin reuptake inhibitor (SSRI) citalopram (20mg/kg or 10m/kg, i.p.) once to male and female mice prior to auditory fear conditioning and tested the effects on fear memory the next day. Research has helped to shed light on the functional organization and neural mechanisms of distractors on memory-guided saccades. In our current study we have utilized eye tracking and EEG technology to simultaneously record the changes in saccadic eye movement (SEM) behavior and event-related potentials (ERPs) associated with performance on a memory-guided saccade task with distractor conditions. Thirteen healthy control participants (n = 13; 6 female) were tasked to complete 864 memory-guided saccade trials with both visible (white) and invisible (black) distractors presented on a black background before saccade initiation. Current thinking suggests that genotypes associated with impulse-control disorders and negative emotionality, such as monoamine oxidase-a (MAOA), interact with negative early environmental factors like childhood maltreatment and develop into the disorders know as Borderline Personality Disorder (BPD) and Antisocial Personality Disorder (ASPD). Trait anxiety refers to an individual’s sensitivity to perceived threat. Though it is not itself diagnosable according to the DSM-5, trait anxiety scales are often administered in a clinical context and serve as an indicator for anxiety disorders. High levels of trait anxiety can result in prolonged periods of intense worry and dysfunction, even in those who are not diagnosed with an anxiety disorder. An activity-based anorexia (ABA) paradigm in adolescent female mice was used to explore whether anorexia affects circuits underlying reward. The ABA paradigm significantly enhanced methamphetamine-induced conditioned place preference (CPP), but not wheel-induced CPP. These results indicate that the ABA paradigm enhances the rewarding properties of methamphetamine, but not wheel running.

Animals learn associations between environmental cues and the natural rewards they predict (e.g., food, water, sex). As a result, reward-predictive cues come to trigger vigorous reward-seeking responses. Many neurons in the nucleus accumbens (NAc) become excited upon presentation of an already-learned reward-predictive cue. These NAc responses encode the motivational value of the cue and are necessary for the expression of the subsequent approach behavior. In dynamic environments, split-second sensorimotor decisions must be prioritized according to potential payoffs to maximize overall rewards. The impact of relative value on deliberative perceptual judgments has been examined extensively, but relatively little is known about value-biasing mechanisms in the common situation where physical evidence is strong but the time to act is severely limited. This research examines the behavioral and electrophysiological indices of how value biases split-second perceptual decisions and the possible mechanisms underlying the process. Methamphetamine is an addictive illicit psychostimulant that produces lasting neurochemical and behavioral changes. The mechanisms underlying these deficits have been characterized in animal models using extremely high doses. Currently, better translational models are needed to understand the onset and progression of these deficits that more accurately reflect the gradual and voluntary dosing parameters as chosen by an abuser. To that end, a new model of methamphetamine administration, labeled Voluntary Oral Methamphetamine Administration (VOMA), offers a means to examine the progression of neurotoxicity, behavioral deficits, and the addiction process through a voluntary consumption framework.

Background: Research investigating patterns of electroencephalographic (EEG) brain asymmetry aids our understanding of neural systems involved in the processing of emotion, motivation, and psychopathology. Withdrawal-motivated negative emotions characteristic of depression are associated with relative right prefrontal cortex (PFC) activity, whereas approach- motivated positive emotions are associated with relative left PFC activity. Styles of emotion regulation (ER), or modulation of the intensity and duration of emotional responses, are also associated with presence (e.g., suppression, or maladaptive ER) versus absence (e.g., cognitive reappraisal, or adaptive ER) of depression vulnerability. Psychopathy is a multifaceted disorder characterized by a lack of cognitive and emotional empathy. The traditional model of psychopathy divides the disorder into two factors: Factor 1 consists of the interpersonal and affective traits of psychopathy while Factor 2 measures antisocial behaviors and lifestyle choices. The attention-to-the-eyes hypothesis argues that psychopathic individuals have impaired emotion recognition (specifically for fear) due to deficits in orienting attention to salient facial features like the eyes.
